Genetic instability is generally thought to underlie the process of aging and is predominantly associated with meiosis and mitosis. This review will discuss DNA damage and repair, somatic mutations and somatic recombination events in non-dividing neurons in relation to aging. In general it can be concluded that mutagenesis operates at high frequency in the brain. Present data do not provide clear evidence for accumulating DNA damage or a change in DNA repair activity in the brain with age. However, a linear age-related increase in frameshift mutations has been shown to occur in vasopressin neu

We report on linkage analysis and haplotype characterization in 12 Cuban families with autosomal dominant polycystic kidney disease (ADPK) using PKD1-linked markers. They included both standard restriction fragment length polymorphisms (26.6., BLu24, and pGGG1) as well as microsatellite polymorphisms (CW2, 16AC2.5, and SM6). All of the examined families were fully informative for genetic diagnosis and no evidence of unlinked families was found. Analysis of two recombination events places PKD1 distal to the marker BLu24 and reduces the size of the region likely to contain the disease gene by ap

TAL1 gene rearrangement is the most common genetic defect associated with T cell acute lymphoblastic leukaemia (T-ALL). Tumour-specific rearrangements of TAL1 arise as a result of either chromosome translocation or local DNA recombination. TAL1 gene products possess the basic helix-loop-helix (bHLH) motif, a DNA-binding domain common to several known transcription factors. The bHLH domain of TAL1 is especially homologous to those encoded by TAL2 and LYL1, distinct genes that were also identified on the basis of chromosomal rearrangement in T-ALL. Thus, TAL1, TAL2 and LYL1 constitute a unique f

We have studied the inheritance of several polymorphic Xq27/28 DNA marker loci in two three generation families with the X linked neonatal lethal form of centronuclear/myotubular myopathy (XL MTM). We found complete linkage of XLMTM to all four informative Xq28 markers analysed, with GCP/RCP (Z = 3.876, theta = 0.00), with DXS15 (Z = 3.737, theta = 0.00), with DXS52 (Z = 2.709, theta = 0.00), and with F8C (Z = 1.020, theta = 0.00). In the absence of any observable recombination, we are unable to sublocalise the XLMTM locus further within the Xq28 region. This evidence for an Xq28 localisation

The Stickler syndrome is an autosomal dominant hereditary disorder of connective tissue with pleiotropic features including premature osteoarthropathy, mild spondyloepiphyseal dysplasia, vitreoretinal degeneration, and the Pierre-Robin sequence. Genetic linkage studies in two families with the Stickler syndrome have been performed using restriction fragment length polymorphisms associated with the structural gene for type II collagen, COL2A1. No recombinants between the Stickler phenotype and COL2A1 were observed. The total LOD score for linkage of the Stickler syndrome and COL2A1 at a recombi

Recent progress in research into ectopic hormone production and hormone gene expression in tumors is reviewed. Biochemical, immunochemical and morphological studies have shown that differentiation between "ectopic" and "eutopic" has become difficult and that the types of peptide hormones, neural peptides and growth factors produced, their molecular heterogeneities and ability of plural hormone production are greatly different in each tumor. Recently, the gene structures of many hormones and amino acid sequences of hormone precursors have been determined by genetic recombination techniques. How

Hepatitis C virus (HCV) is an important cause of chronic liver disease. Several highly diverse HCV genotypes exist with potential key functional differences. The HCV NS5A protein was associated with response to interferon (IFN)-α based therapy, and is a primary target of currently developed directly-acting antiviral compounds. NS5A is important for replication and virus production, but has not been studied for most HCV genotypes. We studied the function of NS5A using infectious NS5A genotype 1-7 cell culture systems, and through reverse genetics demonstrated a universal importance of the amphi

The advent of recombinant DNA has introduced novel ways of diagnosing genetic disorders. The application of these techniques in affected families, coupled with genetic counselling, should help prevent the recurrence of a proportion of cases, and thereby reduce a considerable amount of suffering, and in some cases the expense of caring for a severely affected individual. However, such aims may be achieved only when the importance of such services is more widely recognized and they become more generally available. Counselling and prenatal diagnostic services have been available in University Cen

Infectious hematopoietic necrosis virus is a highly contagious disease of juvenile salmonid species. From the IHNV HLJ-09 isolated in China, two recombinant viruses were generated by reverse genetics using the RNA polymerase II transcription system. The recombinant viruses were confirmed by RT-PCR, indirect immunofluorescence assay and electron microscopy. They were referred to as rIHNV HLJ-09 and rIHNV-EGFP. rIHNV HLJ-09 and rIHNV-EGFP could stably replicate in EPC cell lines and had the same cellular tropism as wtIHNV HLJ-09. But the titer of rIHNV-EGFP was significantly lower than rIHNV HLJ

BACKGROUND: Ovarian cancer (OC) is one of the leading causes of death from gynecological cancers in the world, primarily due to late stage diagnosis. Genetic mutations in genes involved in key cellular functions such as BRCA1, and BRCA2 significantly contribute to ovarian tumorigenesis. This study investigates the genetic landscape of Tunisian patients with familial or sporadic high-grade serous ovarian carcinoma (HGSOC) to guide therapeutic strategies and genetic counseling.
METHODS: We conducted a genetic study by targeted Next-Generation Sequencing (NGS) on 54 Tunisian HGSOC patients. A ca

Is the oxytocin-vasopressin (OT-AVP) system a part of the unseen force that subtly (in a clever and indirect way) directs our human fascination to ourselves? And is it possible that this fundamental drive is the inevitable handmaiden of the genetic selection for survival and reproduction that is played out at the level of the individual, the family and the society? Perhaps. But an equally intense biological drive to experience the unknown is intertwined and exists in the individual as "curiosity". Both are essential for survival and success of the species. Curiously, the path to understanding

Thyroid-stimulating hormone (TSH) is a member of a family of heterodimeric glycoprotein hormones which have a common alpha-subunit but differ in their hormone-specific beta-subunit. To date, numerous studies have been carried out to elucidate the structure-function relationship of glycoprotein hormones and molecular mechanism of hormone action. On the other hand, several types of abnormality in the structure and function of glycoprotein hormones, as well as TSH, have been observed in clinical samples, especially in malignant diseases. To approach the structure-function relationships of bioacti

Among the multiple SARS-CoV-2 variants identified since summer 2020, several have co-circulated, creating opportunities for coinfections and potentially genetic recombinations that are common in coronaviruses. Viral recombinants are indeed beginning to be reported more frequently. Here, we describe a new SARS-CoV-2 recombinant genome that is mostly that of a Omicron 21L/BA.2 variant but with a 3' tip originating from a Omicron 21K/BA.1 variant. Two such genomes were obtained in our institute from adults sampled in February 2022 in university hospitals of Marseille, southern France, by next-gen

Linkage data using the markers DXS51, F9, DXS15, and DXS52 are presented from 14 pedigrees segregating with the fragile X. Cytogenetic and DNA data were combined by two- or three-point linkage analysis for estimation of lod scores and carrier probabilities in potential carriers. Recombination frequencies (theta) corresponding to maximum z scores (zeta) were obtained for DXS51 (zeta = 3.45, theta = 0.0), DXS15 (zeta = 0.40, theta = 0.06), F9 (zeta = 3.15, theta = 0.09), and DXS52 (zeta = 3.60, theta = 0.11) with the fragile X. Considerable alterations to carrier probabilities occurred in some c

Immunologically mediated protection against H. pylori infection is an attractive alternative to antibiotic treatment. We compared the efficacy of conventional protein vaccination with that of genetic vaccination against experimental infection with H. pylori in mice. For oral immunization, we used the recombinant peptide of an antigenic fragment of UreB (rUreB) or H. pylori-whole cell lysate antigens, and for genetic immunization, we used recombinant pcDNA and pSec plasmids inserted with the fragment of ureB or DNA of the H. pylori genomic library. Mice were challenged with the mouse stomach-ad

Human immunodeficiency virus-1 (HIV-1) is among the most genetically diverse pathogens due to expeditious molecular evolution. The rapid change in HIV genomes intricates HIV transmission and progression and attributes HIV resistance to antiretroviral therapy (ART). In East Africa, as in other parts of the globe, HIV-1 occurs in various subtypes, circulating recombinant form (CRF) and unique recombinant forms, with subtype A1 being the most predominant. Surveillance of HIV-1 molecular diversity and drug resistance mutations (DRMs) is a linchpin for monitoring viral evolution and treatment effic

Although HIV-1 infection has now become a treatable chronic condition and not the deadly illness it once was, the costs of that treatment are substantial, and each infection prevented saves both financial and other costs. In China, the most predominant subtypes are CRF07_BC, CRF01_AE, and CRF55_01B, and the various second-generation recombinants are produced from the recombination between these subtypes. HIV full-length genome sequences can provide important information on their epidemiology. In this study, we identified two unique recombinant forms (URFs) designated as JLCC230106 and XJWQ2300

Complete genomic sequences of a non-redundant set of 70 recombinants between three serotypes of attenuated Sabin polioviruses as well as location (based on partial sequencing) of crossover sites of 28 additional recombinants were determined and compared with the previously published data. It is demonstrated that the genomes of Sabin viruses contain distinct strain-specific segments that are eliminated by recombination. The presumed low fitness of these segments could be linked to mutations acquired upon derivation of the vaccine strains and/or may have been present in wild-type parents of Sabi

Duck enteritis virus (DEV) is considered an ideal vector for waterfowl vaccine development due to its favorable safety profile and multiple genomic sites that accommodate foreign gene insertion. Duck hepatitis A virus (DHAV) causes acute hepatitis, neurological symptoms, and high mortality in young ducklings, and the predominant circulating serotype has shifted from type 1 to type 3, underscoring the urgent need for improved vaccines. In this study, we employed a CRISPR/Cas9 genome editing platform combined with dual single-guide RNAs (sgRNAs) and a homologous directed repair (HDR) strategy to
