Fast and highly sensitive fiber-enhanced Raman spectroscopic monitoring of molecular H2 and CH4 for point-of-care diagnosis of malabsorption disorders in exhaled human breath.

Breath gas analysis is a novel powerful technique for noninvasive, early-stage diagnosis of metabolic disorders or diseases. Molecular hydrogen and methane are biomarkers for colonic fermentation, because of malabsorption of oligosaccharides (e.g., lactose or fructose) and for small intestinal bacterial overgrowth. Recently, the presence of these gases in exhaled breath was also correlated with obesity. Here, we report on the highly selective and sensitive detection of molecular hydrogen and methane within a complex gas mixture (consisting of H2, CH4, N2, O2, and CO2) by means of fiber-enhance

A novel approach to non-invasive lung cancer screening via exhaled breath analysis using proton transfer reaction time-of-flight mass spectrometry.

Lung cancer (LC) remains a global public health problem. To minimize late diagnosis, new detection techniques are needed. The analysis of volatile organic compounds (VOCs) in human breath (the volatilome) represents a non-invasive alternative to current methods for the early detection of LC. In this study, VOCs were determined in the breath of individuals with LC (N= 22) and without LC (non-LC,N= 21) using proton transfer reaction time-of-flight mass spectrometry. Orthogonal partial least squares discriminant analysis and hierarchical cluster analysis were conducted on a subset of 54 VOC ions

Associations of PM2.5 carbonaceous and ionic constituents with fractional exhaled nitric oxide (FeNO): A panel study with 16 follow-up visits over 4 years.

Epidemiological studies have linked fine particulate matter (PM2.5) to elevated fractional exhaled nitric oxide (FeNO), biomarker of airway inflammation. However, the specific roles of PM2.5 chemical components remain underexplored. This panel study examined relationships between temperature-resolved carbon subfractions or ionic components of PM2.5 and FeNO. A cohort of 69 self-reported healthy undergraduate students was followed 16 times between 2014 and 2018. PM2.5 samples were collected from the roof of a campus building, while carbonaceous and ionic components were quantified using a carbo

Computational modeling method to estimate secondhand exposure potential from exhalations during e-vapor product use under various real-world scenarios.

Potential secondhand exposure of exhaled constituents from e-vapor product (EVP) use is a public health concern. We present a computational modeling method to predict air levels of exhaled constituents from EVP use. We measured select constituent levels in exhaled breath from adult e-vapor product users, then used a validated computational model to predict constituent levels under three scenarios (car, office, and restaurant) to estimate likely secondhand exposure to non-users. The model was based on physical/thermodynamic interactions between air, vapor, and particulate phase of the aerosol.

Fraction of Exhaled Nitric Oxide Levels Are Elevated in People Living With Human Immunodeficiency Virus Compared to Uninfected Controls, Suggesting Increased Eosinophilic Airway Inflammation.

BACKGROUND: Increased risk of asthma and chronic obstructive pulmonary disease has been reported in people living with human immunodeficiency virus (PLWH). Fraction of exhaled nitric oxide (FeNO) is a marker of eosinophilic airway inflammation. We assessed FeNO levels in PLWH and matched uninfected controls and investigated whether human immunodeficiency virus (HIV) status is independently associated with elevated FeNO. METHODS: FeNO was quantified by NIOX Vero and pulmonary function was assessed by spirometry in 432 PLWH from the Copenhagen Comorbidity in HIV Infection Study and in 1618 age-

Real-time breath analysis of exhaled compounds upon peppermint oil ingestion by secondary electrospray ionization-high resolution mass spectrometry: technical aspects.

Breath analysis by secondary electrospray ionization high-resolution mass spectrometry (SESI-HRMS) has potential for clinical diagnosis and drug monitoring. However, there is still a lack of benchmarking data that shows the capability of this technique and allows comparability with other breath analysis techniques. In this regard, the goal of this study was the identification of volatile compounds upon ingestion of a specific peppermint oil capsule to get benchmark data for real-time breath analysis with SESI-HRMS. This was done in the framework of a consortium set up by the International Asso

Fractional exhaled nitric oxide as a predictor of response to inhaled corticosteroids in patients with non-specific respiratory symptoms and insignificant bronchodilator reversibility: a randomised controlled trial.

BACKGROUND: Chronic non-specific respiratory symptoms are difficult to manage. This trial aimed to evaluate the association between baseline fractional exhaled nitric oxide (FeNO) and the response to inhaled corticosteroids in patients with non-specific respiratory symptoms. METHODS: In this double-blind randomised placebo-controlled trial, we enrolled undiagnosed patients, aged 18-80 years, with cough, wheeze, or dyspnoea and less than 20% bronchodilator reversibility across 26 primary care centres and hospitals in the UK and Singapore. Patients were assessed for 2 weeks before being randoml

Characteristics of haloethylene-induced acetonemia in rats.

A series of halogenated ethylenes (vinyl chloride, vinylidene fluoride, cis- and trans-1,2-dichloroethylene, perchloroethylene) induces increased acetone exhalation in rats. Exposures of differently pre-treated rats to vinylidene fluoride suggest that a metabolite of the haloethylene must be envolved in eliciting this formation of acetone. This conclusion is based on (a)dependence of acetone exhalation on the concentration of vinylidene fluoride, (b)effect of inducing agents, (c)effect of pyrazol, a metabolic inhibitor, (d)effect of cysteine, (e)effect of hypoxia and (f)the time course of acet…

Respiration phase dependent modulation of somatosensory evoked potentials.

The respiration phase (inhalation vs. exhalation) has been shown to influence behavioral variables such as action execution, perception, and cognition, possibly due to interactions between the breathing cycle and higher cortical functions. However, whether somatosensory processing differs between inhalation and exhalation remains unclear. The aim of this study was to investigate the effects of respiration phase on somatosensory evoked potentials (SEPs) in healthy individuals. While simultaneously recording high-density electroencephalography and respiratory signals during normal breathing, ele…

Short-term longitudinal variation in exhaled nitric oxide fraction predicts future moderate-to-severe exacerbations in COPD patients on triple therapy: a prospective cohort study.

BACKGROUND: Fractional exhaled nitric oxide (F ENO) reflects type 2 airway inflammation, yet the prognostic utility of longitudinal F ENO changes in COPD remains unclear. We aim to explore whether 6-month F ENO variation patterns predict subsequent moderate-to-severe exacerbations in COPD patients receiving triple therapy. METHODS: We prospectively evaluated COPD patients receiving triple inhaled therapy from 2023 to 2024. Four F ENO-related variables were used to derive F ENO variation phenotypes via K-means unsupervised clustering. RESULTS: Three distinct F ENO-variation phenotypes were id

Ultrasensitive detection of colorectal cancer biomarkers in exhaled breath using superhydrophobic HCPs-2Br-based SPME-GC-MS with a two-step pre-enrichment strategy.

BACKGROUND: Exhaled breath has emerged as a promising biospecimen for disease diagnosis, particularly for non-invasive screening. However, the detection of disease biomarkers in exhaled breath presents significant challenges due to their ultra-trace levels and the complex, high-humidity composition of the breath matrix. Consequently, there is a pressing need to develop innovative pretreatment technologies to enable the sensitive detection of colorectal cancer (CRC)-related biomarkers for early non-invasive screening. RESULTS: In this study, a superhydrophobic dibromodibenzothiophene-based hyp
