Amanitin-Based Fc-Small Molecule-Drug Conjugates with Noncleavable Linker: A Novel Therapeutic Strategy for Prostate Cancer Targeted Therapy.

Prostate cancer remains a major global health burden, with limited options and poor prognosis in advanced stages. To extend survival and improve the quality of life for patients, targeted therapies have become an emerging treatment modality. Antibody-drug conjugates (ADC) have shown huge clinical success but only limited therapeutic effects in prostate cancer as their large size limits tumor penetration. Small molecule-drug conjugates (SMDC), consisting of a small molecule as a binding moiety and a cytotoxic drug, offer advantages due to their smaller size, but their short plasma half-life and

Engineering Cathepsin S Selective Chemical Probes and Antibody-Drug Conjugates through Substrate Profiling with Unnatural Amino Acids.

Cysteine cathepsins, particularly cathepsin S, regulate proteolytic signaling in cancer progression and immune modulation, yet selective tools for individual cathepsins remain limited. Here, we report the design of cathepsin S-selective probes and cathepsin S-cleavable antibody-drug conjugates (ADCs) using substrate profiling with unnatural amino acids. Hybrid Combinatorial Substrate Library (HyCoSuL) screening identified selective tetrapeptide motifs that were used to develop optimized fluorogenic substrates, irreversible inhibitors, and fluorescent activity-based probes with high selectivity

CD74-Targeted Cathepsin-Inhibitor Antibody-Drug Conjugate Triggers Apoptosis in DLBCL.

Transcriptomic analyses of public datasets (TCGA and GTEx) revealed that both CD74 and Cathepsin L (CTSL) are significantly overexpressed in diffuse large B-cell lymphoma (DLBCL) compared to normal tissues, and that their expression levels are highly correlated to each other (Spearman R = 0.64, p = 3 × 10-46). Kaplan-Meier analysis showed that elevated expression of both genes is associated with reduced overall survival (OS), defining a high-risk CD74+/CTSL+ DLBCL subgroup. This is the first study demonstrating coordinated overexpression of CD74 and CTSL and proposing their dual targeting via

Disease control with disitamab vedotin in HER2-low polymorphous adenocarcinoma of the tongue base after chemotherapy failure: A case report.

Polymorphous adenocarcinoma (PAC) is a distinct subtype of salivary gland carcinoma (SGC), and its occurrence at the tongue base is uncommon. Effective systemic treatment options for patients with advanced disease remain limited. Although HER2-low expression has been identified in a subset of SGCs, evidence supporting the use of HER2-targeted antibody-drug conjugates (ADCs) in HER2-low PAC is currently lacking. CASE PRESENTATION: A 72-year-old female with HER2-low (immunohistochemistry 1+) PAC of the tongue base presented with cervical nodal metastases (cT3N2cM0, Stage IVA). The tumor progress

The Nonclinical Safety Assessment of a Novel Anti-CEACAM5 Antibody Exatecan Conjugate Predicts a Low Risk for Interstitial Lung Disease (ILD) in Patients-The Putative Mechanism Behind ILD.

The therapeutic window of antibody drug-conjugates (ADC) remains challenging due to safety issues such as interstitial lung disease (ILD) observed with specific deruxtecan-based ADCs. To avoid ILD, we designed M9140 by conjugating the maleimide-containing hydrophilic β-glucuronide linker to exatecan and our anti-CEACAM5 (CarcinoEmbryonic Antigen-related Cell Adhesion Molecule 5) specific antibody. Following repeated iv-infusion at 3 to 30 mg/kg of M9140 every 3 weeks, the pathological findings obtained in cynomolgus monkeys were confined to gastrointestinal and hematolymphoid tissues and resem

Antibody-Drug Conjugates as Novel Therapeutic Agents for Non-Small Cell Lung Carcinoma with or without Alterations in Oncogenic Drivers.

Antibody-drug conjugates (ADCs) are an emerging class of therapeutics for lung cancer, and several are currently in development for this malignancy. The structure of these molecules is based on an antibody that targets a protein on the lung cancer cell surface and a cytotoxic payload attached by a linker. Many protein targets, including TROP2, c-MET, CEACAM5, HER2, and HER3 have been identified. In metastatic non-small cell lung carcinoma (NSCLC) without alterations in oncogenic drivers, platinum-based chemotherapy and immune checkpoint inhibitors (ICIs) targeting the programmed death-1/progra

Sacituzumab govitecan in triple-negative breast cancer: from bench to bedside, and back.

Triple-negative breast cancer (TNBC) represents a major therapeutic challenge due to its heterogeneous and aggressive phenotype, and limited target-specific treatment options. The trophoblast cell surface antigen (Trop-2), a transmembrane glycoprotein overexpressed in various cancers, has emerged as a promising target for TNBC. Sacituzumab govitecan (SG), an antibody-drug conjugate (ADC) that targets Trop-2, has recently entered treatment algorithms for advanced and metastatic TNBC, independently from Trop-2 expression status, with manageable toxicity. Despite the impressive results, questions

Understanding the Effects of Site-Specific Light Chain Conjugation on Antibody Structure Using Hydrogen Exchange-Mass Spectrometry (HX-MS).

Antibody drug conjugates (ADCs) represent one of the fastest growing classes of cancer therapeutics. Drug incorporation through site-specific conjugation in ADCs leads to uniform drug load and distribution. These site-specific modifications may have an impact on ADC quality attributes including protein higher order structure (HOS), which might impact safety and efficacy. In this study, we conducted a side-by-side comparison between the conjugated and unconjugated mAb. In the ADC, the linker-pyrrolobenzodiazepine was site specifically conjugated to an engineered unpaired C215 residue within the

[Our experience with the treatment of Hodgkin lymphoma patients].

Overall and disease-free survival of Hodgkin lymphoma patients has improved significantly since the 2000s. This is due to the use of ABVD (adriamycin, bleomycin, vinblastine, dacarbazine) polychemotherapy and modern radiotherapy. In recent years, further diagnostic and therapeutic changes have been made, which further improve patients' survival. The most significant role in this is the improvement of diagnostics, such as the 18FDG-PET/CT, which is now routinely used repeatedly during treatment, and the response-adapted treatment(s) based on it. The main role of ABVD treatment in first-line tre
