Pretreatment with flurazepam (3 mg/kg s.c.) or chlordiazepoxide (5.5-30 mg/kg s.c.) at an ambient temperature of 34 degrees C, markedly enhanced the cataleptic response of mice to delta-9-tetrahydrocannabinol (THC; 5-20 mg/kg i.p.) as measured in a bar test. A double-blind comparative trial was carried out in 64 depressed out-patients, comparing mianserin with a fixed combination of amitriptyline + chlordiazepoxide. Both preparations proved to be equally effective at the end of the trial, but the mianserin group showed a considerable therapeutic improvement at the end of the second week, in comparison to the other group.
