Interaction of three-finger toxins with phospholipid membranes: comparison of S- and P-type cytotoxins.

The CTs (cytotoxins) I and II are positively charged three-finger folded proteins from venom of Naja oxiana (the Central Asian cobra). They belong to S- and P-type respectively based on Ser-28 and Pro-30 residues within a putative phospholipid bilayer binding site. Previously, we investigated the interaction of CTII with multilamellar liposomes of dipalmitoylphosphatidylglycerol by wide-line (31)P-NMR spectroscopy. To compare interactions of these proteins with phospholipids, we investigated the interaction of CTI with the multilamellar liposomes of dipalmitoylphosphatidylglycerol analogously.

A semi-parametric accelerated failure time cure model.

A cure model is a useful approach for analysing failure time data in which some subjects could eventually experience, and others never experience, the event of interest. A cure model has two components: incidence which indicates whether the event could eventually occur and latency which denotes when the event will occur given the subject is susceptible to the event. In this paper, we propose a semi-parametric cure model in which covariates can affect both the incidence and the latency. A logistic regression model is proposed for the incidence, and the latency is determined by an accelerated fa

Modelling the kinetics of chromosome exchange formation in human cells exposed to ionising radiation.

A biophysical model has been applied to study the kinetics of chromosome exchange formation in human cells. Chromosomal exchange induction (for example dicentrics) by ionising radiation was modelled by means of the Monte Carlo technique. This involved energy deposition by electrons, production of chromosomal breaks (assumed to be DNA double-strand breaks) and their repair and exchange. Exchanges were assumed to result from pairwise interaction between two DNA breaks in a distance-dependent manner. The rate at which exchanges are formed was found to depend upon how the exchange to no-exchange p

Deep unrolled regularization for limited-angle dual-panel positron emission mammography.

A dual-panel Positron Emission Mammography (PEM) scanner is a breast-dedicated device with better spatial resolution and sensitivity than conventional Positron Emission Tomography (PET). However, the main limitation of dual-panel PEM is the limited-angle artifacts in the cross-plane slices of the reconstructed images, arising from the acquisition geometry. This work proposes incorporating deep unrolled regularization into the image reconstruction algorithm to mitigate these artifacts. A U-Net was trained to identify and correct the artifacts in cross-plane slices. Dual-panel PEM and ring-shape

Spatial distribution and human health risk assessment of nitrate and fluoride in drinking water of Maragheh County, Iran (2018-2023) using Monte Carlo simulation.

Monitoring nitrate and fluoride levels in drinking water is essential due to their potential adverse health effects. While studies have assessed these contaminants across Iran, comprehensive analyses of their spatial-temporal distribution and probabilistic health risks remain scarce for Maragheh County. This study addresses this gap by applying Monte Carlo simulation (MCS) and principal component analysis (PCA) to 132 drinking water samples collected from 2018 to 2023. This novel framework identifies contamination sources and quantifies risks across demographic groups. Results revealed that 97

Radiological and elemental hazards of informal gold mining at OAU: health risk modelling with spectrometric and probabilistic indices.

This study investigates the environmental and radiological health risks associated with artisanal gold mining activities along the Opa River section of Obafemi Awolowo University (OAU), Ile-Ife, Nigeria. The research addresses a critical gap in understanding the long-term stochastic health effects of heavy metals and naturally occurring radionuclides in a vulnerable academic environment. Artisanal mining, though economically beneficial, has led to significant soil contamination, posing potential risks to over 40,000 campus residents and surrounding communities. The study evaluates the extent o

Analytical quality by design-based development and validation of a robust impurity analysis method for albumin-bound paclitaxel nanoparticles.

Analytical Quality by Design (AQbD) was applied to develop a robust impurity (IMP) analysis method for albumin-bound paclitaxel nanoparticles (Nab-PTX). Preliminary studies revealed that the USP method could induce epimerization, increasing IMP VI (7-epi-paclitaxel) formation. Risk assessment (Ishikawa/FMEA) identified four critical method parameters: flow rate, acetonitrile ratio (ACN), column temperature, and formic acid (FA) content. A 27-run Box-Behnken design (BBD) and response surface modeling (R² = 0.8688-0.9950) revealed complex nonlinear relationships. Specifically, FA content suppres

A spatial scan statistic for group testing data.

Group testing protocols physically combine biomaterial from multiple individuals into a single 'pooled' specimen, which is then tested for markers of infection. Group testing offers a cost-effective approach for surveillance of low-prevalence infections because it greatly reduces the number of tests required relative to individual testing protocols. However, group testing poses challenges for detecting spatial clusters of disease because only pooled test results are observed. This article introduces a spatial scan statistic tailored to group testing data with variable pool sizes, using a likel

Technical note: reconstructing dose distributions from manually planned electron boosts in breast radiotherapy.

PURPOSE: In breast radiotherapy, delivery of manually-calculated electron boosts limits retrospective dose-response analyses as dose distribution is unavailable. This work evaluates the feasibility of reconstructing dose distributions from manually planned electron boosts in breast-conserving radiotherapy. METHODS: Only 72 out of 198 breast cancer patients had complete stored dose distributions from sequential electron boosts in the REQUITE study. Arbitrary data from 70/72 patients were used to develop and validate dose reconstruction method. Twenty patients were used to determine optimal par

A carbon ion minibeam treatment planning method with scissor beams.

BACKGROUND: Minibeam radiation therapy (MBRT) employs a highly modulated spatial dose distribution characterized by the peak-to-valley dose ratio (PVDR). Carbon minibeam radiation therapy (cMBRT) offers higher PVDR and relative biological effectiveness (RBE) compared to proton MBRT. However, achieving uniform target dose (UTD) coverage while maintaining a high PVDR in organs at risk (OAR) remains challenging. PURPOSE: To address this challenge and optimize the balance between PVDR in OAR and target dose homogeneity, we introduce the scissor-beam (SB) approach for cMBRT. METHODS: The SB metho

Converging lens radiotherapy (CLRT) employing kilovoltage x-ray source: Treatment planning study.

BACKGROUND: Converging lens radiotherapy (CLRT) is a novel radiotherapy modality employing a lens which produces a nearly monoenergetic x-ray beam at 60 keV to focus radiation on deep-seated tumors, enabling highly precise dose delivery. PURPOSE: This study represents the first comparison of CLRT treatment plan quality to conventional intensity-modulated radiation therapy (IMRT) and volumetric modulated arc therapy (VMAT). The goal is to provide a proof-of-concept of CLRT treatment planning and to evaluate whether CLRT could potentially offer improved organ-at-risk (OAR) sparing and target co

Investigating the use of a solid water and copper electron filter for reducing skin dose in an inline MRI-linac.

BACKGROUND: Skin dose measurements for the inline MRI-linac show a highly concentrated skin dose hot spot at the central axis of the beam. This is because the electron contamination is focused in the presence of the inline magnetic field, causing a significant entry surface (or skin) dose hot spot. The future clinical treatment will employ a 2 cm water electron filter to reduce electron contamination from the X-ray beam; however, this approach affects the overall dose distribution, and the skin dose remains significantly high. PURPOSE: To evaluate and compare the effectiveness of a copper ele

Power and sample size considerations for test-negative design with bias correction: a case study on the world first malaria vaccine.

BACKGROUND: Test-negative design (TND) studies are increasingly common in evaluating vaccine effectiveness (VE) for various infectious diseases. TND studies are susceptible to bias due to disease outcome misclassification caused by imperfect test sensitivity and specificity. Several bias correction methods have been proposed. However, sample size or power considerations for TND studies incorporating bias correction for such misclassification have not yet been investigated. METHODS: We used Monte Carlo simulations to assess how bias correction affects the statistical power and sample size for

The epidemiology of pathogens with pandemic potential: A review of key parameters and clustering analysis.

INTRODUCTION: In the light of the COVID-19 pandemic many countries are trying to widen their pandemic planning from its traditional focus on influenza. However, it is impossible to draw up detailed plans for every pathogen with epidemic potential. We set out to try to simplify this process by reviewing the epidemiology of a range of pathogens with pandemic potential and seeing whether they fall into groups with shared epidemiological traits. METHODS: We reviewed the epidemiological characteristics of 19 different pathogens with pandemic potential (those on the WHO priority list of pathogens,

Assessing sensitivity of predictive shoulder simulations to uncertain soft tissue properties.

Accurate modeling of glenohumeral joint mechanics requires reliable representations of passive structures such as ligaments and articular cartilage, yet these tissue properties are un-available in vivo on a subject-specific basis. This study evaluated how variability in ligament and cartilage properties propagates to joint-level outcomes. Using a validated musculoskeletal shoulder model and two Monte Carlo simulation frameworks, we independently varied glenohumeral ligament (stiffness and slack length) and cartilage (elastic modulus and thickness) parameters to predict secondary kinematics and
