Association of itraconazole and potassium iodide in the treatment of feline sporotrichosis: a prospective study.

Feline sporotrichosis is an endemic disease in Rio de Janeiro, Brazil, where zoonotic transmission of Sporothrix spp. has been reported since 1998. Itraconazole (ITZ) remains the first choice for treating this disease in cats. However, there have been reports of therapeutic failure and a long-term endeavor. Potassium iodide (KI), considered in the past as a drug with variable effectiveness in cats with sporotrichosis, arises as an important option in the treatment of cats from the endemic area of Rio de Janeiro. In order to evaluate the effectiveness of the association of ITZ and KI in naive c

Sporotrichosis outbreak and mouldy
hay in NSW.

BACKGROUND: Sporotrichosis is a subacute/chronic fungal infection caused by Sporothrix schenckii. The fungus thrives on organic matter including hay, wood, moss, soil and plants. Zoonotic transmission has also been reported. Lesions present as superficial skin nodules that become purulent and ulcerate. Lymphocutaneous spread is common. Haematogenous spread is uncommon and case fatality is rare. OBJECTIVE: The aim of this article is to describe an outbreak of sporotrichosis in New South Wales. DISCUSSION: A cluster of six cases was identified in the NSW mid-north coast area in the first half

Endemic of zoonotic sporotrichosis: profile of cases in children.

BACKGROUND: Sporotrichosis in childhood is rare in most countries. Isolated cases and small outbreaks related to recreational activities or without identification of the transmission mechanism have been reported. METHODS: Series of case reports. The isolation of Sporothrix schenckii from exudates or fragments of lesions obtained from the patients was used as the criterion of inclusion in the study. RESULTS: A total of 81 cases of sporotrichosis in children younger than 15 years of age were diagnosed at the Evandro Chagas Research Institute, Fiocruz, Brazil, between 1998 and 2004. These cases

Positive Montenegro skin test among patients with sporotrichosis in Rio De Janeiro.

We studied 52 patients with sporotrichosis confirmed by isolation of Sporothrix schenckii and reactivity to the Montenegro skin test (MST) during an ongoing outbreak of this mycosis in Rio de Janeiro. The objective was to emphasize the importance of parasitological confirmation and the possibility of incorrect diagnosis based on the lesion's appearance, epidemiological information, and immunological tests. The antigen used for the MST was conserved in either thimerosal 1:10,000 (group 1) or 0.4% phenol (group 2). Nineteen patients (39%) in group 1 and seven (12%) in group 2 presented an indura

[Importance of the sporotrichosis asteroid body for the rapid diagnosis of sporotrichosis].

Eighty patients with cutaneous sporotrichosis were studied between 1985 and 1996. The investigation of asteroid bodies (AB) was done by direct microscopic slides examination of pus obtained from the lesions. Patients were divided into two groups: 32 consulting before 1989, and 48 consulting after 1990. In the first group, material was obtained as usual by simple digital pressure of the most productive lesion, and then wet preparation microscopic examination was performed. Fourteen patients with AB were found in this group (43.75%). In the second group the initial pus was discarded and new samp

PURPOSE: To describe Sporothrix schenckii conjunctivitis in 2 owners of cats with sporotrichosis. METHODS: Small case series and literature review. RESULTS: Two women had been caring for their pet cats with sporotrichosis for 2 months but did not recall any traumatic injury such as scratches or bites. Each presented a conjunctival granulomatous lesion measuring 4 to 5 mm accompanied by local hyperemia, secretion, and edema, in addition to painful facial subcutaneous nodes and regional lymph node enlargement. Pyogenic material was collected from the conjunctival sac and from cutaneous lesions

Activity of orally, topically, and parenterally administered itraconazole in the treatment of superficial and deep mycoses: animal models.

The activity of itraconazole in vitro was evaluated for 2,094 strains of 132 fungal species, one achloric alga, nine actinomycetes, and six bacterial species. Itraconazole was active against dermatophytes (271 strains), Candida species (1,303), Cryptococcus neoformans (27), Torulopsis species (170), Pityrosporum species (40), Aspergillus species (87), Sporothrix schenckii (12), dimorphic fungi, Dematiaceae, and various other organisms. This activity depended largely on the test conditions and the medium used. Ittraconazole was as active as ketoconazole in the treatment of dermatophytoses and o

Skin test and blastogenic responses to Sporotrichun schenckii.

In vivo skin testing and in vitro lymphocyte blastogenesis were evaluated in a young adult population as methods for detecting cellular immunity to Sporotrichum schenckii. Similar procedures for Candida albicans and Coccidioides immitis were also investigated. 5 of 143 subjects had positive skin tests and 14 had positive blastogenic responses to S. schenckii. These 14 subjects also exhibited unusually high responses to C. albicans in vitro and 11 of the 14 were female. Data demonstrated a correlation coefficient of 0.89 when comparing the blastogenic assays for S. schenckii and C. albicans, su

Experimental therapy of cladosporiosis and sporotrichosis with 5-fluorocytosine.

Cladosporium trichoides and Sporothrix schenckii are fungi known to be pathogenic for man. No effective chemotherapy is available for cladosporiosis, and systemic sporotrichosis can be very resistant to antifungal chemotherapy. Experimental infections of mice with these fungi resembled their respective infections in man and provided a model for evaluating a new antifungal agent, 5-fluorocytosine (5-FC). Our results with four isolates of C. trichoides demonstrated a statistically significant dose-related therapeutic effect with 5-FC. Mortality was significantly reduced in all treatment groups,

Comparative evaluation of five serological methods for the diagnosis of sporotrichosis.

The diagnosis of sporotrichosis can be time consuming. Serological procedures could facilitate the rapid and accurate diagnosis of this disease. A slide latex agglutination (SLA) test for sporotrichosis was developed and compared with the tube agglutination (TA), complement fixation (CF), and immunodiffusion (ID) tests in the serological study of 80 proven human cases of sporotrichosis representing the cutaneous, subcutaneous, and extracutaneous forms of the disease. In addition, the indirect fluorescent antibody (IFA) technique was applied to 61 case sera. In the SLA test, latex particles sen

Seasonal trend analysis of sporotrichosis with zoonotic potential.

Sporotrichosis is a zoonotic subcutaneous mycosis caused by fungi of the genus Sporothrix. It is considered a neglected disease, already reported in almost all Brazilian states in both animals and humans, highlighting the urgent need to understand the disease's behavior to guide public control and surveillance policies. Therefore, the objective of this study was to elucidate the hypothesis of seasonality in sporotrichosis based on the cases recorded in the municipality of Guarulhos, where notification is mandatory. For this purpose, animal sporotrichosis cases from 2016 to 2023 were provided b

Rapid diagnosis of sporotrichosis by immunofluorescent method.

Histopathological examinations fungal cultures and direct immunofluorescent staining of the pus and skin biopsy microsections with fluorescein isothiocyanate-labelled specific antiserum were performed for 13 patients suspected to be infected with Sporotrichum schenckii. While histopathological examinations could not give useful information, the latter two gave more reliable results which could support the clinical diagnosis. The direct immunofluorescent staining test could be used as a method for rapid diagnosis of sporotrichosis.

Synthesis, characterization, DNA interaction studies, and biological evaluation of copper(II) hybrids containing azole drugs and intercalating ligands against neglected diseases.

In an attempt to treat neglected diseases such as leishmaniasis and fungal infections, three novel copper(II) hybrid have been developed: [Cu(dppz)(CTZ)(NO3)](NO3) (1), [Cu(dppz)(KTZ)(H2O)(NO3)](NO3) (2), and [Cu(dppz)(FLZ)(NO3)]2(NO3)2 (3). They were synthesized by coordinating antifungal imidazole based drugs and dipyridophenazine as ligands to copper(II) under mild conditions and in good yields. These coordination compounds were characterized by various analytical and spectroscopic techniques which confirmed the coordination of both ligands to the metal, and the monodentate (1) or bidentate

Disseminated Sporotrichosis in a Liver Transplant Patient: A Case Report.

Sporotrichosis is an infection caused by the fungus of the Sporothrix schenckii complex and can be particularly harmful in immunocompromised patients. We report the case of a 26-year-old male patient with a previous history of pulmonary infection who underwent a liver transplant for Budd-Chiari syndrome. After the procedure, he presented with persistent fever and leukocytosis. On the 13th post-operative day, he was diagnosed with thrombosis of the hepatic artery and underwent a second liver transplant 15 days after the first procedure. After the retransplant, he presented daily episodes of fev

Immunopathogenesis of Human Sporotrichosis: What We Already Know.

Sporotrichosis is a subacute/chronic mycosis caused by dimorphic fungus of the genus Sporothrix. This mycosis may affect both human and domestic animals and in the last few years, the geographic dispersion and increase of sporotrichosis worldwide has been observed. The occurrence of cases related to scratching/bites of domestic felines have increased, characterizing the disease as predominantly a zoonosis. In humans, sporotrichosis mainly involves the cutaneous tegument of infected patients, but other tissues may also present the infection. The main forms of clinical presentation are lymphocut
