Exosomes Derived from Human Umbilical Cord Mesenchymal Stem Cells Promote Ulcer Healing in Diabetes Mellitus Type 2 Mice.

BACKGROUND: With the passage of time, the incidence rate of diabetes continues to rise. As a common and serious complication of diabetes, the economic burden of diabetes ulcers is also increasing. However, there is currently no unified clinical treatment strategy for its repair and research, and the treatment effect is not satisfactory. Exosomes derived from MSCs play a crucial role in improving diseases. METHODS: This study used the explant culture method to isolate human umbilical cord mesenchymal stem cells (hucMSCs) from Wharton's jelly, which were identified by flow cytometry and differe

MicroRNA-489-3p plays a significant role in congenital hypothyroidism through regulating neuronal cell apoptosis via targeting translationally controlled tumor protein 1.

Accumulating reports have indicated that congenital hypothyroidism (CH) is an endocrine disorder caused by underdeveloped thyroid gland or thyroid dyshormonogenesis. It has been also reported that certain microRNAs (miRNAs) may exert protective effects against the development of CH. However, whether miR-489-3p regulates CH progression remains unclear. The aim of the present study was to investigate the effects of miR-489-3p on CH and elucidate the underlying mechanisms. Therefore, Sprague Dawley rats were injected with propylthiouracil (50 mg/day) to establish a CH model. Reverse transcription

Effect of resolution enhancement using metal ion assisted strategy based on electrospray ionization-ion mobility spectrometry: A case study of carbendazim and thiabendazole in fruits.

A method for the rapid and simultaneous determination of carbendazim and thiabendazole residues by electrospray ionization-ion mobility spectrometry (ESI-IMS) combined with a metal ion-assisted technique was developed and validated in different fruit matrices. The metal ion assisted strategy was performed instead of tedious pre-separation procedures to overcome the limitation of low resolution of IMS. Four transition metal cations, Co(II), Ni(II), Cu(II), and Zn(II), were screened and their interactions with carbendazim and thiabendazole were investigated. The injection flow rate and metal ion

Repurposed antipsychotic chlorpromazine inhibits colorectal cancer and pulmonary metastasis by inducing G2/M cell cycle arrest, apoptosis, and autophagy.

PURPOSE: Despite efforts in developing effective therapeutic strategies, colorectal cancer (CRC) remains one of the most prevalent and lethal neoplasms. Repurposing approved drugs is an alluring strategy for developing anticancer agents. Some antipsychotic drugs, including chlorpromazine (CPZ), possess anticancer activities. However, the pharmacological effects of CPZ on CRC have not been clearly established. METHODS: MTT assay, flow cytometry, western blotting analysis, subcutaneous mice tumor, and tail-vein-injection established lung metastasis model were used to investigate the anticancer

Reduction in cerebral perfusion after heroin administration: a resting state arterial spin labeling study.

Heroin dependence is a chronic relapsing brain disorder, characterized by the compulsion to seek and use heroin. Heroin itself has a strong potential to produce subjective experiences characterized by intense euphoria, relaxation and release from craving. The neurofunctional foundations of these perceived effects are not well known. In this study, we have used pharmacological magnetic resonance imaging (phMRI) in 15 heroin-dependent patients from a stable heroin-assisted treatment program to observe the steady state effects of heroin (60 min after administration). Patients were scanned in a cr

Quantitative assessment of cerebral hemodynamics using single photon emission computed tomography with venous blood sampling.

OBJECTIVE: Quantitative cerebral blood flow (CBF) measured by single photon emission computed tomography (SPECT) with arterial blood sampling is one of the most reliable methods to assess the hemodynamics in individual patients. SPECT with venous blood sampling is less invasive. The present study compared the measurement of CBF using N-isopropyl-p-(iodine-123)-iodoamphetamine SPECT with venous blood sampling and with arterial blood sampling in patients with major cerebral artery occlusive disease. METHODS: Two normal subjects and 14 patients with major cerebral artery occlusive disease underw

Peritoneal challenge modulates expression of pneumococcal surface protein C during bacteremia in mice.

Differential expression of pneumococcal virulence proteins has been demonstrated. We previously demonstrated challenge route-dependent differences in pneumococcal surface protein C (PspC) expression during bacteremia. In this study, we investigated differences in PspC expression during the transition of pneumococci from the peritoneum to the blood. Time course analysis of PspC expression using flow cytometry demonstrated that Streptococcus pneumoniae D39 collected from blood expressed significantly more PspC than did D39 collected from the peritoneum of intraperitoneally (i.p.)-infected mice.

A Novel Magnetically Targeted Intramedullary (MagIC-TI) Xenograft Model for Precise Leukemia Modeling and Drug Resistance Evaluation in the Bone Marrow Niche.

Acute myeloid leukemia (AML) remains a therapeutic challenge due to drug resistance and relapse, which are often driven by the protective bone marrow (BM) niche. Conventional xenograft models fail to adequately recapitulate this niche-specific pathophysiology. To overcome this limitation, a novel magnetically targeted intramedullary (MagIC-TI) xenograft model was developed. Magnetically labeled doxorubicin (DOX)-resistant HL60 cells (Mag-Re) were injected into the femurs of NSG (nonobese diabetic [NOD] Cg-PrkdcscidIL2rgtm1Wjl/SzJ) mice using a patented microinjection syringe under localized ma

Toll-like receptor 4 (TLR4) deficiency impedes Toxoplasma gondii excreted-secreted antigens (ESA)-induced abortion.

INTRODUCTION: Toxoplasma gondii is an opportunistic intracellular parasite that is a major pathogenic factor in miscarriage, especially when it occurs early in pregnancy. We have previously demonstrated that the regulation of forkhead box transcription factor (Foxp3) is associated with abortion in early pregnancy caused by excretory-secretory antigen (ESA) of strain China 1. We aimed to reveal the underlying mechanism of miscarriage caused by ESA. METHODS: A TLR4-/- pregnant mouse model was successfully constructed. Pregnant mice at gestational day 5 (G5) were injected with ESA. All animals w
