Bread Wheat Microspore Culture.

Microspore culture is one of the doubled-haploid-producing techniques that can speed up the breeding process and shorten the time for releasing new cultivars. Growing world population and changing climate demand to expedite the development of new resilient and high-yielding cultivars for staple crops such as bread wheat (Triticum aestivum L.). Here, we describe a comprehensive protocol for obtaining bread wheat doubled haploids through microspore culture. Key steps including donor plant growth and spike pretreatment, microspore isolation and density-gradient-based purification, microspore-ovar

A Rapid Point-of-Care Ultrasound Diagnosis and Treatment of Tamponade in a Patient With Rare and Lethal Purulent Pericarditis: A Case Report.

This article describes the prompt diagnosis and life-saving intervention facilitated by point-of-care ultrasound (POCUS) in a patient presenting with tamponade secondary to a rare case of purulent pericarditis. Within a remarkable timeframe of only 40 minutes from emergency department (ED) presentation to pericardial drainage, POCUS not only confirmed the diagnosis but also expedited critical treatment measures. The etiology of the underlying purulent pericardial effusion was traced to a Streptococcus pneumoniae infection. Ultimately, the patient had untreated X-linked agammaglobulinemia (XLA)

Photoredox Autocatalysis: Towards a Library of Generally Applicable Sulfonamide Reductive Photocatalysts.

Dichotomous thinking dominates the field of synthetic photochemistry-either a reaction needs a photocatalyst or not. Herein, we report the identification of a photoredox autocatalytic pathway - an alternative to the existing mechanistic paradigms - to access cyclic biaryl sulfonamides (BASNs). This reaction does not require exogenous catalyst as the visible light absorbing deprotonated product - with potent excited state reductive power - acts as the photocatalyst for its own synthesis. This finding implicated BASN as a novel organophotocatalyst architecture and allowed a rapid, modular, and l

Mucocutaneous Adverse Effects of Methotrexate Toxicity: A Case Series.

Methotrexate is a widely used medication in dermatology, rheumatology, and oncology. However, patient misunderstandings or attempts to expedite symptom relief can lead to overuse, resulting in severe toxicity. This case series presents five instances of methotrexate toxicity due to daily dosing and excessive use for underlying disorders. All patients exhibited fever, skin, and mucosal lesions, alongside abnormal hematological parameters. Management involved immediate cessation of methotrexate, administration of leucovorin rescue therapy, and supportive care. Three patients fully recovered with

A curious case of unsuspected intracranial metallic splinter with complications.

Penetrating injuries to the brain, due to low-velocity, non-missile objects are rare in civilian lament, but can carry significant morbidity. We describe a 20-year-old male with altered sensorium, headache and imbalance, months following what was deemed a trivial head injury from cricket. After imaging, a 2.5 cm metallic splinter lodged in the left cerebellum, which was subsequently surgically removed. The patient developed meningitis and hydrocephalus, required a shunt thereafter, but was complicated by the development of a pseudocyst, cherry-picking a CSF leak, malfunction of the original sh

Discovery of Potent Antimalarial Type II Kinase Inhibitors with Selectivity over Human Kinases.

While progress has been made in the effort to eradicate malaria, the disease remains a significant threat to global health. Acquired resistance to frontline treatments is emerging in Africa, urging a need for the development of novel antimalarial agents. Repurposing human kinase inhibitors provides a potential expedited route given the availability of a diverse array of kinase-targeting drugs that are approved or in clinical trials. Phenotypic screening of a library of type II human kinase inhibitors identified compound 1 as a lead antimalarial, which was initially developed to target human ep

Osseous Stress Injuries: Treatment Algorithms and Return to Play.

Osseous stress injuries are common in athletes. Specifically, lower extremity injuries are prevalent in running athletes and upper extremity injuries are prevalent in throwing athletes. Such injuries are suspected when there is focal bone tenderness and increased pain with the inciting activity. In elite athletes, osseous stress injuries are a relatively common culprit in lost play time. Thus rapid diagnosis and treatment is imperative to expedite return to play (RTP). The radiologist's role in these cases is not only for diagnosis, but also to grade the injury, which has implications in deter

Biomarkers for progressive multifocal leukoencephalopathy: emerging data for use of JC virus DNA copy number in clinical trials.

Progressive multifocal leukoencephalopathy is a rare but devastating demyelinating disease caused by the JC virus (JCV), for which no therapeutics are approved. To make progress towards addressing this unmet medical need, innovations in clinical trial design are needed. Quantitative JCV DNA in CSF has the potential to serve as a valuable biomarker of progressive multifocal leukoencephalopathy disease and treatment response in clinical trials to expedite therapeutic development, as do neuroimaging and other fluid biomarkers such as neurofilament light chain. Specifically, JCV DNA in CSF could b

Water-based efficient alkyne transformation towards α-acetoxy/imido-ketones via oxidative coupling reactions using an alkylamine catalyst.

A novel approach is unveiled for the expedited synthesis of valuable α-substituted ketones, utilising aliphatic amine catalysis to drive the oxidative C-O/C-N coupling reaction between alkynes and an appropriate nucleophile. This one-pot synthesis employs hypervalent iodine as both the oxidant and coupling agent. A fast, metal-free, and environmentally benign method is developed for synthesising α-acetoxyketones and α-imidoketones in an aqueous medium. To demonstrate the potential for larger-scale production, a gram-scale reaction is conducted. Moreover, the newly developed methodology has suc

Immunodetection of Pyruvate Carboxylase Expression in Human Astrocytomas, Glioblastomas, Oligodendrogliomas, and Meningiomas.

Pyruvate carboxylase (PC) is an enzyme catalyzing the carboxylation of pyruvate to oxaloacetate. The enzymatic generation of oxaloacetate, an intermediate of the Krebs cycle, could provide the cancer cells with the additional anaplerotic capacity and promote their anabolic metabolism. Recent studies revealed that several types of cancer cells express PC. The gained anaplerotic capability of cells mediated by PC correlates with their expedited growth, higher aggressiveness, and increased metastatic potential. By immunohistochemical staining and immunoblotting analysis, we investigated PC expres

Possible Donor-Derived Infection in a Pediatric Liver Transplant Patient With Granulomatous Hepatitis.

Pediatric liver transplant recipients are a high-risk group for the development of adenovirus hepatitis and other manifestations of disseminated adenoviral disease. The risk is greatest during periods of increased immunosuppression, including immediately post-transplantation and following treatment for rejection. Manifestations of adenovirus hepatitis are heterogeneous with a wide spectrum of clinical severity, ranging from mild, focal disease to fulminant liver failure. Here we report a case of liver transplantation-associated adenovirus hepatitis presenting with fever and multifocal liver le

Helicopter Emergency Medical Services Ultrasound Use in the Diagnosis of Pericardial Effusion Due to Aortic Dissection.

Ultrasound has historically been limited to in-hospital use for the diagnosis and management of various conditions. However, with the advent of smaller and more portable devices, this technology can be used outside the hospital. This report describes a patient with chest pain and hypotension for whom a point-of-care cardiac ultrasound (POCUS) was performed to diagnose cardiac tamponade during critical care transport. She was subsequently found to have an acute type A aortic dissection, and her care was expedited to the operating room. Cardiac tamponade in the setting of acute type A aortic dis

Reimagining Family Involvement in Residential Long-Term Care.

Although descriptions of family involvement in residential long-term care (RLTC) are available in the scientific literature, how family involvement is optimized in nursing homes or assisted living settings remains underexplored. During the facility lockdowns and visitor restrictions of the COVID-19 pandemic, residents experienced social deprivation that may have resulted in significant and adverse health outcomes. As with so many other critical issues in RLTC, the COVID-19 pandemic has magnified the need to determine how families can remain most effectively involved in the lives of residents.

Point-of-Care Ultrasound Diagnosis of Cesarean Scar Ectopic Pregnancy: A Case Series.

BACKGROUND: A cesarean scar pregnancy (CSP) is a clinically important form of ectopic pregnancy that carries a high risk of maternal morbidity and mortality. As the rate of cesarean sections has risen, this diagnosis is becoming an increasingly important consideration for providers caring for patients in early pregnancy. CASE REPORT: We present three cases of CSPs in which point-of-care ultrasound expedited the diagnosis and treatment in the emergency department. WHY SHOULD AN EMERGENCY PHYSICIAN BE AWARE OF THIS?: Given the risks of an undiagnosed CSP, the increasing incidence of CSP, and th

Emancipating Chlamydia: Advances in the Genetic Manipulation of a Recalcitrant Intracellular Pathogen.

Chlamydia species infect millions of individuals worldwide and are important etiological agents of sexually transmitted disease, infertility, and blinding trachoma. Historically, the genetic intractability of this intracellular pathogen has hindered the molecular dissection of virulence factors contributing to its pathogenesis. The obligate intracellular life cycle of Chlamydia and restrictions on the use of antibiotics as selectable markers have impeded the development of molecular tools to genetically manipulate these pathogens. However, recent developments in the field have resulted in sign

Toward T cell protein-protein interaction activity relevant to alopecia areata.

Development of better therapies for the T cell-mediated autoimmune disease alopecia areata (AA) could be expedited by an improved understanding of the immunologic signals underlying its pathogenesis. To approach this, our group is mounting a new technological and analytical platform, multiplex immunoprecipitation detected by flow cytometry (MIF). MIF is designed to allow analysis of collections of protein-protein interactions that participate in T cell signaling webs. Early experiments suggest that MIF can detect the increased protein-protein interaction network activity that occurs under cond
