Malignant hematopoietic cell lines: in vitro models for the study of multiple myeloma and plasma cell leukemia.

Multiple myeloma (MM) is a neoplasm of a terminally differentiated B-cell. The disease is progressive and always lethal characterized by the slow proliferation of malignant plasma cells in the bone marrow. Much of our current understanding of the biology of MM has been obtained by studying MM-derived cell lines. Human myeloma cell lines were shown to be suitable model systems for use in various fields of the biological sciences. However, it has proved very difficult to establish cell lines from plasma cell dyscrasias. Most reported MM cell lines have been derived from patients with advanced di…

Expression of the bcl-2 gene in human multiple myeloma cell lines and normal plasma cells.

The bcl-2 gene, encoding a mitochondrial membrane protein suggested to play an important role in cell survival, is translocated into the Ig loci in about 80% of human follicular lymphomas, which results in a high level of expression. This report shows that bcl-2 was expressed in eight of eight human multiple myeloma cell lines and in normal lymph node and bone marrow plasma cells. In the majority of the myeloma lines, the level of expression was comparable with that observed in Karpas 422, a follicular lymphoma cell line carrying a 14;18 translocation of the bcl-2 gene. DNA rearrangements of t…

Diabetic GK rat plasma but not normal Wistar rat plasma induces insulin-stimulated DNA synthesis in primary cultured smooth muscle cells in GK rat aorta.

We investigated the effect of diabetic plasma on insulin-stimulated DNA synthesis in primary cultured aortic smooth muscle cells (SMC) of the GK rat, a model of non-insulin-dependent diabetes mellitus, and compared it with that of Wistar normal rat plasma. We measured the incorporation of 3H-thymidine into cultured SMC. The diabetic plasma (3%) of GK rat, but neither the plasma (3%) of Wistar normal rat nor the plasma (3%) (not containing both insulin-like growth factor-I (IGF-I) and corticosterone) of Wistar hypophysectomized rat induced insulin-stimulated DNA synthesis in GK rat SMC. The res

Concomitant mobilization of plasma cells and hematopoietic progenitors into peripheral blood of multiple myeloma patients: positive selection and transplantation of enriched CD34+ cells to remove circulating tumor cells.

One advantage of the use of peripheral blood stem cells (PBSCs) over autologous bone marrow would be a reduced risk of tumor cell contamination. However, the level of neoplastic cells in the PB of multiple myeloma (MM) patients after mobilization protocols is poorly investigated. In this study, we evaluated PB samples from 27 pretreated MM patients after the administration of high dose cyclophosphamide (7 g/m2 or 4 g/m2) and granulocyte-colony stimulating factor for the detection of myeloma cells as well as hematopoietic progenitors. Plasma cells containing intracytoplasmic lg were counted by …

miR-15a/16-1 deletion in activated B cells promotes plasma cell and mature B-cell neoplasms.

Chromosome 13q deletion [del(13q)], harboring the miR-15a/16-1 cluster, is one of the most common genetic alterations in mature B-cell malig … s including an increased number of GC B cells, percentage of dark zone B cells, and maturation into plasma cells. With time, this leads to development of mature B-cell neoplasms resembling human extramedullary plasmacytoma (EP) as well as follicular and diffuse large B-cell lymphomas. The indolent nature and lack of bone marrow involvement of EP in our murine …

A novel method for simultaneously measuring boronophenylalanine uptake in brain tumor cells and number of cells using inductively coupled plasma atomic emission spectroscopy.

Boron neutron capture therapy (BNCT) combines neutron irradiation with boron compounds that are selectively uptaken by tumor cells. Boronophenylalanine (BPA) is a boron compound used to treat malignant brain tumors. The determination of boron concentration in cells is of great relevance to the field of BNCT. This study was designed to develop a novel method for simultaneously measuring the uptake of BPA by U87 and U251 cells (two brain tumor cell lines) and number of cells using inductively coupled plasma atomic emission spectroscopy (ICP-AES). The results revealed a linear correlation between

B-cell and plasma cell proliferations associated with T follicular helper cell lymphomas: clinical-pathological and genetic features.

Nodal T-follicular helper cell lymphomas (nTFHL) are aggressive peripheral T-cell lymphomas with unique clinical-biological features. B-cell and/or plasma cell proliferations (B/PCP) are frequently reported in nTFHL, yet their histological spectrum remains incompletely defined. This study aims to delineate the clinical, pathological and molecular features of a retrospective series of nTFHL-associated B/PCP, with particular emphasis on recurrent disease patterns. To this aim, we retrospectively analyzed 39 nTFHL with 45 synchronous and/or metachronous B/PCP, collected from five Italian referral

Expression of SLAM (CD150) cell-surface receptors on human B-cell subsets: from pro-B to plasma cells.

The SLAM (CD150) family receptors are leukocyte cell-surface glycoproteins involved in leukocyte activation. These molecules and their adaptor protein SAP contribute to the effective germinal center formation, generation of high-affinity antibody-secreting plasma cells, and memory B cells, thereby facilitating long-term humoral immune response. Multi-color flow cytometric analysis was performed to determine the expression of CD48 (SLAMF2), CD84 (SLAMF5), CD150 (SLAM or SLAMF1), CD229 (Ly9 or SLAMF3), CD244 (2B4 or SLAMF4), CD319 (CRACC, CS1, or SLAMF7), and CD352 (NTB-A or SLAMF6) on human cel…

Plasma protein oxidation is correlated positively with plasma iron levels and negatively with hemolysate zinc levels in sickle-cell anemia patients.

INTRODUCTION: Sickle-cell anemia (SCA) is a disease of high oxidative stress. The oxidative medium of SCA was evaluated by protein oxidation parameters and their correlation with lipids and ions were investigated both in the plasma and in the erythrocyte. METHODS: Blood samples were obtained from 15 steady-state patients with SCA, 10 carriers, and 10 controls. Plasma protein carbonyl, total sulfhydryl, total cholesterol, triglyceride, phospholipids, and bilirubin levels were determined by spectrophotometric methods. Plasma copper, zinc, and iron levels were determined by atomic absorption spe

Bioelectric regulation of stem cell fate: From the plasma membrane to organelles.

Stem cell identity has traditionally been viewed through the lens of transcriptional and metabolic programs. Recent work has revealed an additional layer of complexity: the plasma membrane and intracellular organelles maintain compartment-specific membrane potentials, pH gradients, and ion dynamics that can influence stemness and fate. In this review, we synthesize recent advances identified across diverse stem cell populations to highlight convergent and divergent bioelectric mechanisms driving cell fate decisions. We explore how plasma membrane mechanosensors convert physical cues into inter

Plasma heme pool compartmentalization is linked to pathophysiology in Sickle Cell Disease.

Heme toxicity plays a central role in the pathophysiology of Sickle Cell Disease (SCD), contributing to severe complications such as vaso-occlusion and acute chest syndrome. The continuous release of hemoglobin and heme from increased intravascular hemolysis can exceed the capacity of protective scavenger proteins, leading to heme accumulation in plasma. Interactions with various binding partners result in the formation of different plasma heme species and the compartmentalization of the plasma heme pool. In an observational biomarker study, we used novel bioanalytical assays to quantify plasm

Plasma Metabolic Signature and Single-Cell Regulatory Network for Ischemic Stroke.

Ischemic stroke (IS) is a leading cause of mortality and long-term disability worldwide, with ultra-early diagnosis critical for effective reperfusion therapy but challenged by nonspecific symptoms and limited sensitivity of imaging in the hyperacute phase. Plasma metabolomic alterations occur rapidly after ischemia onset, offering potential for accessible biomarkers and deeper insights into underlying pathogenic mechanisms. We therefore aimed to explore plasma metabolic alterations and their associated regulatory networks in IS. We conducted a multicenter case–control study involving 493 IS p

Epigenetic and metabolic reprogramming support plasma cell differentiation in germinal centers.

The germinal center (GC) is a specialized structure that ensures the production of high-quality antibodies. Although recent studies have pinpointed the existence of a pre-plasma cell (prePC) population within mouse GC B cells, it remains unclear how these prePCs mechanistically differentiate into PCs. Additionally, there is a lack of validation of these findings in human cells. Here, we demonstrate CD205 is highly expressed in prePCs both in mouse and in human. The histone H3 lysine 27 demethylase Kdm6b, not Kdm6a, potently enhances the differentiation of prePCs into bona fide PCs by removing

Plasma Cell Selection and Migration in the Establishment of Long-Lived Antibody Responses.

As B cells are activated and differentiate into plasma cells, they execute migration programs that allow them to traffic within secondary lymphoid tissues and reach the bone marrow, where they can mature as long-lived plasma cells. Recent studies have revealed mechanisms underlying not only affinity-driven but also isotype-driven plasma cell selection. In addition, the importance of migration programs in plasma cell longevity has become increasingly appreciated. This review focuses on mechanisms that regulate plasma cell numbers and their migration to the bone marrow, with emphasis on our find…

A Pilot Study of Plasma Cell-Free DNA Fragmentomics in Gallbladder Cancer.

BACKGROUND & AIMS: Gallbladder cancer (GBC) is a rare and highly lethal biliary tract cancer with limited treatment options and lack of diagnostic and prognostic noninvasive biomarkers. Circulating cell-free DNA (cfDNA) offers a noninvasive means to capture fragmentomics alterations that may assist with diagnosis and biological characterization. This pilot study aimed to identify cfDNA-based features that differentiate GBC from individuals with gallstones and healthy controls. METHODS: cfDNA was extracted from archived plasma samples from 67 individuals in two case-control studies from China

Plasma Proteome Signatures in Sickle Cell Anemia and the Effect of Hydroxyurea Treatment.

Sickle Cell Anaemia (SCA) is a monogenic blood disorder caused by a mutation in the β-globin gene, yet it presents with marked clinical variability. Although hydroxyurea (HU) is an established therapy, its precise mechanism of action remains incompletely understood. Plasma proteins represent valuable biomarkers for elucidating disease mechanisms and treatment responses. In this study, plasma proteome profiling of 31 healthy controls and 76 SCA patients identified 43 differentially abundant proteins (DAPs) that form a highly interconnected interaction network. Proteins with increased abundance
