The genomic architecture of local adaptation in two connected populations of three-spined stickleback.

Populations often adapt to their local environments despite the homogenizing effects of gene flow, but the genomic mechanisms enabling this process remain unclear. Theory predicts that adaptive divergence under high connectivity is favored when beneficial alleles cluster in regions of reduced recombination, a pattern that can be reinforced by structural variants (SVs). We investigated this in three-spined sticklebacks (Gasterosteus aculeatus) from the St. Lawrence Estuary, where distinct freshwater and marine ecotypes meet and interbreed along a short ecological gradient. Using long- and short

6D Building information modelling (BIM) for demolition waste management of semi-submersible wind turbines.

The rapid global expansion of offshore wind is matched by an emerging wave of decommissioning. By the 2030s, over 30 GW of Europe's capacity will reach end-of-life (EoL), rising to 40 million tonnes of material waste globally by 2050, with blades alone contributing approximately 325,000 tonnes of waste per year in Europe if not effectively recovered. This shift presents a risk of waste accumulation and an opportunity to embed circular economy (CE) practices into infrastructure renewal. To address this challenge, this study develops a BIM-based framework for dismantling a semi-submersible float

The canine gut microbiome as a translational model for human health and aging.

The domestic dog is increasingly recognized as a complementary translational model for microbiome research relevant to human health while also offering direct benefits for veterinary medicine. Dogs share environments and dietary exposures with humans and show substantial taxonomic and gene-content similarity to the human gut microbiome, although the apparent degree of similarity depends strongly on whether read recruitment, gene-catalog overlap, or functional overlap is assessed. Dogs also develop spontaneous chronic enteropathies, obesity-associated metabolic dysfunction, cancers, cognitive d

A Multi-Tissue Yak (Bos grunniens) ceRNA Atlas with Ribo-Seq-Informed lncRNA Curation and Candidate Prioritization.

The yak (Bos grunniens) thrives under chronic hypoxia and cold on the Qinghai-Tibet Plateau, yet a cross-tissue view of post-transcriptional regulation in this species remains limited. Here, we integrated multi-tissue RNA-seq and miRNA-seq data (tissues pooled from three Maiwa yaks) to construct and compare tissue-specific competing endogenous RNA (ceRNA) networks, while explicitly addressing a major source of false positives in ceRNA inference-misclassified lncRNA candidates with translational signatures. We cataloged 10,037 high-confidence lncRNAs (9360 non-redundant), 234 circRNAs, and 1030

A Cattle BodyMap of Transcriptome across 52 Tissues and 3 Developmental Stages Reveals New Genetic Insights into Beef Production Traits.

Cattle are important livestock that provide essential meat and milk resources. However, a comprehensive analysis of gene expression, alternative splicing (AS), and RNA editing across various organs and developmental stages in cattle has not been reported. This study aims to create a comprehensive transcriptomic BodyMap across various tissues and developmental stages, integrating this information into genomic predictions of beef production traits. We created a comprehensive transcriptomic BodyMap using 400 samples collected from 52 organs of newborn, young, and adult cattle, and estimated their

Leveraging genetic propensity to identify modifiable factors for the age at onset of Alzheimer's disease.

INTRODUCTION: Knowledge of modifiable factors influencing age at onset (AAO) of Alzheimer's disease (AD) remains limited. This study utilizes genetic information to uncover such factors. METHODS: Using 43 exposure genome-wide association studies (GWAS) summary statistics, we calculated corresponding polygenic scores (PGS) for 9219 AD cases and 10,345 controls from the Alzheimer's Disease Genetic Consortium (ADGC). Linear mixed model and survival analyses were performed to identify exposure-PGS associated with AAO. Top exposures were cross-evaluated using PGS from the PGS Catalog and Mendelian

Comprehensive Schistosoma mansoni Hierarchical Transcriptome Assembly Points to Novel lncRNAs Associated with Sexual Dimorphism.

Background/Objectives: Schistosomiasis is a neglected tropical disease affecting >200 million people worldwide. Praziquantel is the sole recommended drug against Schistosoma mansoni; however, it lacks activity against juvenile forms and cannot prevent reinfection. Thus, there is an urgent need to identify novel therapeutic targets. Long noncoding RNAs (lncRNAs) are known to regulate various biological processes in S. mansoni, including parasite pairing and fertility; therefore, screening for novel lncRNAs could reveal new potential targets. Methods: We compiled all publicly available RNA-seq d

Helicobacter pylori infection is causally associated with extra-gastric tumors: a Mendelian randomization study.

BACKGROUND: Helicobacter pylori (H. pylori) infection is a major pathogen associated with gastric cancer. However, the causal relationship between H. pylori infection and extra-gastric tumors has not been fully elucidated. METHODS: A two-sample Mendelian randomization (MR) study was conducted using genome-wide association study (GWAS) data on serum H. pylori IgG levels as the exposure variable and GWAS data on 268 benign and malignant tumors as the outcome variable. Single nucleotide polymorphisms (SNPs) associated with the exposure were identified from the OpenGWAS and GWAS Catalog databases

EBV-informed multi-omics target prioritization and structure-based drug repurposing reveal potential therapeutic strategies for multiple sclerosis.

Multiple sclerosis (MS) is a chronic neuroinflammatory disorder characterized by progressive neurodegeneration, demyelination, and genetic susceptibility. Epstein-Barr virus (EBV) infection has been implicated as a major environmental risk factor in MS pathogenesis. Despite the availability of disease-modifying therapies, effective targeted interventions for progressive disease remain limited. In this study, an EBV-informed integrative computational framework combining MS transcriptomic datasets, EBV-associated transcriptional signatures, GWAS susceptibility genes, and network analyses was emp

Personalized Auditory Rhythmic Cues to Optimize Gait in Older Adults and People With Parkinson Disease.

BACKGROUND AND PURPOSE: Gait dysfunction in people with Parkinson disease (PwPD) is common and debilitating. Internal cues (ie, mental singing) may provide a useful alternative to external cueing (ie, listening to music) to improve walking for PwPD. In this study, we individually tailored cues to each participant by allowing them to choose their own cue song and by selecting the cue rate that elicited the longest strides. METHODS: Fifty-six participants (28 controls and 28 PwPD) performed walking trials in a single-session, cross-sectional study. Initially, uncued trials were compared with cu

Shifts in Cutaneous Melanocytic Tumor Diagnostic Terminology: Melanocytoma, MPATH-Dx V2.0 and the WHO Skin5.

In this Special Issue of the Journal of Cutaneous Pathology in memory of Dr. Martin C. Mihm, Jr, we highlight his many contributions over more than 50 years to the catalog of specific melanocytic tumor terminology. Dr. Mihm was an active participant in the International Melanoma Pathology Study Group (IMPSG). Discussions led to proposed recommendations for changes in the terminology of melanocytic tumors and their standardized diagnostic reporting. Histopathological reports of melanocytic tumors provide critical information that guides patient counseling and therapy. Importantly the pathology

Insights from the Evolution of Coagulation: A New Perspective on Anti-Inflammatory Strategies in the ICU-Focus on the Contact Activation System.

This review reappraises the anti-inflammatory potential of the contact activation system (CAS) in intensive care through an evolutionary lens. The authors propose that coagulation factor XII (FXII) and related components evolved in terrestrial animals as a "foreign-surface sensing-immunothrombosis" module, helping to explain the minimal bleeding phenotype of FXII deficiency and the secondary loss of F12 in marine mammals. CAS shares components with the kallikrein-kinin system (KKS): alpha-coagulation factor XIIa (α-FXIIa) drives coagulation factor XI (FXI) activation to amplify coagulation, wh

Multi-ancestry polygenic risk scores for the prediction of type 2 diabetes and complications in diverse ancestries.

BACKGROUND: Polygenic risk scores (PRSs) improve type 2 diabetes (T2D) prediction beyond clinical risk factors but perform poorly in non-European populations, where T2D burden is often higher, undermining their global clinical utility. METHODS: We conducted the largest global effort to date to harmonize T2D genome-wide association study (GWAS) meta-analyses across five ancestries-European (EUR), African/African American (AFR), Admixed American (AMR), South Asian (SAS), and East Asian (EAS)-including 360,000 T2D cases and 1·8 million controls (41% non-EUR). We constructed ancestry-specific and
