Unlocking the molecular engineering of Geobacillus glycoside hydrolases as a source of industrial biocatalysts.

This review examines Geobacillus sensu stricto as a source of thermostable glycoside hydrolases (GH) for biomass conversion, food processing, and enzyme engineering. Recent peer-reviewed literature was assessed with emphasis on taxonomy, genome-based Carbohydrate-Active Enzymes (CAZyme) prediction, biochemical validation, structural data, and engineering case studies. Taxonomic boundaries were interpreted using current Anoxybacillaceae frameworks, with Parageobacillus treated as a related comparator rather than as Geobacillus. The strongest evidence supports GH13 alpha-amylases, xylan-active s

Co-Producing Reducing Sugars, Furfural and Xylo-Oligosaccharides from Reeds via Effective Pretreatment by Ternary Deep Eutectic Solvent Lactic Acid:Cetyltrimethylammonium Bromide:CuCl₂.

Recently, deep eutectic solvents (DESs) have attracted much attention for biomass pretreatment. This study explored the use of a ternary DES system to enhance the lignin removal during the pretreatment of biomass and improve the enzymolysis efficiency of residual biomass after pretreatment. It also investigated the possibility of co-production of furfural and xylo-oligosaccharides. A ternary DES system synthesized using lactic acid (LA), cationic surfactant cetyltrimethylammonium bromide (CTAB) and metal chloride CuCl₂ was successfully used to achieve highly efficient removal of lignin and xyl

Ammonia-Derived Acidic Compound Pretreatment: Fractionation of Lignocellulosic Biomass for Boosting Enzymatic Hydrolysis to Efficiently Produce Bioethanol by Yeast.

Development of pretreatment to reduce biomass recalcitrance is essential to produce fermentable sugars, which is used as carbon sources for microorganisms to synthesize biofuels. Lignin in lignocellulose is generally removed by alkaline pretreatment (e.g., NH4OH, ethanediamine), while the severe environment causes equipment corrosion and forms toxic inhibitors. Using ─NH2/NH4 +-derived acidic compounds (ADACs) might achieve ideal lignin removal efficiency and fermentability. Herein, five ADACs including ammonium chloride (AC), ammonium sulfate (AS), ammonium acetate (AA), butanamide (BuA), and

Isolation of cellulolytic phycomycete fungi from the caecum of the horse.

Microscopic examination of horse caecum contents revealed vegetative growth of phycomycete fungi on particles of digesta, and uniflagellated cells similar to fungal zoospores in the liquid phase. Three morphologically distinct isolates of strictly anaerobic phycomycete fungi were obtained from the caecum contents and cultured in vitro. Two of the isolates were able to utilize a wide range of plant carbohydrates for growth, including alpha-cellulose, xylan and particulate starch, and extensively digested water-insoluble plant tissues.

Identification of enzymes from genus Trichoderma that can accelerate formation of ferulic acid and ethyl ferulate in collaboration with rice koji enzyme in sake mash.

The enzymes responsible for acceleration of ferulic acid and ethyl ferulate formation in sake mash were studied. Ferulic acid and ethyl ferulate are formed during the sake brewing process from feruloylated glucuronoarabinoxylan. Cellulase reagent from genus Trichoderma was used instead of rice koji, because rice koji for sake brewing produces extremely low levels of xylan-degrading enzymes. A combination of the reagent with rice koji enzymes accelerated the formation of ferulic acid from α-rice powder. Addition of the reagent to sake mash increased ferulic acid and ethyl ferulate formation. Th

Utilizing Anaerobic Fungi for Two-stage Sugar Extraction and Biofuel Production from Lignocellulosic Biomass.

Lignocellulosic biomass is a vast and underutilized resource for the production of sugars and biofuels. However, the structural complexity of lignocellulosic biomass and the need for multiple pretreatment and enzymatic steps for sugar release renders this process economically challenging. Here, we report a novel approach for direct, single container, exogenous enzyme-free conversion of lignocellulosic biomass to sugars and biofuels using the anaerobic fungal isolate strain C1A. This approach utilizes simple physiological manipulations for timely inhibition and uncoupling of saccharolytic and f

Two β-xylanases from Aspergillus terreus: characterization and influence of phenolic compounds on xylanase activity.

Sugarcane bagasse was used as an inexpensive alternative carbon source for production of β-xylanases from Aspergillus terreus. The induction profile showed that the xylanase activity was detected from the 6th day of cultivation period. Two low molecular weight enzymes, named Xyl T1 and Xyl T2 were purified to apparent homogeneity by ultrafiltration, gel filtration and ion exchange chromatographies and presented molecular masses of 24.3and 23.60 kDa, as determined by SDS-PAGE, respectively. Xyl T1 showed highest activity at 50 °C and pH 6.0, while Xyl T2 was most active at 45 °C and pH 5.0. Mas

Development of an Efficient Bifunctional Endoxylanase/Feruloyl Esterase Chimera for Coproduction of Ferulic Acid, p-Coumaric Acid, and Xylooligosaccharides.

Bifunctional endoxylanases/feruloyl esterases that can efficiently produce hydroxycinnamic acids and xylooligosaccharides from agricultural residues are highly desirable but underexplored. Here, we report the development of a bifunctional chimera, XynB-SnT-FaeB, which shares low sequence identity (<23%) with its homologues and contains a novel linker peptide. It can hydrolyze model hydroxycinnamate esters and xylans. Due to interdomain synergism, its feruloyl esterase and endoxylanase catalytic efficiencies (kcat/Km) toward feruloylated arabinoxylan were 2.95 and 1.60 times those of FaeB and X…

Streptomyces scabies 87-22 possesses a functional tomatinase.

The actinomycete Streptomyces scabies 87-22 is the causal agent of common scab, an economically important disease of potato and taproot crops. Sequencing of the S. scabies 87-22 genome revealed the presence of a gene with high homology to the gene encoding the alpha-tomatine-detoxifying enzyme tomatinase found in fungal tomato pathogens. The tomA gene from S. scabies 87-22 was cotranscribed with a putative family 1 glycosyl hydrolase gene, and purified TomA protein was active only on alpha-tomatine and not potato glycoalkaloids or xylans. Tomatinase-null mutants were more sensitive to alpha-to…

Fecal fermentation behavior and immunomodulatory activity of arabinoxylan from wheat bran.

Arabinoxylan (AX) is the predominant non-starch polysaccharide in wheat bran, known for its significant immunomodulatory activity. However, existing literature lacks comprehensive studies on AX fermentation by gut microbiota and its subsequent immunomodulatory mechanisms. In the present study, we aimed to investigate the effects of AX on the composition of gut microbiota and the characteristics of its immunomodulatory activity. For this purpose, an in vitro fermentation system and a cyclophosphamide-induced immunosuppressed mouse model were established to explore both the in vitro and in vivo

Exploring the gelling properties of Plantago ovata-based Arabinoxylan: Fabrication and optimization of a topical emulgel using response surface methodology.

Prime objective of the current research was to develop a stable nimesulide emulgel with the help of arabinoxylan, a natural gelling agent extracted from Plantago ovata. The response surface methodology was used by a Design Expert 10 software to formulate and optimize the emulgel. The experimental design approach evaluated the impact of independent and dependent variables. Independent variables were different concentrations of arabinoxylan, span 80 and tween 20, whereas, dependent variables were viscosity, pH, and content uniformity. FTIR demonstrated the compatibility of nimesulide with the ex

Arabinoxylan and Pectin Metabolism in Crohn's Disease Microbiota: An In Silico Study.

Inflammatory bowel disease is a chronic disorder including ulcerative colitis and Crohn's disease (CD). Gut dysbiosis is often associated with CD, and metagenomics allows a better understanding of the microbial communities involved. The objective of this study was to reconstruct in silico carbohydrate metabolic capabilities from metagenome-assembled genomes (MAGs) obtained from healthy and CD individuals. This computational method was developed as a mean to aid rationally designed prebiotic interventions to rebalance CD dysbiosis, with a focus on metabolism of emergent prebiotics derived from

CAZyme-guided modification of cactus and regional polysaccharides for food hydrocolloid applications.

Food-hydrocolloid performance depends on polymer architecture and processing history rather than ingredient name alone. This critical narrative review evaluates CAZyme-guided modification of cactus-derived pectic materials and related regional polysaccharides for food hydrocolloid applications. Opuntia mucilage and pectic fractions are the focal substrates; non-cactus pectins provide the closest mechanistic evidence, and arabinoxylan is retained only as a bounded comparison for substitution and oxidative crosslinking. Starch is considered only as a limited process benchmark and is not used to

Mass distribution profiling of cell wall matrix polysaccharide biosynthesis mutants reveals alterations in non-targeted matrix polysaccharides.

Plant cell walls contain multiple matrix polysaccharides whose interactions help determine wall mechanics, growth, and integrity, but it remains unclear how disruption of one biosynthetic or modification pathway reshapes other, non-target polymers. We used gel permeation chromatography coupled with enzyme-linked immunosorbent assay (GPC-ELISA) to compare mass distributions of major matrix polysaccharides in Arabidopsis thaliana mutants affecting xylan biosynthesis and decoration (irx14L, irx15, gux1-1, and gux2-2), glucomannan synthesis (csla2-1), pectin homogalacturonan (HG) biosynthesis (gau

A green biomass pretreatment strategy to produce 3-hydroxypropionic acid and ethanol with engineered Saccharomyces cerevisiae, Rhodosporidium toruloides, and Issatchenkia orientalis.

Chemical and water overconsumption involved in solid-liquid separation and post-washing after biomass pretreatment pose significant challenges to scaling up the production of platform chemicals and biofuels. This study explored a novel one-pot biomass valorization strategy that merges CaO pretreatment of corn stover (CS) or brewer's spent grain (BSG) with CO2 carbonation. The pretreated biomass slurry was used for 3-hydroxypropionic acid (3-HP) or ethanol production without solid-liquid separation or post-washing. Results showed that carbonation reduced the pH to around 6 of BSG slurries while

Cost impact of hexose-to-pentose sugar ratios for biomanufacturing.

Central to the long-term vision for biomanufacturing is the ability to deconstruct plant cell walls to sugars that microbes can convert to products. Aside from glucose, the most abundant sugar in biomass is xylose, a pentose sugar. Industrially relevant microbes have been engineered to co-ferment xylose and glucose. Most nth plant technoeconomic analyses (TEAs) assume similar consumption rates and product yields for both sugars, but in reality, xylose is consumed more slowly. Feedstocks can be selected, or engineered, to alter the glucan-to-xylan ratio (GXR) but no TEAs have quantified the imp

Enhancing anaerobic digestibility and phosphorus recovery of dairy manure through microwave-based thermochemical pretreatment.

Anaerobic digestion and struvite precipitation are two effective ways of treating dairy manure for recovering biogas and phosphorus. Anaerobic digestion of dairy manure is commonly limited by slow fiber degradation, while struvite precipitation is limited by the availability of orthophosphate. The aim of this work is to study the possibility of using microwave-based thermochemical pretreatment to simultaneously enhance manure anaerobic digestibility (through fiber degradation) and struvite precipitation (through phosphorus solubilization). Microwave heating combined with different chemicals (N

A hierarchical surrogate approach to biomass ethanolysis reaction kinetic modelling.

The reaction mechanism and kinetics of the sulfuric acid catalysed ethanolysis of glucose, cellulose, xylan, and corncob were investigated using a combination of experiments and empirical reaction mechanism modelling. The experimental study was carried out in ethanol at various temperatures between 150 °C and 200 °C. Ethanol mediates the depolymerisation and formation of ethyl levulinate from the carbohydrates in the substrates. Ethanol itself is converted to the corresponding ether in a parallel acid-catalysed condensation reaction. The complementary synergistic thermal and combustion propert
