Peripheral exosomal miRNAs as candidate molecular indicators associated with acupuncture at ST36 in an LPS-induced neuroinflammation model.

This study aimed to identify circulating exosomal microRNA (miRNA) candidates showing a control-like expression pattern following ST36 acupuncture treatment in an LPS-induced neuroinflammation model, using empirical Bayes reanalysis of previously published microarray data. Two independent Affymetrix GeneChip miRNA 4.0 microarray datasets from C57BL/6 male mice were reanalysed using the limma empirical Bayes framework with Benjamini-Hochberg false discovery rate (FDR) correction. After detection-call filtering, 286 commonly expressed probes were identified. Control-like pattern candidates were

xBind: an integrated webserver for large language model-enabled cross-molecular protein binding site prediction.

xBind is an interactive, freely accessible, and fully configurable webserver for large language model (LLM)-enabled cross-molecular protein binding-site prediction. xBind leverages LLM embeddings from the ESM-2 model together with sequence- and structure-derived features to predict protein-protein, protein-DNA, and protein-RNA binding sites using symmetry-aware deep graph neural networks. The input to xBind is either a single-chain protein sequence in FASTA format or a monomer protein structure in PDB or mmCIF format and it outputs predicted residue-level binding sites of the input protein wit

A novel composite model of PTSD induced by social bullying: validation via multidimensional behavioral and molecular biomarkers.

Post-traumatic stress disorder (PTSD) is a debilitating psychiatric condition characterized by persistent fear memory, hyperarousal, and social impairment. Mounting evidence highlights the critical role of social stress, such as bullying, in triggering PTSD. However, progress in elucidating the underlying mechanisms and developing effective treatments has been hindered by the lack of animal models that accurately recapitulate the multidimensional etiology of social trauma-related PTSD. To address this gap, we developed a novel triple-composite social bullying (SB) model in mice, which integrat

Production of electrospun cellulose fibres via water-assisted rapid deacetylation: Experimental and molecular modelling insights.

Cellulose acetate (CA) enables the production of cellulose nanofibers via electrospinning, overcoming the solubility limits of native cellulose. However, rapid regeneration of CA into cellulose remains a bottleneck for scalable manufacturing. We evaluated three regeneration methods: immersion, vapour exposure, and spray, targeting near-instant deacetylation of electrospun CA fibres. Immersion was the most effective: 0.5 N KOH in ethanol with 5% (v/v) water achieved complete deacetylation in 30 s. Water was essential, promoting ion mobility and hydrolysis while preserving fibre integrity. FTIR

A stratified urine-based molecular diagnostic and prognostic model for non-muscle-invasive bladder cancer management.

BACKGROUND: Non-muscle-invasive bladder cancer (NMIBC) is characterized by a high recurrence rate requiring lifelong cystoscopic surveillance. Existing urine-based molecular assays mainly rely on mutations or methylation, which fail to capture large-scale genomic instability. Copy number variation (CNV) profiling offers complementary information on tumor evolution and aggressiveness, but its application in urinary diagnosis remains limited. We aimed to integrate CNV and DNA methylation signals from urinary DNA to establish a noninvasive and biologically informed stratified diagnostic model for

Hydrophobic Natural Deep Eutectic Solvents for Sustainable CO2 Capture: Molecular Insights From Multiscale Modeling.

This research delves into the physicochemical properties and CO2 capture ability of a hydrophobic natural deep eutectic solvent (HNADES) consisting of piperitone (PIP) and linoleic acid in a 1:1 molar ratio. Through a multiscale computational approach involving COSMO-RS calculations, quantum chemistry methods, and molecular dynamics simulations, we demonstrate that the considered system, via the development of strong hydrogen bonding interactions, both homo- and hetero-associations, and in parallel with nonpolar domains through van der Waals interactions, creates a suitable situation for CO2 c

Emergence and molecular characterization of bovine alphaherpesvirus 1 subtype 1.1 in China, with in vivo characterization in a rabbit model.

Bovine alphaherpesvirus 1 (BoAHV-1; formerly bovine herpesvirus 1, BoHV-1) remains endemic in China, whereas available surveillance and genomic data have largely suggested predominance of genotype 1.2. In this study, molecular subtyping using a 451-nt fragment from the UL44 carboxyl terminal region identified 15 genotype 1.1 viruses among 60 recent BoAHV-1 positive clinical specimens. This proportion (25%) contrasts with previous Chinese surveillance studies that have predominantly reported genotype 1.2, particularly subtype 1.2b, suggesting that genotype 1.1 may have been underrecognized in e

Molecular Characterization of Calu-3 Cells from Submerged to Air-Liquid Interface to Model Lung Infections.

The air-liquid interface (ALI) model using Calu-3 cells has been used to model lung diseases. In ALI, Calu-3 polarizes and changes to a mucus-producing cell. Polarized Calu-3 similarity with primary cells has been proven; however, no studies have been focusing on the pathways differentially expressed in ALI. Here, we profiled the proteome and transcriptome of Calu-3 from submerged (nonpolarized) to ALI (polarized) conditions, and in the omics data, we observed an increase in cell replication in the nonpolarized condition while polarized cells presented higher activation of cellular energy prod

Spatial multi-omics reveals region-specific molecular signatures in a 6-OHDA model of Parkinson's disease.

Parkinson's disease (PD) is characterized by complex molecular and circuit-level alterations that extend beyond dopaminergic neurodegeneration, yet the spatial integration of metabolic and proteomic changes remains insufficiently explored. Here, we combined time-of-flight secondary ion mass spectrometry-based metabolite imaging with laser cell sorting proteomics to interrogate molecular alterations in the substantia nigra and striatum of the 6-hydroxydopamine toxin model of PD. Our analyses revealed distinct region-specific metabolic and proteomic signatures within primary lesion sites, and ad

A triazole-derived compound mitigates behavioral, biochemical, and molecular alterations in a chronic stress-induced depression model.

The increase in psychiatric disorders such as depression, largely associated with chronic stress exposure, highlights the need for safer and more effective therapeutic strategies. Compounds containing the triazole ring, such as 1-(2-(4-(4-ethylphenyl)-1H-1,2,3-triazol-1-yl)phenyl)ethan-1-one (ETAP), have gained attention due to their antidepressant potential. Previous studies demonstrated antidepressant-like effects of ETAP through different pathways. Based on these findings, the present study evaluated its protective effect in C57BL/6 male mice subjected to the chronic unpredictable mild stre

Molecular Mechanisms of Pulmonary Fibrosis: The Interaction of Epithelial-mesenchymal Transition and AMPK Pathways in a Bleomycin-induced Model.

Idiopathic pulmonary fibrosis (IPF) is a progressive lung disease marked by excessive extracellular matrix (ECM) deposition, largely mediated by activated fibroblasts. Epithelial-mesenchymal transition (EMT), regulated by transcription factors such as TGF-β, Twist1, and Snail, is a critical mechanism in fibrosis progression. AMP-activated protein kinase (AMPK) has been implicated in modulating fibrotic pathways, but its role in EMT remains unclear. This study aimed to explore the interaction between EMT and AMPK signaling in pulmonary fibrosis. A bleomycin-induced pulmonary fibrosis mouse mode

Neuroprotective Effects of a Benzofuran-Containing Selenium in a Mouse Alzheimer's Disease Model: Molecular, Biochemical, and Behavioral Analyses.

Alzheimer's disease (AD) is a neurodegenerative disorder mainly characterized by progressive cognitive decline, for which effective treatments remain limited, and selenium is known for its neuroprotective actions. Thus, this study evaluated the neuroprotective effects of the compound 2-(((3-trifluoromethyl)phenyl(selenyl)methyl)-2,3-dihydrobenzofuran (TFSeB) in a streptozotocin (STZ)-induced AD model in male Swiss mice. The animals received intracerebroventricular injections of STZ (3 mg/kg, a neurotoxic agent) to induce cognitive deficits, followed by treatment with TFSeB (1 and 5 mg/kg, intr

Multidimensional Evaluation of the Streptozotocin-Induced Alzheimer's Model: Integrating Molecular, Histopathological, and Behavioral Correlates Over Time.

Alzheimer's disease (AD) is characterized by cognitive deterioration, oxidative stress and neuroinflammation. Reliable preclinical models for sporadic AD are lacking. The intracerebroventricular (ICV) streptozotocin (STZ) model simulates pathology of sporadic AD; its timeline remains ambiguous. We assessed multiple endpoints in male Wistar rats 1 and 3 months post-ICV STZ (3 mg/kg; n = 72; control, sham, STZ). Spatial and associative memory was tested via the Morris Water Maze and Passive Avoidance Task. Hippocampal superoxide dismutase (SOD), tumor necrosis factor-α (TNF-α), and brain-derived

1,2,4-triazole incorporated into polyheterocyclic scaffolds as anti-glioblastoma agents: Biological evaluation and molecular modeling studies.

Glioblastoma (GBM) remains the most lethal brain tumor with limited treatment options and poor prognosis. The design of new anti-GBM drug candidates is of utmost need. Here we assessed the activity of 1,2,4-triazolo[3,4-a]phthalazine and 1,2,4-triazolo[4,3-b]pyridazine derivatives in GBM cell lines, patient-derived primary GBM cells as well as normal human astrocytes (NHA). The tested compounds presented considerable cytotoxicity against GBM cells in MTT assay, and two of them were much less harmful to NHA. They also significantly decreased the ability of GBM cells to form colonies. The initia

Dietary and Pharmacological Modulation of Aging-Related Metabolic Pathways: Molecular Insights, Clinical Evidence, and a Translational Model.

Advances in geroscience suggest that aging is modulated by molecular pathways that are amenable to dietary and pharmacological intervention. We conducted an integrative critical review of caloric restriction (CR), intermittent fasting (IF), and caloric restriction mimetics (CR-mimetics) to compare shared mechanisms, clinical evidence, limitations, and translational potential. Across modalities, CR and IF consistently activate AMP-activated protein kinase and sirtuins, inhibit mTOR (mechanistic target of rapamycin) signaling, and enhance autophagy, aligning with improvements in insulin sensitiv
