The gut epithelial barrier is a strategic place to prevent, or at least to limit, parasite dissemination upon oral infection with Toxoplasma gondii. Innate immunity to this pathogen results from delicate interactions involving different components of the infecting agent and the host. We herein aimed to examine the molecular mechanism by which protozoan DNA boosts the production of α-defensin-5 (DEFA-5), the main antimicrobial peptide at the target site of infection. The present study shows that DEFA-5 is rapidly upregulated in intestinal epithelial cells following intracellular Toll-like recep

PROBLEM: Toll-like receptors (TLRs) are innate immune receptors that mediate the pattern recognition of, and response toward, pathogens and host-derived danger signals. We reported that cyclooxygenase-2 (COX-2) and microsomal prostaglandin E synthase (mPGES) mRNA were expressed in cases of endometriosis. The relationship between COX-2, mPGES-1, and TLR4 in endometriotic lesions has yet to be determined.
METHOD OF STUDY: Endometriosis samples were obtained from 37 patients with endometrial cysts. Endometrial tissues were obtained from patients undergoing surgical procedures for benign gynecolo

Listeria monocytogenes (Lm) is a foodborne Gram-positive pathogenic bacterium that primarily causes clinical manifestations such as sepsis, meningitis, and monocytosis. The main susceptible populations include newborns, pregnant women, adults over 40 years old, and immunocompromised individuals. Currently, antibiotic therapy remains the primary clinical treatment, but in recent years the issue of antibiotic resistance has become increasingly prominent. This review summarizes recent advances in host immune mechanisms against Lm infection, focusing on: (1) the defensive roles of host barrier sys

BACKGROUND & AIMS: Hepatitis B virus (HBV) and D virus (HDV) co-infections cause the most severe form of viral hepatitis. HDV induces an innate immune response, but it is unknown how the host cell senses HDV and if this defense affects HDV replication. We aim to characterize interferon (IFN) activation by HDV, identify the responsible sensor and evaluate the effect of IFN on HDV replication.
METHODS: HDV and HBV susceptible hepatoma cell lines and primary human hepatocytes (PHH) were used for infection studies. Viral markers and cellular gene expression were analyzed at different time points

Peptidoglycan recognition proteins (PGRPs) are a class of pattern recognition receptors (PRRs) that activate the innate immune system in response to microbial infection by detection of peptidoglycan, a distinct component of bacterial cell walls. Bioinformatic studies have revealed four PGRPs in the red imported fire ant Solenopsis invicta; nonetheless, the mechanism of the immune response of S. invicta induced by pathogens is still poorly understood. The peptidoglycan recognition protein full-length cDNA (designated as SiPGRP-S1/S2/S3/L) from S. invicta was used in this investigation. Accordin

Hepatitis B virus (HBV) and hepatitis C virus (HCV) are among the most endemic pathogens worldwide, with more than 500 million people globally currently infected with these viruses. These pathogens can cause acute and chronic hepatitis that progress to liver cirrhosis or hepatocellular carcinoma. Both viruses utilize multifaceted strategies to evade the host surveillance system and fall below the immunological radar. HBV has developed specific strategies to evade recognition by the innate immune system and is acknowledged to be a stealth virus. However, extensive research has revealed that HBV

Tobacco smoking is strongly linked to vascular damage contributing to the development of hypertension, atherosclerosis, as well as increasing the risk for neurodegeneration. Still, the involvement of the innate immune system in the development of vascular damage upon chronic tobacco use before the onset of clinical symptoms is not fully characterized. Our data provide evidence that a single acute exposure to tobacco elicits the secretion of extracellular vesicles expressing CD105 and CD49e from endothelial cells, granting further recognition of early preclinical biomarkers of vascular damage.

Inflammatory disease is often associated with an increased incidence of venous thromboembolism in affected patients, although in most instances, the mechanistic basis for this increased thrombogenicity remains poorly understood. Acute infection, as exemplified by sepsis, malaria and most recently, COVID-19, drives 'immunothrombosis', where the immune defence response to capture and neutralise invading pathogens causes concurrent activation of deleterious prothrombotic cellular and biological responses. Moreover, dysregulated innate and adaptive immune responses in patients with chronic inflamm

Attenuation in the activity of the negative regulators or the hyperactivity of plant innate immune receptors often causes ectopic defense activation manifested in severe growth retardation and spontaneous lesion formations, referred to as autoimmunity. In this review, we have described the cellular and molecular basis of the development of autoimmune responses for their useful applications in plant defense. Plants are exposed to diverse disease-causing pathogens, which bring infections by taking over the control on host immune machineries. To counter the challenges of evolving pathogenic races

Bacterial meningitis is - despite therapeutical progress during the last decades - still characterized by high mortality and severe permanent neurogical sequelae. The brain is protected from penetrating pathogens by both the blood-brain barrier and the innate immune system. Invading pathogens are recognized by so-called pattern recognition receptors including the Toll-like receptors (TLR) which are expressed by glial immune cells in the central nervous system. Among these, TLR2 is responsible for the detection of Gram-positive bacteria such as the meningitis-causing pathogen Streptococcus pneu

The SARS-CoV-2 spike protein has been shown to activate Toll-like receptor 4 (TLR4), yet the precise molecular structures driving recognition and subsequent activation remain poorly defined. Here, we present in silico structural alignments and molecular docking simulations indicating potential spatial compatibility between the wild-type SARS-CoV-2 HR1HR2 fusion core and the human TLR4/MD-2 heterodimer. The computational models project candidate interfaces involving salt bridges, as well as polar and non-polar interactions, with both TLR4 and MD-2 dimerization partners, suggesting a theoretical

The Enterococcus genus comprises an unusual subset of the human microbiota with the dual role of beneficial commensals and opportunistic pathogens. The biological activity of Enterococcus species is not limited to colonization and is mainly driven by numerous bioactive compounds. Metabolites, extracellular vesicles (EVs), exopolysaccharides, and cell wall structures serve as mediators of complex host-microbe interplay. Through the transfer of these bioactive substances, Enterococcus-derived EVs shape host signaling pathways and help bacteria to adapt to environmental conditions. Additionally,

Hemolin, a distinctive member of the immunoglobulin superfamily in invertebrates, functions as a pivotal molecular nexus that integrates innate immunity with developmental programming in Lepidoptera. This review synthesizes evidence expanding its role from a canonical pattern recognition receptor to a central signaling hub. We delineate its multifaceted functions in antibacterial, antifungal, and virus family specific antiviral defenses and detail its regulatory crosstalk with core immune pathways, including the Toll/Imd and prophenoloxidase cascades. A critical focus is the transcriptional re

Therapeutic cancer vaccines are designed to induce tumor-specific immunity and durable antitumor surveillance. However, their clinical efficacy remains limited by antigen heterogeneity, defective antigen presentation, immune exhaustion, and immunosuppression within the tumor microenvironment (TME). Advances in sequencing, immunopeptidomics, artificial intelligence (AI)-assisted neoantigen prioritization, nucleic acid engineering, dendritic cell (DC) biology, and combination therapy have shifted the field from empirical antigen exposure toward mechanism-guided vaccine design. Myocarditis is an inflammatory disease of the myocardium with heterogeneous aetiologies, in which innate and adaptive immune responses critically influence clinical outcomes, ranging from complete recovery to progression toward dilated cardiomyopathy. In viral myocarditis, pathogen- and damage-associated molecular patterns activate pattern-recognition receptors, triggering NF-κB and inflammasome signaling and amplifying pro-inflammatory cytokine responses. Early myocardial injury involves neutrophils, mast cells, natural killer cells, and inflammatory monocytes, followed by activation of adapt

Phosphorylation is a reversible post-translational modification that dynamically regulates immune responses by reshaping the antigen profiles. Phosphoproteomics and immunopeptidomics studies reveal that phosphorylated peptides (phosphopeptides) are naturally processed and presented by MHC class I and II molecules and can elicit robust T-cell responses. These epitopes influence proteasome cleavage, TAP transport, and MHC loading, defining the immunopeptidome under physiological and pathological states. Structurally, phosphate groups enhance MHC binding and immunogenicity, positioning phosphoryl

NK cells belong to innate lymphoid cells and able to eliminate infected cells and tumor cells. NK cells play a valuable role in controlling viral infections. Also, they have the potential to shape the adaptive immunity via a unique crosstalk with the different immune cells. Murine models are important tools for delineating the immunological phenomena in viral infection. To decipher the immunological virus-host interactions, two major infection models are being investigated in mice regarding NK cell-mediated recognition: murine cytomegalovirus (MCMV) and lymphocytic choriomeningitis virus (LCMV

The reemergence of the Zika virus (ZIKV) infection in recent years has posed a serious threat to global health. Despite being asymptomatic or mildly symptomatic in a majority of infected individuals, ZIKV infection can result in severe manifestations including neurological complications in adults and congenital abnormalities in newborns. In a human host, ZIKV is primarily recognized by RIG-like receptors and Toll-like receptors that elicit anti-viral immunity through the secretion of type I interferon (IFN) to limit viral survival, replication, and pathogenesis. Intriguingly, ZIKV evades its h
