In this study, we investigated the effects of medication electromagnetic waves (MEMW) derived from the anticancer drug doxorubicin on tumor growth of MDA-MB-231 breast cancer cell line in a mouse model. Building on prior research, we explored how MEMW inhibits cancer cell proliferation without the toxic side effects associated with traditional chemotherapy. Twenty female nude mice were xenografted with MDA-MB-231 cells and divided into five groups: a control group, a group treated with doxorubicin only, a group treated with doxorubicin plus MEMW, a group treated with MEMW only, and a group tre…

Combination chemotherapy is often limited by the distinct physicochemical properties and intracellular behaviors of individual drugs. This limitation leads to poorly coordinated delivery in tumor tissues. In this study, pH-responsive ionizable nanoliposomes were developed using the ionizable lipid DLin-DMA to co-deliver erlotinib and doxorubicin hydrochloride. This approach introduces a novel spatial and temporal synchronization strategy for two mechanistically distinct drugs within a single carrier. The resulting nanoliposomes formed stable nanoscale assemblies with an optimized particle size

Doxorubicin (DOX) loaded liposomes have been used and studied in the last decades due to the significant decrease in DOX induced cardiac and systemic toxicity relative to administration of free drug. Therefore, new strategies are sought to improve DOX delivery and antitumor activity, while avoiding side effects. Recently, folate-coated pH-sensitive liposomes (SpHL-Fol) have been studied as a tool to enhance cellular uptake and antitumor activity of paclitaxel and DOX in breast cancer cells expressing folate receptor (FR+). However, the elucidation of folate functionalization relevance in DOX-l

Wee1-like protein kinase (Wee1) is a validated target for cancer therapy, though no inhibitors have yet received regulatory approval. To identify new Wee1 inhibitors, we conducted a virtual screening campaign using the Eurofins-Villapharma library. Based on the initial hit Compound 1 (IC50 = 240 nM in the biochemical assay), we synthesized and tested additional compounds. Analog 20 showed a significant improvement in potency (IC50 = 33 nM). It was also the most potent in cellular assays, inhibiting the proliferation of lung carcinoma cell lines A427 (IC50 = 0.761 μM) and NCI-H23 (IC50 = 0.449

Granzyme B (GrB)-producing B cells have dual roles in tumor immunity, either killing tumor cells or suppressing antitumor responses by eliminating effector T cells. In this study, we aimed to investigate how breast cancer cells influence GrB-producing B cells from tumor-draining lymph nodes and whether B cell activation enhances their cytotoxic potential. Mononuclear cells were isolated from 14 fresh axillary lymph node samples by density gradient centrifugation using Ficoll-Hypaque. Lymphocytes were co-cultured with breast tumor cell lines (MCF-7 and MDA-231) in the presence of recombinant in

Experimental studies were conducted to compare the relative effects of γ-tocotrienol, a natural isoform within the vitamin E family of compounds, with established galectin-3 inhibitor β-lactose, on galectin-3 levels and distribution in highly metastatic human MDA-MB-231, and mouse + SA and TS/A breast cancer cell lines. In contrast to β-lactose, γ-tocotrienol displays a dual-mode binding profile with the carbohydrate recognition domain and hydrophobic pocket located on galectin-3, as determined using Schrödinger molecular modeling. However, treatment with either γ-tocotrienol or β-lactose indu

BACKGROUND: Identifying reliable biomarkers for immunotherapy response remains challenging. Solute carrier family 7 member 11 (SLC7A11), a key regulator of redox homeostasis and ferroptosis, demonstrates significant tumor-promoting potential across cancers, yet its pan-cancer associations with immunotherapy-related features and interaction with microRNAs in triple-negative breast cancer pathogenesis remain elusive.
METHODS: We systematically analyzed SLC7A11 expression across cancer types using TCGA and GTEx data, and evaluated its associations with immunotherapy-related features. Survival an

Human cathepsin B is a cysteine-dependent protease whose roles in both normal and diseased cellular states remain yet to be fully delineated. This is primarily due to overlapping substrate specificities and lack of unambiguously annotated physiological functions. In this work, a selective, cell-permeable, clickable and tagless small molecule cathepsin B probe, KDA-1, is developed and kinetically characterized. KDA-1 selectively targets active site Cys25 residue of cathepsin B for labeling and can detect active cellular cathepsin B in proteomes derived from live human MDA-MB-231 breast cancer c

Women with aggressive triple-negative breast cancer (TNBC) are at high risk of brain metastasis, which has no effective therapeutic option partially due to the poor penetration of drugs across the blood-brain barrier. Trifluoperazine (TFP) is an approved antipsychotic drug with good bioavailability in brain and had shown anticancer effect in several types of cancer. It drives us to investigate its activities to suppress TNBC, especially the brain metastasis. In this study, we chose three TNBC cell lines MDA-MB-468, MDA-MB-231, and 4T1 to assess its anticancer activities along with the possible

Newer therapeutic strategies aim to increase the sensitivity of cancer cells to apoptosis. The phytochemical, oleuropein (OLP) has shown promising anticancer properties; however, its low bioavailability and non-specific delivery limit its anticancer potential. Therefore, we developed a novel aptamer-conjugated nanocarrier delivery system for the targeted, efficient and controlled delivery of OLP to the MDA-MB-231 breast cancer cells. The oleuropein-loaded nanoparticles (OLP-NPs) were synthesized, characterized, and conjugated with a Mucin-1 specific aptamer (APT) (APT-OLP-NPs) for an efficient

Warburg's effect and elevated glucose transporter (GLUT) activity are defining features of many tumors and offer a route for selective drug delivery. We synthesized a panel of monosaccharide (mannose)-functionalized Pt(II) complexes (C1-C8) along with a non-glycosylated control (C9) and elucidated their structures by multiple analytical techniques and single-crystal X-ray diffraction analysis (C7, C9). Time-dependent 1H NMR indicated a water-assisted release of the mannose functionalize co-ligand from the Pt(II) center. Biological testing in MCF-7 (ER+), HeLa, and MDA-MB-231 (triple-negative)

Several cardiovascular diseases are associated with tumor development. But the mechanisms underlying how the hypertrophic heart promotes tumor progression remain unknown. Here, we show that extracellular vesicles from hypertrophic cardiomyocytes facilitate cancer progression. Following transverse aortic constriction to induce cardiac hypertrophy, 4T1-Luc cells are orthotopically implanted into mice. Concurrently, AC16 cells are treated with angiotensin II, and extracellular vesicles isolated from these cells are administered to BALB/c nude mice previously implanted with MDA-MB-231 cells in the…

Mitochondrial DNA (mtDNA) transcription is an emerging target in cancer therapy. While enzyme-targeted inhibitors like IMT1B block POLRMT, their organelle specificity remains unclear. Here, we report a distinct strategy using mitochondria-accumulating nonplanar perylene diimides (PDIs). We demonstrate that the fluorescent lead compound PDIC-BL selectively accumulates within mitochondria. Single-molecule stretching assays showed that PDIC-BL interacts with mtDNA-derived double-stranded DNA templates in vitro with an affinity comparable to ethidium (Kd = 32 μM). In cells, this mitochondrial accu

Lysyl Oxidases (LOXs) catalyse elastin and collagen cross-linking in the extracellular matrix and have been implicated in cancer cell progression and metastases, particularly in breast cancer. In this study, we sought small-molecule inhibitors of LOXL2 using a library of 4-thiazolidones supported by molecular docking. Five candidates were prioritised, with Les 5740 displaying the highest LOXL2 inhibitory activity (IC50 of 27 μM). Although Les 5740 showed low cytotoxicity in various cancer and normal-like cell lines, the triple-negative breast cancer (TNBC) cell line MDA-MB-231 showed increased

Breast cancer (BC) is a malignant tumor that develops in the mammary gland due to uncontrolled cell proliferation. Estrogen receptor (ER) signaling, mediated by 17β-estradiol (E2), plays a crucial role in regulating cell proliferation, differentiation, and survival. Specifically, the binding of E2 to the estrogen receptor alpha (ERα) increases cell proliferation. Conversely, selective estrogen receptor beta (ERβ) agonists inhibit cancer cell proliferation by suppressing the expression of oncogenes, making ERβ an important therapeutic target. Given the urgent need for targeted and effective the
