Ecological advantages of autolysis during the development and dispersal of Pseudoalteromonas tunicata biofilms.

In the ubiquitous marine bacterium Pseudoalteromonas tunicata, subpopulations of cells are killed by the production of an autocidal protein, AlpP, during biofilm development. Our data demonstrate an involvement of this process in two parameters, dispersal and phenotypic diversification, which are of importance for the ecology of this organism and for its survival within the environment. Cell death in P. tunicata wild-type biofilms led to a major reproducible dispersal event after 192 h of biofilm development. The dispersal was not observed with a DeltaAlpP mutant strain. Using flow cytometry a

Meningococcal biofilm formation: structure, development and phenotypes in a standardized continuous flow system.

We show that in a standardized in vitro flow system unencapsulated variants of genetically diverse lineages of Neisseria meningitidis formed biofilms, that could be maintained for more than 96 h. Biofilm cells were resistant to penicillin, but not to rifampin or ciprofloxacin. For some strains, microcolony formation within biofilms was observed. Microcolony formation in strain MC58 depended on a functional copy of the pilE gene encoding the pilus subunit pilin, and was associated with twitching of cells. Nevertheless, unpiliated pilE mutants formed biofilms showing that attachment and accumula

Proteomics insight into phosphate limitation response in Burkholderia phytopathogens.

Inorganic phosphate (Pi) limitation is a pervasive environmental constraint that shapes microbial behaviour in soil and plant-associated niches. To determine how Pi scarcity influences the pathogenic potential of rice-infecting Burkholderia species, we performed comparative proteomic and lipidomic analyses of Burkholderia glumae and Burkholderia plantarii grown under Pi-replete and Pi-deplete conditions. Pi starvation triggered strong activation of the Pho regulon in both organisms, including induction of alkaline phosphatases, phosphonate transporters and the high-affinity PstSABC uptake syst

Biofilm and planktonic Staphylococcus aureus exhibit distinct gene expression patterns in response to cinnamaldehyde.

Staphylococcus aureus forms biofilms in the context many infections, including endocarditis, lung infection, and the colonization of implants. How antimicrobials specifically affect S. aureus biofilms as opposed to planktonic S. aureus is an important consideration in the development of treatments of these infections. It is well known that bacteria in biofilms are more resistant to antimicrobials, and the degree and nature of the responses is crucial to understanding the basis of this resistance. While certain antimicrobials such as antibiotics have specific mechanisms that induce pathways rel

The regulation characterization of novel transcription regulator JTY_2262 in Mycobacterium bovis BCG.

Mycobacterium tuberculosis (Mtb) can survive for a long time in vivo and evade host immune attacks, primarily through transcriptional regulation mediated by transcription regulators. JTY_2262 (homologous with Rv2250c) belongs to the Tetracycline Repressor (TetR) family of regulators, and its function is currently unclear. In this study, we combined JTY_2262 overexpressed transcriptomics and electrophoretic mobility shift assays (EMSA) to identify novel targets regulated by JTY_2262. The cofactors containing Cys, VC, VB1, VB3, VB6, Pb2+, Cu2+ and Li + inhibit the binding between JTY_2262 and th

The accessory protein CvnF8 modulates histidine kinase activity in an actinobacterial G protein system in Streptomyces coelicolor.

UNLABELLED: Conservons are operons that encode unusual regulatory systems found in bacteria of the phylum Actinomycetota. These regulatory systems are composed of four core proteins: a sensor histidine kinase-like protein (CvnA homolog), an MglB-type roadblock protein (CvnB homolog), a protein containing a domain of unknown function (CvnC homolog), and a small Ras-like GTPase (CvnD homolog). Based on their conserved small GTPase components and their phylogenetic distribution, we propose that the systems encoded by conservons should be known as actinobacterial G protein systems (AGPSs). The sig

The ArcAB two-component system is associated with the susceptibility of Aggregatibacter actinomycetemcomitans to superoxide and hydrogen peroxide.

Aggregatibacter actinomycetemcomitans is a Gram-negative facultative anaerobe and is associated with periodontal disease. This bacterium is exposed to environmental stresses, such as osmotic pressure, temperature shifts, pH shifts, and antimicrobial substances, including reactive oxygen species (ROS), in the human oral cavity. The bacterial two-component system ArcAB modulates gene expression in response to environmental changes, primarily by sensing oxygen pressure in several pathogens belonging to the γ-proteobacteria. It is also known to provide adaptation to ROS stress; however, its functi

Harnessing the Regulatory Effects of Streptococcus mutans Two-Component Signal Transduction Systems for Therapeutic Interventions Against Dental Caries.

Streptococcus mutans is considered the main pathogen causing dental caries and has a strong ability to establish biofilms and respond to environmental stimuli, which are essential for its survival and cariogenicity. Fourteen two-component signal transduction systems (TCSs) in S. mutans have been reported to regulate a broad range of physiological processes such as bacterial biofilm formation, acid resistance, competence development, and toxic oxygen metabolite resistance. These systems collectively contribute to the cariogenicity of S. mutans by coordinating adaptive responses to environmental

Activation of the ChvG-ChvI pathway promotes survival during cell wall stress in Agrobacterium tumefaciens.

Agrobacterium tumefaciens shifts from a free-living soil bacterium to a plant-invading state upon encountering the plant root microenvironment. The acid-induced two-component sensor system ChvG-ChvI drives this shift and triggers a complex transcriptional program that promotes host invasion and survival against host immune defenses. Remarkably, ChvG-ChvI is also activated under cell wall stress conditions, suggesting that the transcriptional response may have a broader function. Here, we find that blocking cell wall synthesis either genetically or chemically leads to ChvG-ChvI activation. Muta

The critical human pathogen Acinetobacter baumannii exhibits light-regulated circadian rhythms.

Acinetobacter baumannii is recognized as the paradigm of multidrug-resistant superbug, topping the WHO priority list of critical human pathogens. Interestingly, it senses and responds to blue light, which modulates global aspects of its physiology, including pathogenicity. We hypothesized that light could serve as a signal to synchronize the bacterial physiology to the host's behavior and/or to the environment. At environmental temperatures, light regulation is mainly governed by the photoreceptor BlsA. In this work, we identified the existence of daily rhythms in blsA promoter activity displa

Limited Diversity of Thermal Adaptation to a Critical Temperature in Zymomonas mobilis: Evidence from Multiple-Parallel Laboratory Evolution Experiments.

Laboratory evolution is an effective means of understanding microbial adaptation to the environment. We previously isolated four thermoadapted Zymomonas mobilis mutants, which showed a 2 °C rise in the critical high temperature (CHT), by performing multiple-parallel adaptation experiments. In the present study, the individual mutations in these mutants were intensively analyzed. Two mutations in each adapted mutant were found to primarily contribute to the increase in the upper temperature limit. RNA sequencing (RNA-seq) analysis revealed that the two mutations led to the upregulation of 79-18

A cytochrome bd repressed by a MarR family regulator confers resistance to metals, nitric oxide, sulfide, and cyanide in Chromobacterium violaceum.

UNLABELLED: Chromobacterium violaceum is a ubiquitous environmental pathogen. Despite its remarkable adaptability, little is known about the mechanisms of stress resistance in this bacterium. Here, in a screen for iron-susceptible transposon mutants, we identified a cytochrome bd that protects C. violaceum against multiple stresses. The two subunits of this cytochrome bd (CioAB) are encoded by the cioRAB operon, which also encodes a GbsR-type MarR family transcription factor (CioR). A ∆cioAB mutant strain was sensitive to iron and the iron-requiring antibiotic streptonigrin and showed a decrea

Regulatory mechanisms of two-component systems in Vibrio cholerae: Enhancing pathogenicity and environmental adaptation.

Cholera, which is caused by the bacterium Vibrio cholerae, is a highly dangerous disease characterized by severe symptoms such as watery diarrhea, dehydration, and even death. V. cholerae can both colonize the host intestine and survive in environmental reservoirs. Two-component systems (TCSs) are essential regulatory mechanisms that allow bacteria to adapt to changing environments. This review focuses on the regulatory mechanisms of TCS-mediated gene expression in V. cholerae. We first summarize the composition and classification of TCSs in V. cholerae N16961. We then discuss the roles of TCS

Staphylococcus aureus Stress Response to Bicarbonate Depletion.

Bicarbonate and CO2 are essential substrates for carboxylation reactions in bacterial central metabolism. In Staphylococcus aureus, the bicarbonate transporter, MpsABC (membrane potential-generating system) is the only carbon concentrating system. An mpsABC deletion mutant can hardly grow in ambient air. In this study, we investigated the changes that occur in S. aureus when it suffers from CO2/bicarbonate deficiency. Electron microscopy revealed that ΔmpsABC has a twofold thicker cell wall thickness compared to the parent strain. The mutant was also substantially inert to cell lysis induced b

Activation of CzcS/CzcR during zinc excess regulates copper tolerance and pyochelin biosynthesis of Pseudomonas aeruginosa.

UNLABELLED: Zinc is an important transition metal that is essential for numerous physiological processes while excessive zinc is cytotoxic. Pseudomonas aeruginosa is a ubiquitous opportunistic human pathogen equipped with an exquisite zinc homeostatic system, and the two-component system CzcS/CzcR plays a key role in zinc detoxification. Although an increasing number of studies have shown the versatility of CzcS/CzcR, its physiological functions are still not fully understood. In this study, transcriptome analysis was performed, which revealed that CzcS/CzcR is silenced in the absence of the z

MmpL3, Wag31, and PlrA are involved in coordinating polar growth with peptidoglycan metabolism and nutrient availability.

UNLABELLED: Cell growth in mycobacteria involves cell wall expansion that is restricted to the cell poles. The DivIVA homolog Wag31 is required for this process, but the molecular mechanism and protein partners of Wag31 have not been described. In this study of Mycobacterium smegmatis, we identify a connection between wag31 and trehalose monomycolate (TMM) transporter mmpl3 in a suppressor screen and show that Wag31 and polar regulator PlrA are required for MmpL3's polar localization. In addition, the localization of PlrA and MmpL3 is responsive to nutrient and energy deprivation and inhibitio

Lrp Family Regulator SCAB_Lrp2 Responds to the Precursor Tryptophan and Represses the Thaxtomin Biosynthesis in Streptomyces scabies.

Streptomyces scabies is a well-researched plant pathogen belonging to the genus Streptomyces. Its virulence is linked to the production of the secondary metabolite thaxtomin A, which is tightly regulated at the transcriptional level. The leucine-responsive regulatory protein (Lrp) family is conserved in prokaryotes and is involved in various crucial biological processes. However, the regulatory interaction between Lrp protein and pathogenic Streptomyces species remains poorly understood. This study aims to explore the role of SCAB_Lrp2 in regulating thaxtomin biosynthesis and pathogenicity, an

The role of rcpA gene in regulating biofilm formation and virulence in Vibrio parahaemolyticus.

Vibrio parahaemolyticus (V. parahaemolyticus) is a common seafood-borne pathogen that can colonize the intestine of host and cause gastroenteritis. Biofilm formation by V. parahaemolyticus enhances its persistence in various environments, which poses a series of threats to food safety. This work aims to investigate the function of rcpA gene in biofilm formation and virulence of V. parahaemolyticus. Deletion of rcpA significantly reduced motility, biofilm biomass, and extracellular polymeric substances, and inhibited biofilm formation on a variety of food and food contact surfaces. In mice infe

The histidine kinase NahK regulates denitrification and nitric oxide accumulation through RsmA in Pseudomonas aeruginosa.

UNLABELLED: Pseudomonas aeruginosa have a versatile metabolism; they can adapt to many stressors, including limited oxygen and nutrient availability. This versatility is especially important within a biofilm where multiple microenvironments are present. As a facultative anaerobe, P. aeruginosa can survive under anaerobic conditions utilizing denitrification. This process produces nitric oxide (NO) which has been shown to result in cell elongation. However, the molecular mechanism underlying this phenotype is poorly understood. Our laboratory has previously shown that NosP is a NO-sensitive hem

Plant-Derived Phenolic Acids Limit the Pathogenesis of Salmonella Typhimurium and Protect Intestinal Epithelial Cells during Their Interactions.

The incidence of gastrointestinal illness attributable to Salmonella enterica serovar Typhimurium (ST) remains a concern for public health worldwide, as it can progress into systemic infections mediated by the type-three secretion system (T3SS), which allows for adherence and invasion to intestinal epithelial cells. The current study evaluates the ability of gallic acid (GA), protocatechuic acid (PA), and vanillic acid (VA) to impair the adhesion and invasion abilities of ST to a human epithelial (INT-407) cell monolayer while also assessing their cytotoxicity. GA, PA, and VA inhibited detecta
