Cultured Human Foreskin as a Model System for Evaluating Ionizing Radiation-Induced Skin Injury.

PURPOSE: Precise molecular and cellular mechanisms of radiation-induced dermatitis are incompletely understood. Histone variant H2A.J is associated with cellular senescence and modulates senescence-associated secretory phenotype (SASP) after DNA-damaging insults, such as ionizing radiation (IR). Using ex vivo irradiated cultured foreskin, H2A.J was analyzed as a biomarker of radiation-induced senescence, potentially initiating the inflammatory cascade of radiation-induced skin injury. METHODS: Human foreskin explants were collected from young donors, irradiated ex vivo with 10 Gy, and culture

A new assay capturing chromosome fusions shows a protection trade-off at telomeres and NHEJ vulnerability to low-density ionizing radiation.

Chromosome fusions threaten genome integrity and promote cancer by engaging catastrophic mutational processes, namely chromosome breakage-fusion-bridge cycles and chromothripsis. Chromosome fusions are frequent in cells incurring telomere dysfunctions or those exposed to DNA breakage. Their occurrence and therefore their contribution to genome instability in unchallenged cells is unknown. To address this issue, we constructed a genetic assay able to capture and quantify rare chromosome fusions in budding yeast. This chromosome fusion capture (CFC) assay relies on the controlled inactivation of

An investigation of the age-dependency of chromosome abnormalities in human populations exposed to low-dose ionising radiation.

Among various cytogenetic changes stable chromosome aberrations (SCHA) seem to be the most significant for ageing and carcinogenesis. Being nonlethal they can persist through cell divisions and accumulate in time. We studied the age response of SCHA (translocations and insertions) in normal and radiation exposed human populations. Two cohorts of people at the age range of 3--72 years were studied: control (43 persons) and exposed to low doses of accidental irradiation due to Chernobyl accident and atomic bomb testing in Semipalatinsk (67 persons). FISH method was used for visualisation of chro

[Relationship between transcription and repair of radioinduced DNA damage].

The DNA repair is occurred on genome level with different rates. This rates are depend on a transcription activity of various genome sequences. The repair rates are low in non-transcribed genes and DNA sequences, more higher in transcribed genes and the highest in actively transcribed strands of active genes. Such classification could be applied to the repair of UV-induced cyclobutane pyrimidine dimers and for other (but not for all) DNA lesions. This observation has become a basement of such important findings as involvement of transcription into repair or existence of transcription-coupling

Cytogenetic damage induced in human lymphocytes by low doses of 60Co gamma rays delivered at high and low dose rates.

Chromosomal aberrations were investigated in human peripheral blood lymphocytes after exposure to low doses of 60Co gamma rays delivered acutely or at low dose rates (0.1 or 0.03 Gy/h). Chromosome analysis was performed in cells collected after a 44 to 46 hour culture time in order to avoid scoring of second division cells, and the dose-related induction of aberrations was analysed by the maximum likelihood method. In all cases, the induction of dicentrics was well described by a linear-quadratic dose response model (Y = aD + bD2), the data obtained at a low dose rate being equally well fitted

Mitochondrial genetics. VI. The petite mutation in Saccharomyces cerevisiae: interrelations between the loss of the p+ factor and the loss of the drug resistance mitochondrial genetic markers.

The survival of the rho(+) factor and of Drug(R) mitochondrial genetic markers after exposure to ethidium bromide has been studied. A technique allowing the determination of Drug(R) genetic markers among a great number of both grande and petite colonies has been developed. The results have been analyzed by the target theory. The survival of the rho(+) factor is always less than the survival of any Drug(R) genetic marker. The survivals of C(R) and E(R) are similar to each other, while that of O(R) is greater than that of the other two Drug(R) markers. All possible combinations of Drug(R) marker

A note on the maternal effect mutants daughterless and abnormal oocyte in Drosophila melanogaster.

Two deficiencies for, and a dominant enhancer of, the second chromosome maternal effect mutant, "daughterless" (da), were induced with X-irradiation. Their properties were studied with respect to both da and the linked maternal effect mutant, "abnormal oocyte" (abo), with the following conclusions. (1) The most probable map positions of da and abo are: J-(1/2)-da-2(1/2)-abo, where J is a dominant marker located at 41 on the standard map. (2) The da locus is in bands 31CD-F on the polytene chromosome map; abo is to the right of 32A. (3) Because homozygous da individuals survive while individual

Melasma pathogenesis and influencing factors - an overview of the latest research.

Melasma is an acquired, symmetrical hypermelanosis of the face. The pathogenesis of melasma is complex and the treatment is often challenging with frequent relapses. Genetic background, exposure to ultraviolet radiation, and female sex hormones are classical influencing factors. To the light of the recent literature, other factors could promote melasma lesions. Moreover, there are increasing evidences showing that melanocytes are not the only cells involved, and that other players probably have a key role in the development and the relapses of melasma. Identifying those associated factors shou

Development of breast cancer in a 21-year-old childhood Wilms' tumor survivor with a BRCA1 2634delC mutation.

Breast cancer at age 21 is rare, even in individuals who have a genetic predisposition. These early diagnoses are usually the result of a hereditary cancer syndrome. Other contributing factors, such as chemotherapy and radiation for previous malignancies, can also increase the risk of secondary malignancies, including breast cancer. Here we present one of the youngest cases of breast cancer reported in the literature: a 21-year-old Wilms' tumor survivor (diagnosed at age 6), who was found to have a familial BRCA1 mutation and was diagnosed with ductal carcinoma in situ at age 21.

Alport syndrome: a genetic study of 31 families.

Thirty one families with Alport syndrome including 3 families with associated syndromes were studied. The location of the COL4A5 gene, responsible for the Alport syndrome, was determined by linkage analysis with eight probes of the Xq arm and by a radiation hybrid panel. Concordant data indicated the localization of the Alport gene between DXS17 and DXS11. Four deletions and one single base mutation of the COL4A5 gene were detected. Homogeneity tests failed to show any evidence of genetic heterogeneity superimposed on clinical heterogeneity for ophthalmic signs and end-stage renal disease age.

Preventative Options and the Future of Chemoprevention for Cutaneous Tumors.

Chemoprophylaxis against nonmelanoma skin cancer (NMSC) should be considered in high-risk populations such as those with certain genetic disorders, immunosuppressive states, chronic radiation, excessive UV exposure, or extensive personal or family history of NMSC. The methods for chemoprevention have progressed beyond traditional sunscreen into more effective strategies including DNA repair enzymes, nicotinamide, systemic retinoids, and nonsteroidal anti-inflammatory drugs. Other therapies are still being investigated and include treatments that target premalignant lesions, capecitabine, hedge

Glioblastoma multiforme: a glance at advanced therapies based on nanotechnology.

Glioblastoma multiforme (GBM, grade IV) is the most common malignant and invasive central nervous system tumor with poor survival outcome. Various pathogenesis signatures such as genetic mutation, hypoxia, necrosis and neo-angiogenesis are involved in GBM. Standard treatment includes surgical resection along with radiation therapy and temozolomide (TMZ) chemotherapy that do not improve the overall survival of patients. In this review, we focused on the diagnosis, risk factors and novel therapies, using advanced therapies such as nanotechnology in drug delivery, gene therapy and hyperthermia th

Bone marrow transplantation for genetic disorders.

Allogeneic bone marrow transplantation has been successfully utilized to correct the lymphoid and/or hematopoietic abnormalities characterizing a wide array of lethal genetic disorders. Examples include severe combined immunodeficiency, Wiskott-Aldrich syndrome, thalassemia major, sickle cell anemia, and several types of lysosomal storage diseases. Most marrow transplant recipients require preparation with myeloablative doses of chemotherapy, with or without additional radiation therapy, to ensure engraftment of allogeneic marrow. This approach has dramatically changed the long-term outlook fo

A genome-wide association study on radiation induced fibrosis in breast cancer patients.

BACKGROUND AND PURPOSE: Radiation-induced fibrosis affects women having undergone post-lumpectomy radiotherapy to a varying degree and remains a significant late morbidity for breast cancer patients. While Single Nucleotide Polymorphisms (SNPs) have been linked to fibrosis after radiotherapy (RT), the genetic architecture remains incompletely understood. We aimed to identify new genetic variants in a cohort of early breast cancer patients representing two cohorts treated within the Danish Breast Cancer Group (DBCG) protocols. MATERIAL AND METHODS: A genome-wide association study (GWAS) was co

Homozygous hypomorphic BRCA2 variant in primary ovarian insufficiency without cancer or Fanconi anaemia trait.

BACKGROUND: Primary ovarian insufficiency (POI) affects 1% of women under 40 years and is a public health problem. The genetic causes of POI are highly heterogeneous with isolated or syndromic forms. Recently, variants in genes involved in DNA repair have been shown to cause POI. Notably, syndromic POI with Fanconi anaemia (FA) traits related to biallelic BRCA2 truncated variants has been reported. Here, we report a novel phenotype of isolated POI with a BRCA2 variant in a consanguineous Turkish family. METHODS: Exome sequencing (ES) was performed in the patient. We also performed functional

Immunogenetic influences on the initiation stage of the cutaneous chemical carcinogenesis pathway.

While it is generally agreed that environmental exposure to solar radiation and to certain classes of chemicals are the major causes of nonmelanoma skin cancer, it is also believed that genetic polymorphisms regulating immunological responses are important determinants of individual susceptibility to skin cancer. However, little is known about their interactions with the chemical carcinogenesis pathway prior to the actual development of tumors. This issue was examined by comparing susceptibility to skin cancer in C3H/HeN and C3H/HeJ mice, two strains that differ only at the lipopolysaccharide
