Modulation of lipid digestion by food-derived compounds has attracted increasing interest. This study explored the effects of four common carotenoids (lutein, zeaxanthin, lycopene, and β-carotene) on lipid digestion using simplified in vitro model systems from biochemical and interfacial perspectives. Lutein showed noncompetitive inhibition of pancreatic lipase, whereas zeaxanthin, lycopene, and β-carotene exhibited mixed-type inhibition. Spectroscopy and molecular simulation showed that these carotenoids can directly interact with pancreatic lipase, altering its structure and reducing activit

BACKGROUND: The relationship between serum levels of carotenoids and endometriosis remains largely unknown. The aim of this study is to assess the association between serum levels of major carotenoids (α-carotene, β-carotene, β-cryptoxanthin, lutein/zeaxanthin, and trans-lycopene) and the risk of endometriosis in US women.
METHODS: The data were obtained from the 2001-2006 National Health and Nutrition Examination Surveys (NHANES), which included a total of 3,636 women aged 20 to 54. Serum levels of α-carotene, β-carotene, β-cryptoxanthin, lutein/zeaxanthin, and trans-lycopene were measured u

Concentrations of retinol, α-tocopherol, and major carotenoids in dairy products are often determined simultaneously by liquid chromatography. These compounds have different polarity and solubility; thus, extracting them simultaneously can be difficult and inefficient. In milks with low carotenoid concentrations, the xanthophylls lutein and zeaxanthin may not be completely resolved using common extraction techniques. A simplified method was developed to optimize extraction efficiency and the limit of detection and limit of quantification (LoQ) of lutein and zeaxanthin in bovine milk without de

A polymorphism in the promoter of the β,β-carotene 15,15'-monooxygenase 1 (BCMO1) gene recently was identified in an experimental cross between 2 chicken lines divergently selected on growth rate and found to be associated with variations in the yellow color of the breast meat. In this study, the effects of the polymorphism on several aspects of carotenoid metabolism were evaluated in chickens sharing the same genetic background except for their genotype at the BCMO1 locus. We confirmed that BCMO1 mRNA abundance varied (P < 0.001) between the 2 homozygous genotypes (GG << AA) and in the pector

Hyperglycemia-linked oxidative stress and/or consequent endoplasmic reticulum (ER) stress are the causative factors of pathogenesis of diabetic retinopathy. Dietary bioactive components which mitigate oxidative stress may serve as potential chemopreventive agents to prevent or slow down the disease progression. Wolfberry is a traditional Asian fruit consumed for years to prevent aging eye diseases in Asian countries. Here we report that dietary wolfberry ameliorated mouse retinal abnormality at the early stage of type 2 diabetes in db/db mice. Male mice at six weeks of age were fed the control

An efficient combination strategy based on high-speed shear dispersing emulsifier technique and high-performance countercurrent chromatography was developed for on-line extraction and isolation of carotenoids from the fruits of Lycium barbarum. In this work, the high-speed shear dispersing emulsifier technique has been employed to extract crude extracts using the upper phase of high-performance countercurrent chromatography solvent system composed of n-hexane-dichloromethane-acetonitrile (10:4:6.5, v/v) as the extraction solvent. At the separation stage, the high-performance counter-current ch

Xanthophylls constitute a major part of carotenoids in nature. They are an oxidized version of carotenoid. Xanthophyll has widely drawn scientists' attentions in terms of its functionality, bioavailability and diversity. An assortment of xanthophyll varieties includes lutein, zeaxanthin, β-cryptoxanthin, capsanthin, astaxanthin, and fucoxanthin. Chemically, lutein and zeaxanthin are dipolar carotenoids with hydroxyl groups at both ends of their molecules that bestow hydrophilic properties to them. Hydrophilic affinity in lutein and zeaxanthin makes better bioavailability in reaction with singl

BACKGROUND: A higher dietary intake of carotenoid-rich foods and higher circulating concentrations of carotenoids have been associated with better lung function in cross-sectional studies; however, the longitudinal association between carotenoids and lung function has shown conflicting results.
OBJECTIVE: We examined the longitudinal association between serum carotenoids (β-cryptoxanthin, α-carotene, β-carotene, lutein/zeaxanthin, and lycopene) and the evolution of lung function.
DESIGN: We evaluated our hypothesis in the Coronary Artery Risk Development in Young Adults (CARDIA) prospective

BACKGROUND: Age-related macular degeneration (AMD) is one of the most common causes of irreversible impairment of central vision in the elderly population. Although anti-VEGF therapy remains the standard treatment for neovascular AMD, nutritional factors may influence disease progression and retinal health. The aim of this prospective controlled study was to evaluate the effects of an individualized dietary intervention with nutraceutical characteristics on serum concentrations of lutein, zeaxanthin, lycopene, vitamin A, and vitamin E in patients with neovascular AMD receiving anti-VEGF therap

This study examined the impact of co-culturing Chlorella saccharophila (UTEX247) with Exiguobacterium sp. strain AMK1 on carotenoid production under nitrate-depleted conditions and 3% CO₂ supplementation. The co-culture significantly enhanced the productivity of lutein (238.31 µg.L⁻¹d⁻¹), zeaxanthin (220.72 µg.L⁻¹d⁻¹), violaxanthin (185.42 µg.L⁻¹d⁻¹), and antheraxanthin (84.07 µg.L⁻¹d⁻¹). Compared to nitrate-repleted mono-cultures, these carotenoids increased by 3.54-fold, 4.81-fold, 12.28-fold, and 9.34-fold, respectively. The violaxanthin cycle, activated by CO₂ supplementation, resulted in

Our goal was to determine whether anthocyanin-producing species (red) use different photoprotective strategies to cope with excess light during fall senescence compared with non-anthocyanin-producing species (yellow). In a previous study, we found that a yellow species retained the photoprotective PsbS protein in late autumn, while a red species did not. Specifically, we tested the hypothesis that red species make less use of zeaxanthin and PsbS-mediated thermal dissipation, as they rely on anthocyanins for photoprotection. We monitored four red (Acer ginnala, Rhus typhnia, Parenthocissus quin

Suggestions that carotenoid-containing foods are beneficial in maintaining health have led to several studies of circulating carotenoid concentrations of adults. Because few data are available for children, we report serum carotenoid concentrations of 493 children in Belize. Carotenoid concentrations were determined as part of a survey of vitamin A status of children, most between 65 and 89 mo of age. Reproducibility was tested by collecting a second blood sample 2 wk after the first collection from a subset of children (n = 23) who consumed their habitual diet with no treatment during the int

The separation of carotenoids and retinoids on a beta-cyclodextrin-bonded stationary phase with conventional mobile phases is reported. Compounds studied include beta-carotene (all-trans), 15,15'-cis-beta-carotene, 7,8,7',8'-dihydro-beta-carotene, alpha-carotene, lycopene, lutein, zeaxanthin, retinal, retinol, retinol palmitate and retinol acetate. The best resolution of carotenes was obtained with low concentrations (less than or equal to 1%) of polar solvents (e.g., 2-propanol or ethyl acetate) in hexane or cyclohexane. Xanthophylls required much higher concentrations of polar solvents. The

Age-related macular degeneration (AMD) is the leading cause of blindness in people over the age of 65 particularly in those who are smokers, obese, White race, genetically predisposed and environmentally exposed. The root cause is thought to be photochemical damage causing oxidative stress to the macula coupled with low grade inflammation over many years which also contributes to the progression of the disease. The hallmark studies Age-Related Eye Disease Study (AREDS) and AREDS2 found a formulation consisting of 500 mg vitamin C, 400 IU vitamin E, 25 mg zinc, 2 mg copper, 10 mg lutein and 2 m

INTRODUCTION: All-trans-crocin I (t-crocin I) is the predominant and most extensively studied crocin congener, with antidepressant effects comparable to some first-line antidepressants. However, t-crocin I is still obtained mainly by plant extraction, leading to a high retail price and an unsustainable supply. Despite substantial efforts in heterologous production, most studies generate mixtures of multiple crocin congeners and their cis isomers. Approaches for selective t-crocin I production remain scarce.
OBJECTIVES: This study aims to establish and optimize an in vitro multi-enzyme cascade

There is increasing evidence that the macular pigment carotenoids, lutein and zeaxanthin, may play an important role in the prevention of age-related macular degeneration, cataract, and other blinding disorders. Although it is well known that the retina and lens are enriched in these carotenoids, relatively little is known about carotenoid levels in the uveal tract and in other ocular tissues. Also, the oxidative metabolism and physiological functions of the ocular carotenoids are not fully understood. Thus, we have set out to identify and quantify the complete spectrum of dietary carotenoids
