Long-range axonal projections of transplanted mouse embryonic stem cell-derived hypothalamic neurons into adult mouse brain.

The hypothalamus is comprised of heterogenous cell populations and includes highly complex neural circuits that regulate the autonomic nerve system. Its dysfunction therefore results in severe endocrine disorders. Although recent experiments have been conducted for in vitro organogenesis of hypothalamic neurons from embryonic stem (ES) or induced pluripotent stem (iPS) cells, whether these stem cell-derived hypothalamic neurons can be useful for regenerative medicine remains unclear. We therefore performed orthotopic transplantation of mouse ES cell (mESC)-derived hypothalamic neurons into adu

The morphological and histochemical neurosecretory magnocellular system in the rat after administration of chlorpromazine.

The effect of chlorpromazine (CPZ) on the neurosecretory action of the hypothalamo- hypophyseal system was investigated in 72 male rats. The experimental animals received CPZ in a dose of 0.4 mg, 4.0 mg and 20.0 mg/kg b.w. for 30 days. The rats were sacrificed by decapitation at 24 h and 7 days after the last dose of the drug. The neurosecretory material was stained with paraldehyde fuchsin in the supraoptic nucleus, paraventricular nucleus, eminentia mediana and neurohypophysis, the tigroid was stained with toluidine blue and the acid phosphatase activity was evaluated histoenzymatically. It

Little or no response to 24-hr water-deprivation of Fos-like immunoreactivity in vasopressinergic magnocellular neurons in the hypothalamus of hereditary microphthalmic rats.

In normal rats, 24-hr water-deprivation was found to cause a significant increase in the plasma concentration of arginine vasopressin (AVP), and marked induction of Fos in the AVP-positive magnocellular neurons in the paraventricular nucleus (PVN) and the supraoptic nucleus (SON) of the hypothalamus. On the other hand, 24-hr water-deprivation also caused a significant increase in the plasma AVP concentration in hereditary microphthalmic rats, but much less than in normal rats. Consistent with this, the extent of induction of Fos in the nuclei of the PVN and SON of these rats was lesser than in

Endoplasmic reticulum stress in vasopressin neurons of familial diabetes insipidus model mice: aggregate formation and mRNA poly(A) tail shortening.

The immunoglobulin heavy chain binding protein (BiP) is an endoplasmic reticulum (ER) chaperone, which binds to newly synthesized secretory and transmembrane proteins to facilitate protein folding. BiP mRNA is expressed in the arginine vasopressin (AVP) neurons in the supraoptic nucleus of wild-type mice even in basal conditions, and the expression levels increase in response to dehydration. These data suggest that AVP neurons are subjected to ER stress. Familial neurohypophysial diabetes insipidus (FNDI) is caused by mutations in the gene locus of AVP. The mutant proteins could accumulate in

The topography and cytoarchitectonics of the nuclei of supraoptic and preoptic areas in insectivores.

In regio supraoptica there are nucleus paraventricularis, nucleus supraopticus, area hypothalamica anterior and nucleus suprachiasmaticus. These centers are more developed in hedgehog than in mole and common shrew. Area preoptica is divided into 4 parts (medial, lateral, periventricular and middle) from among the middle part in hedgehog is less developed than in other two species. Only in common shrew within area preoptica there is accessory cell group (nucleus preopticus accessorius).

Atrial natriuretic peptides in brain nuclei of rats with inherited diabetes insipidus (Brattleboro rats).

The concentration of atrial natriuretic peptides (ANF) was measured radioimmunologically in 18 selected microdissected brain areas of rats with inherited diabetes insipidus (Brattleboro rats) and their control rats (Long-Evans rats). In 7 brain areas known to be involved in the regulation of fluid homeostasis or blood pressure (subfornical organ, organum vasculosum lamina terminalis, periventricular preoptic nucleus, perifornical nucleus, nucleus of the solitary tract, tegmentum pontis, arcuate nucleus) ANF concentration was significantly changed. After restoration of antidiuresis with the V2

Mechanism and function of phasic firing in vasopressin-releasing magnocellular neurosecretory cells.

Magnocellular neurosecretory cells that release vasopressin (MNCVP ) from axon terminals in the neurohypophysis display a unique pattern of action potential firing termed phasic firing. Under basal conditions, only a small proportion of MNCVP display spontaneous phasic firing. However, acute and chronic conditions that stimulate vasopressin release, such as hemorrhage and dehydration, greatly enhance the number of MNCVP that fire phasically. Phasic firing optimizes VP neurosecretion at axon terminals by allowing action potential broadening to promote calcium-dependent frequency-facilitation, a

Modulation of synaptic inputs in magnocellular neurones in a rat model of cancer cachexia.

In cancer cachexia, abnormal metabolism and neuroendocrine dysfunction cause anorexia, tissue damage and atrophy, which can in turn alter body fluid balance. Arginine vasopressin, which regulates fluid homeostasis, is secreted by magnocellular neurosecretory cells (MNCs) of the hypothalamic supraoptic nucleus. Arginine vasopressin secretion by MNCs is regulated by both excitatory and inhibitory synaptic activity, alterations in plasma osmolarity and various peptides, including angiotensin II. In the present study, we used whole-cell patch-clamp recordings of brain slices to determine whether h

Beyond the GnRH axis: kisspeptin regulation of the oxytocin system in pregnancy and lactation.

Circulating oxytocin is critical for normal birth and lactation. Oxytocin is synthesised by hypothalamic supraoptic and paraventricular neurons and is released from the posterior pituitary gland into the circulation. Oxytocin secretion depends on action potentials initiated at the cell body, and we have shown that intravenous (IV) administration of kisspeptin-10 transiently increases the firing rate of supraoptic nucleus oxytocin neurons in anaesthetised, non-pregnant, pregnant and lactating rats. This peripheral effect is likely via vagal afferent input, because disruption of vagal afferents

Sexually dimorphic distribution of calcium-binding protein, calretinin in the preoptic area of the freshwater catfish, Clarias batrachus (Linn.).

Preoptic area (POA) plays an important role in the hormonal regulation of the pituitary gland in vertebrates. In this study we report the sexually dimorphic distribution of calcium-binding proteins calretinin (CR) in the POA in the freshwater catfish, Clarias batrachus. Nissl staining highlighted the presence of the nucleus praeopticus periventricularis (NPP) and other subdivisions of the nucleus praeopticus (NPO), including supraoptic (NPOs), paraventricular (NPOp) and magnocellular (NPOm) divisions. In NPO, CR immunoreactivity was noted only in females but not in males. In both sexes, CR sta

[Gomori-positive elements of the hypothalamo-hypophyseal neurosecretory system of the rat (immunohistochemical study)].

Vasopressiergic and oxytocinergic cells and fibers in the hypothalamo-hypophysial neurosecretory system have been identified by means of immuno-histochemical reactions. Vasopressin-producing cells localize mainly in the ventral part of the supraoptic nucleus (SON) and in the dorsolateral part of the paraventricular nucleus (PVN). Oxytocin-producing cells predominantly concentrate in the dorsal part of the SON and in the ventromedial part of the PVN. In the central part of the posterior pituitary lobe vasopressin-containing fibers are mainly situated, and in the peripheral parts--both oxytocin-

Reduced Numbers of Corticotropin-Releasing Hormone Neurons in Narcolepsy Type 1.

Narcolepsy type 1 (NT1) is a chronic sleep disorder correlated with loss of hypocretin(orexin). In NT1 post-mortem brains, we observed 88% reduction in corticotropin-releasing hormone (CRH)-positive neurons in the paraventricular nucleus (PVN) and significantly less CRH-positive fibers in the median eminence, whereas CRH-neurons in the locus coeruleus and thalamus, and other PVN neuronal populations were spared: that is, vasopressin, oxytocin, tyrosine hydroxylase, and thyrotropin releasing hormone-expressing neurons. Other hypothalamic cell groups, that is, the suprachiasmatic, ventrolateral

Hexokinase I messenger RNA in the rat central nervous system.

Hexokinase I (ATP:D-hexose 6-phosphotransferase, EC 2.7.1.1) is the first enzyme required in the metabolism of glucose in the central nervous system and plays a major role in regulation of the cerebral glycolytic rate. The distribution of hexokinase I mRNA was examined throughout the central nervous system of the rat by use of oligonucleotide probes and in situ hybridization histochemistry. In the rhinencephalon, strong hexokinase I mRNA labeling was demonstrated in the glomerular, mitral, internal granular, and internal plexiform layers, whereas the olfactory nerve, external plexiform, and su

Metabolic activation of efferent pathways from the rat area postrema.

We used the quantitative [14C]deoxyglucose method and autoradiography to evaluate metabolic activity in 47 individual cerebral structures or subregions that are part of neural pathways emanating from the brain stem circumventricular organ, area postrema. Electrical stimulation of the dorsocentral area postrema in halothane-ventilated rats produced hypotension and increased glucose metabolism by several structures within the ascending trajectories of efferent neural projections from the nucleus. Structures in the caudal medulla oblongata, including three subnuclei of the nucleus of the solitary

Age-dependent accumulation of hybrid vasopressin-oxytocin gene products but not hybrid oxytocin-vasopressin products in the endoplasmic reticulum of Brattleboro rats.

The age-dependence of the incidence of magnocellular neurosecretory neurons containing abnormal accumulations of peptide in the rough endoplasmic reticulum was examined in homozygous Brattleboro rats and in their wild-type Long Evans counterparts. Neurons in which the immunophenotype of the peptide aggregates indicate that somatic cross-over mutations involving the 5' end of the vasopressin gene and the 3' end of the oxytocin gene have occurred, increased with age in homozygous Brattleboro rats, reaching a maximum of 24 cells per hypothalamus (approximately 0.6% of the vasopressin neurons). [Reaction of the neurosecretory system of dogs with reno-vascular hypertension in the postnatal ontogenesis].

In dogs of different age with simulated renovascular hypertension neurosecretion of the hypothalamo-neurohypophysial system was studied. Experiments were conducted with puppies 18-22 days, 11/2-3 months and dogs 3-5 years old. It was established that in the ontogenesis of dogs the differentiation of neurons in the supraoptic nucleus occurred earlier than that of cells in the paraventricular nucleus. The simulation of hypertension accelerates the differentiation of tbe neurosecretory cells. The development of arterial hypertension leads to prevalence of precesses of the neurosecretory material

Nervous mechanisms of restraint water-immersion stress-induced gastric mucosal lesion.

Stress-induced gastric mucosal lesion (SGML) is one of the most common visceral complications after trauma. Exploring the nervous mechanisms of SGML has become a research hotspot. Restraint water-immersion stress (RWIS) can induce GML and has been widely used to elucidate the nervous mechanisms of SGML. It is believed that RWIS-induced GML is mainly caused by the enhanced activity of vagal parasympathetic nerves. Many central nuclei, such as the dorsal motor nucleus of the vagus, nucleus of the solitary tract, supraoptic nucleus and paraventricular nucleus of the hypothalamus, mediodorsal nucl

Alterations in the nuclear volume of neurones from the hypothalamic magnocellular nuclei of fetuses born to rats subject to chronic alcoholism.

Previous authors have noticed alterations of the neuroendocrine system in the offspring of chronically alcoholic animals. Forty Sprague-Dawley rats, were used fed a liquid control diet and a 5%-alcohol liquid diet (Lieber and DeCarli). They were divided into four groups: Control (C), Pair Fed (P), Embryonic alcohol (E) and Fetal alcohol (F). Results obtained in the statistical analysis were as follows: from the paraventricular nucleus: between E and F, E and P, E and C, p less than 0.001. F and C p less than 0.01. P and C p less than 0.005. Between F and P there were no significant differences

TRPV1 gene deficiency attenuates miniature EPSC potentiation induced by mannitol and angiotensin II in supraoptic magnocellular neurons.

The release of arginine vasopressin (AVP) from the magnocellular neurosecretory cells (MNCs) in the supraoptic nucleus (SON) is crucial for body fluid homeostasis. The MNC activity is modulated by synaptic inputs and humoral factors. A recent study demonstrated that an N-terminal splice variant of the transient receptor potential vanilloid type 1 (TRPV1) is essential for osmosensory transduction in the SON. In the present study, we examined the effects of mannitol and angiotensin II on miniature EPSCs (mEPSCs) in the supraoptic MNCs using whole-cell patch-clamp recording in in vitro slice prep
