Use of Artificial Intelligence to Identify New Mechanisms and Approaches to Therapy of Bone Disorders Associated With Chronic Kidney Disease.

Chronic kidney disease (CKD) leads to clinically severe bone loss, resulting from the deranged mineral metabolism that accompanies CKD. Each individual patient presents a unique combination of risk factors, pathologies, and complications of bone disease. The complexity of the disorder coupled with our incomplete understanding of the pathophysiology has significantly hampered the ability of nephrologists to prevent fractures, a leading comorbidity of CKD. Much has been learned from animal models; however, we propose in this review that application of multiple techniques of mathematical modeling

Leveraging quantitative systems pharmacology and artificial intelligence to advance treatment of chronic kidney disease mineral bone disorder.

Chronic kidney disease mineral bone disorder (CKD-MBD) is a complex clinical syndrome responsible for the accelerated cardiovascular mortality seen in individuals afflicted with CKD. Current approaches to therapy have failed to improve clinical outcomes adequately, likely due to targeting surrogate biochemical parameters as articulated by the guideline developer, Kidney Disease: Improving Global Outcomes (KDIGO). We hypothesized that using a Systems Biology Approach combining machine learning with mathematical modeling, we could test a novel approach to therapy targeting the abnormal movement

Isolation and artificial production of atypical porcine pestivirus, using the swine-kidney-derived cell line SK-L.

Congenital tremor (CT) in piglets was first reported in 1922, and although the causative pathogen was unknown for many years, atypical porcine pestivirus (APPV) was recently shown to be the cause. APPV is difficult to isolate, and there have been few reports of APPV isolated from field materials. Here, we successfully isolated infectious particles from a tonsillar emulsion from a CT-affected piglet using the established swine-kidney-derived cell line SK-L. In addition, we produced APPV artificially using these cells. Thus, SK-L cells are useful for both isolation and artificial production of A

Extracorporeal organ support (ECOS) in critical illness and acute kidney injury: from native to artificial organ crosstalk.

The complex nature of single organ failure potentially leading to multiple organ dysfunction syndrome (MODS) in critically ill patients necessitates integrated supportive therapy. Rather than a primary disease, acute kidney injury (AKI) is considered a window to a potentially serious underlying systemic disease, which may partially explain the high morbidity and mortality rates associated with the condition. Renal replacement therapy (RRT) has been routinely used for more than a decade in various intensive care settings and there has also been an increase in the use of extracorporeal membrane

Artificial Urinary Sphincter With Bilateral Atrophic Kidneys and Accessory Renal Arteries in a Male Cadaveric Subject: A Case Report and Clinicopathological Reconciliation of Urinary Abnormalities and Embryogenetic Correlation of Vascular Aberrations.

A unique combination of triple abnormality in a willed male body donor dissection, with putative clinicopathological correlations during the subject's lifetime, is described in this case report. The subject had a three-piece artificial urinary sphincter surgically implanted around the proximal corpus spongiosum, left scrotal pouch and in the lower left abdominal wall, ostensibly for urinary incontinence during his lifetime, though the etiology of the latter was not immediately obvious. He also had a total of three accessory renal arteries involving both sides, complicated by bilateral diffuse

Prediction of Five-Year Mortality Risk of Chronic Kidney Disease Using Artificial Intelligence-Based Models: A Retrospective Study.

BACKGROUND AND AIMS: Most countries are experiencing an increasing trend of chronic kidney disease (CKD). Preventive strategies, such as early prediction of CKD progression and mortality, are crucial in reducing this disease rate. Although artificial intelligence (AI) models have demonstrated outstanding predictive performance in the prognosis of CKD, no study has been conducted on the 5-year mortality risk of this disease. This study aims to perform this task to gain a deeper understanding of this technology and optimize preventive and treatment strategies. METHODS: This retrospective study

A representative mechanistic model of multisolute transport in low-flow hollow-fiber dialyzers for wearable artificial kidney-oriented design.

To support the analysis of compact hollow-fiber dialyzers operated under low-flow conditions relevant to wearable artificial kidney development, a mechanistic model of multisolute transport was developed. Unlike conventional homogeneous-membrane descriptions, the dialysis membrane was represented as a three-layer asymmetric porous structure. Within a continuum framework, pore-scale hindered diffusion, ultrafiltration-driven convection, and transmembrane hydraulics were coupled to describe diffusion-convection transport under low-flow conditions. Comparisons with manufacturer-stylein vitroclear

Emerging multidimensional biomarker system for cardiovascular-kidney-metabolic syndrome: from multi-omics integration to clinical artificial intelligence.

Cardiovascular-kidney-metabolic (CKM) syndrome is an emerging clinical entity that highlights the complex, bidirectional interplay among cardiovascular disease, chronic kidney disease, and metabolic disorders, representing a substantial and growing global health burden. This conceptualization marks a paradigm shift from viewing these conditions in isolation to understanding them as an interconnected disease continuum. Traditional biomarkers face significant limitations in the early detection, risk stratification, and precise management of CKM, necessitating a transition towards an integrated f

Artificial intelligence assisted risk prediction in organ transplantation: a UK Live-Donor Kidney Transplant Outcome Prediction tool.

Introduction: Predicting the outcome of a kidney transplant involving a living donor advances donor decision-making donors for clinicians and patients. However, the discriminative or calibration capacity of the currently employed models are limited. We set out to apply artificial intelligence (AI) algorithms to create a highly predictive risk stratification indicator, applicable to the UK's transplant selection process. Methodology: Pre-transplant characteristics from 12,661 live-donor kidney transplants (performed between 2007 and 2022) from the United Kingdom Transplant Registry database wer

GDF15, EGF, and Neopterin in Assessing Progression of Pediatric Chronic Kidney Disease Using Artificial Intelligence Tools-A Pilot Study.

Cell-mediated immunity and chronic inflammation are hallmarks of chronic kidney disease (CKD). Growth differentiation factor 15 (GDF15) is a marker of inflammation and an integrative signal in stress conditions. Epidermal growth factor (EGF) is a tubule-specific protein that modulates the regeneration of injured renal tubules. Neopterin is a product of activated monocytes and macrophages and serves as a marker of cell-mediated immunity. Our aim was to assess the role of the above-mentioned parameters in the progression of CKD in children using artificial intelligence tools. The study group con

Artificial Intelligence-Driven Risk Stratification in Chronic Kidney Disease Progression: Minimizing Bias via Race-Specific Algorithms.

Background Chronic kidney disease (CKD) is a prevalent condition that affects a substantial portion of the adult population and progresses unevenly across different demographic groups. Recent updates to estimated glomerular filtration rate (eGFR) estimation have removed race adjustments to promote greater equity. Yet, the impact of such changes on model performance and fairness across populations remains uncertain. Objective To ascertain whether, in comparison to a traditional pooled (or "race-blind") model, a race-specific, modular deep-learning architecture can enhance clinical utility and f

Strict blood glucose control by an artificial endocrine pancreas during hepatectomy may prevent postoperative acute kidney injury.

The aim of the present study was to evaluate the usefulness of a closed-loop system (STG-55; Nikkiso, Tokyo, Japan), a type of artificial endocrine pancreas for the continuous monitoring and control of intraoperative blood glucose, for preventing postoperative acute kidney injury (AKI) in patients undergoing hepatectomy. Thirty-eight patients were enrolled in this study. Glucose concentrations were controlled with either a manual injection of insulin based on a commonly used sliding scale (manual insulin group, n = 19) or the programmed infusion of insulin determined by the control algorithm o
