[Function of endogenous opioid peptides and sex steroids at the level of the central nervous system].

Gonadal steroids and beta-endorphin (beta-EP) (and probably other - EOP endogenous opioid peptides) play a role of the pivotal hormones involved in integration of several neurophysiological mechanisms. The reproductive system could be disturbed at hypothalamic level by interference of beta-EP and GnRH secretion and/or at pituitary level with response of gonadotropes to GnRH. Gonadal steroids, through a feedback mechanism, may exert similar effect on hypothalamus and/or pituitary. The action of EOP on the hypothalamo-pituitary-gonadal axis may be influenced by physiological and pathological cha

Reversal of mitochondrial damage in a rat model of Parkinson's disease by a neurotrophic peptide (MPF analogue).

We have previously shown that a metabolically stable analogue of MPF, the C-terminal tetrapeptide of human beta-endorphin of structure Lys-Lys-Gly-Glu, reduces the turning behaviour of rats with unilateral lesions of their nigro-striatal pathways. Transmission electron microscopy (TEM) has now revealed that this effect is related to reversal of the mitochondrial damage to substantia nigra (SN) neurones induced by the lesion. The results are consistent with the concept that an inherited defect in components of the mitochondrial enzyme system is the initial step in the genesis of Parkinson's dis

Negative allesthesia and decreased endogenous opiate system activity in anorexia nervosa.

The combined effects of an intragastric load of glucose compared to water and of naltrexone compared to placebo were tested on preference for sucrose in six anorectic patients. While in normal subjects, glucose-induced negative allesthesia is known to disappear upon loss of weight, it persisted in anorexia nervosa (AN) despite a major weight loss; furthermore, in contrast with its effects in normoponderal subjects, naltrexone at the dose of 25 mg did not decrease the preference for sucrose nor did it enhance glucose-induced allesthesia. Basal plasma beta endorphin level determined by radioimmu

Ectopic ACTH syndrome due to Grawitz tumor.

An extremely unusual case of adrenocorticotropin (ACTH)-producing Grawitz tumor is reported in a 56-year-old female. The clinical feature of the patient was compatible with Cushing's syndrome. The plasma levels of ACTH and cortisol were markedly elevated. At autopsy, a left renal tumor was demonstrated and its histopathological diagnosis was renal cell carcinoma (Grawitz tumor). The adrenal gland was bilaterally enlarged with diffuse hyperplasia of the fasciculate zone. The adenohypophyseal cells were atrophic and showed Crooke's degeneration. The tumor contained extremely high levels of ACTH,

Tumor markers in patients with lung cancer.

The most examined tumor markers in lung cancer patients are CEA, hormonal peptides, and some neurogenic enzymes in small cell carcinoma. Calcitonin, ACTH, ADH, CEA, neurophysin, oxytocin, beta-endorphin, neuron-specific enolase, and CK BB are elevated in serum specimens in 25-75% of cases of small cell carcinoma. The level of these markers is related to the stage of the disease in groups of patients; elevated pretreatment levels decrease with tumor regression. Marker levels are not valid in defining the tumor load and the presence of disease in the individual patient. It has not yet been docum

Novel synaptic responses mediated by dopamine and gamma-aminobutyric acid in neuroendocrine cells of the intermediate pituitary.

Neuroendocrine cells of the pituitary pars intermedia contain pro-opiomelanocortin and secrete mainly alpha-melanocyte-stimulating hormone and, in lesser amounts, corticotropin and endorphin. We have recorded intracellularly from rat pars intermedia cells in vitro and analysed the synaptic inputs to these cells. Electrical stimulation of the pituitary stalk evoked a biphasic postsynaptic response in pars intermedia cells. An initial inhibitory postsynaptic potential was most likely mediated by gamma-aminobutyric acid (GABA) because there was a conductance increase, and it was blocked by bicucu

Effect of specific opioid-receptor antagonist naloxone on rats with hereditary hypothalamic diabetes insipidus (Brattleboro strain) during acute hemorrhagic shock.

The development of an acute hemorrhagic shock in rats with hereditary diabetes insipidus (DI), lacking vasopressin, is very dramatic, compared to rats of the parent strain Long Evans (LE). After removal of 50% of the circulating blood for LE, and 30% for DI, the mortality for both LE and DI groups was 50%, the shock index being 0.049 and 0.028, respectively. Infusion of either vasopressin (107 mU/100 g) or naloxone (0.2 mg/100 g) in DI rats, prevented the progression of hemorrhagic shock into irreversible stage, and augmented survival up to 66% and 57%, respectively. Central and peripheral distribution of corticotropin-releasing factor.

Corticotropin-releasing factor (CRF) is widely distributed in the central nervous system and is associated with nuclei and pathways that control the release of adrenocorticotropin and beta-endorphin from the pituitary, and others that mediate central and peripheral autonomic responses to stress. Major concentrations of CRF immunoreactive neurons have been described in the hypothalamus, parts of the limbic system, several nuclei of the basal forebrain and brain stem, and the cerebral cortex. Recent data on the distribution of CRF immunoreactive perikarya and fibers in the preoptic-septal area,

Pituitary surgery: current status, including transsphenoidal surgery.

Advances in CT scanning and digital subtraction angiography have improved the accuracy of preoperative diagnosis of pituitary disease. Modern radioimmunoassay techniques are able to evaluate "subdivisions" and "relatives" of the classic pituitary hormones, including monomeric and oligomeric growth hormones, somatomedin C, beta-lipotropin, and beta-endorphin, as well as the alpha subunit of follicle-stimulating hormone. Pituitary pathologic states can now be identified immunologically in patients in whom results of radiologic studies are normal. Bromocriptine, a centrally active dopamine antago

Opiate receptor blockade in man reduces 2-deoxy-D-glucose-induced food intake but not hunger, thirst, and hypothermia.

Opioid peptides may act as neuromodulators in the central nervous system to conserve energy stores and water in mammals. To examine this hypothesis in man, the effect of opiate receptor blockade with naloxone on the hunger, thirst, and hypothermic response to 2-deoxy-D-glucose-induced glucoprivic stress was assessed. Opiate receptor blockade decreased stress-induced food intake but did not reduce marked increases in hunger produced by glucoprivation. Naloxone infusions did not change the hypercortisolemic, polydipsic, hypothermic, and thermogenic response to 2-deoxy-D-glucose. While these resu

Biochemistry and the schizophrenia. Old concepts and new hypothesis.

More than a decade of scientific inquiry into the biochemistry of schizophrenia has been organized by the dopamine hypothesis. The evidence … d balance between several neurotransmitters may underlie schizophrenia. The recent discovery of the endorphins has led to speculations about the possible role of these substances in schizophrenia. Both an excess and a deficiency of endorphin activity have been implicated in schizophrenia, and speculative evidence has been used to support both hypotheses. The …

Effects of opiate-like peptides, morphine, and naloxone in the photosensitive baboon, Papio papio.

The effects of intracerebroventricular (i.c.v.) or systemic injections of Met- or Leu-enkephalin, beta-endorphin, FK 33.824 (D-Ala2, MePhe4, Met(O5)-ol-enkephalin) and of morphine and naloxone have been studied in baboons, Papio papio, which spontaneously show photically induced epileptic responses. Animals were chronically implanted with epidural or deep recording electrodes and a cannula in one lateral ventricle, and tested whilst seated in a primate chair. In some animals the natural syndrome was enhanced by the prior administration of DL-allylglycine, 100--200 mg/kg, i.v. Met- or Leu-enkep

Sex differences in pain and hypothalamic-pituitary-adrenocortical responses to opioid blockade.

OBJECTIVE: Sex differences in pain sensitivity and stress reactivity have been well documented. Little is known about the role of the endogenous opioid system in these differences. This study was conducted to compare adrenocortical, pain sensitivity, and blood pressure responses to opioid blockade using naltrexone in men and women. METHODS: Twenty-six participants completed 2 sessions during which placebo or 50 mg of naltrexone was administered, using a double-blind, counterbalanced design. Thermal pain threshold and heat tolerance were assessed. Participants also rated pain during a 90-secon

Pharmacology of enkephalinase inhibitors: animal and human studies.

We have shown that a number of compounds which inhibit the degradation of met-enkephalin can produce naloxone-reversible analgesia in mice. These compounds also potentiate the analgesia produced by acupuncture, foot shock, and transcutaneous nerve stimulation in animals and humans. The potency of their effectiveness as analgesics or potentiators parallels their potency as inhibitors of mouse brain enkephalinase. D-Phenylalanine (DPA), one of these enkephalinase inhibitors, has been used successfully for the management of chronic intractable pain in humans and to potentiate the treatment of man

Deranged modulatory midbrain opioid and gonadotrophin functions: relevance to Tourette's syndrome.

In the following communication we discuss evidence suggesting that an abnormal opioid-gonadotrophin interaction at the midbrain may be essential to the clinical expression of vocal tics in Tourette's Syndrome (TS). Data derived from animal studies and the striking similarity between the symptomatology of TS and encephalitis lethargica (EL) add further support to our hypothesis implicating an abnormal opioid-gonadotrophin interaction in the pathophysiology of TS.

The effects of prolonged opioidergic blockade on LH pulsatile secretion during the menstrual cycle.

Although considerable evidence points towards a pivotal role for the endogenous opioid peptides (EOP) in the neuroendocrine regulation of GnRH-LH secretion, the effects of prolonged opioidergic blockade on LH pulsatile activity during the menstrual cycle have not been thoroughly investigated. Accordingly, 10 women in the early follicular phase (EFP, days 3 and 4), 10 women in the late follicular phase (LFP, days 9 to 13) and 7 women in the midluteal phase (MLP, days 6 to 8 after LH surge) were studied on two consecutive days before and during opioidergic blockade imposed by an opiate receptor
