Cerebral modeling and dynamic Bayesian networks.

The understanding and the prediction of the clinical outcomes of focal or degenerative cerebral lesions, as well as the assessment of rehabilitation procedures, necessitate knowing the cerebral substratum of cognitive or sensorimotor functions. This is achieved by activation studies, where subjects are asked to perform a specific task while data of their brain functioning are obtained through functional neuroimaging techniques. Such studies, as well as animal experiments, have shown that sensorimotor or cognitive functions are the offspring of the activity of large-scale networks of anatomical…

Neuroimaging in eating disorders and obesity: implications for research.

Medicine and psychiatry have benefited from developments in investigational techniques. Neuroimaging is one such domain that has technically progressed enormously in recent years, resulting in, for example, higher temporal and spatial resolution. Neuroimaging techniques have been widely used in a range of psychiatric disorders, providing new insights into neural brain circuits and neuroreceptor functions in vivo. These imaging techniques allow researchers to study not only the configuration of brain structures but also aspects of normal and anomalous human behavior more accurately.

Spontaneous Brain Activity Abnormalities in Patients With Temporal Lobe Epilepsy: A Meta-Analysis of 1474 Patients.

BACKGROUND: Abnormalities in resting-state functional brain activity have been detected in patients with temporal lobe epilepsy (TLE). The results of individual neuroimaging studies of TLE, however, are frequently inconsistent due to small and heterogeneous samples, analytical flexibility, and publication bias toward positive findings. PURPOSE: To investigate the most consistent regions of resting-state functional brain activity abnormality in patients with TLE through a quantitative meta-analysis of published neuroimaging data. STUDY TYPE: Meta-analysis. SUBJECTS: Exactly 1474 TLE patients

DunedinPACNI estimates the longitudinal Pace of Aging from a single brain image to track health and disease.

To understand how aging affects functional decline and increases disease risk, it is necessary to develop measures of how fast a person is aging. Using data from the Dunedin Study, we introduce an accurate and reliable measure for the rate of longitudinal aging derived from cross-sectional brain magnetic resonance imaging, that is, the Dunedin Pace of Aging Calculated from NeuroImaging (DunedinPACNI). Exporting this measure to the Alzheimer's Disease Neuroimaging Initiative, UK Biobank and BrainLat datasets revealed that faster DunedinPACNI predicted cognitive impairment, accelerated brain atr

The effects of APOEe4 allele on cerebral structure, function, and related interactions with cognition in young adults.

In the last decade, extensive research has emerged into understanding the impact of risk factors for Alzheimer's Disease (AD) on brain in pre-symptomatic stages. We investigated the neuroimaging correlates of the APOEe4 genetic risk factor for AD in young adulthood, its relationship with cognition, and potential effects of other variables on the findings. While conventional volumetric analyses revealed no consistent differences, more sophisticated analyses identified subtle structural differences between APOEe4 carriers and non-carriers. Findings from diffusion studies were limited, but functi

Brain alteration of autoimmune thyroid disease: Neuropsychiatric impact, neuroimaging insights, and neurobiological implications.

Autoimmune thyroid disease (AITD) is the most common organ-specific autoimmune disease, characterized by thyroid function disorder and autoimmune imbalance. Previous studies have demonstrated the decreased quality of life and neuropsychiatric manifestations in AITD patients, including anxiety, depression, cognitive impairment and affective disorder. These problems also plague the euthyroid AITD patients. Advanced neuroimaging techniques were well carried out and employed as an explanatory instrument for the above intriguing phenomenon. In recent years, an increasing number of neuroimaging stud

Neuroplasticity and aphasia treatments: new approaches for an old problem.

Given the profound impact of language impairment after stroke (aphasia), neuroplasticity research is garnering considerable attention as means for eventually improving aphasia treatments and how they are delivered. Functional and structural neuroimaging studies indicate that aphasia treatments can recruit both residual and new neural mechanisms to improve language function and that neuroimaging modalities may hold promise in predicting treatment outcome. In relatively small clinical trials, both non-invasive brain stimulation and behavioural manipulations targeting activation or suppression of

Neuroimaging the Effectiveness of Substance Use Disorder Treatments.

Neuroimaging techniques to measure the function and biochemistry of the human brain such as positron emission tomography (PET), proton magnetic resonance spectroscopy ((1)H MRS), and functional magnetic resonance imaging (fMRI), are powerful tools for assessing neurobiological mechanisms underlying the response to treatments in substance use disorders. Here, we review the neuroimaging literature on pharmacological and behavioral treatment in substance use disorder. We focus on neural effects of medications that reduce craving (e.g., naltrexone, bupropion hydrochloride, baclofen, methadone, var

Effect of mind on brain activity: evidence from neuroimaging studies of psychotherapy and placebo effect.

Mentalistic variables must be considered to reach a correct understanding of the neurophysiological basis of behavior in humans. Confusion regarding the relative importance of neurophysiological and mentalistic variables can lead to important misconceptions about causes and effects in the study of human behavior. In this article, we review neuroimaging studies of the effect of psychotherapy in patients suffering from diverse forms of psychopathology (obsessive-compulsive disorder, panic disorder, unipolar major depressive disorder, spider phobia). We also review neuroimaging studies of the pla

The cognitive cerebellum: a hub for motor, emotional, and executive control.

For many decades, the cerebellum was regarded almost exclusively as a structure devoted to motor regulation, responsible for balance, coordination, and the fine-tuning of movement. This view began to change in the 1990s, when studies with patients with isolated cerebellar lesions revealed cognitive and affective disturbances that could not be solely explained by cortical dysfunction. Subsequent anatomical, neuroimaging, and clinical investigations demonstrated robust reciprocal connectivity between the cerebellum and prefrontal regions, while functional imaging confirmed cerebellar activation

Tinnitus and tinnitus disorder: Genetic, neurobiological, and clinical differentiation.

Tinnitus is the conscious perception of sound in the absence of an external acoustic source. When accompanied by emotional distress, cognitive dysfunction, or autonomic arousal leading to behavioral and functional impairment, it is termed "tinnitus disorder." This perspective synthesizes genetic, epidemiological, and neuroimaging evidence supporting the distinction between tinnitus and tinnitus disorder. Genetic studies indicate that tinnitus is linked to multiple common variants of small effect size, whereas tinnitus disorder involves rarer variants exerting larger effects. Epidemiologically,

The Etiology of Auditory Hallucinations in Schizophrenia: From Multidimensional Levels.

Enormous efforts have been made to unveil the etiology of auditory hallucinations (AHs), and multiple genetic and neural factors have already been shown to have their own roles. Previous studies have shown that AHs in schizophrenia vary from those in other disorders, suggesting that they have unique features and possibly distinguishable mechanisms worthy of further investigation. In this review, we intend to offer a comprehensive summary of current findings related to AHs in schizophrenia from aspects of genetics and transcriptome, neurophysiology (neurometabolic and electroencephalogram studi

Charting Frontotemporal Dementia: From Genes to Networks.

Frontotemporal dementia (FTD) is a genetically and clinically heterogeneous syndrome that is characterized by overlapping clinical symptoms involving behavior, personality, language and/or motor functions and degeneration of the frontal and temporal lobes. The term frontotemporal lobar degeneration (FTLD) is used to describe the proteinopathies associated with clinical FTD. Emerging evidence from network-based neuroimaging studies, such as resting state functional MRI and diffusion tensor MRI studies, have implicated specific large-scale brain networks in the pathogenesis of FTD syndromes, sug

Longitudinal Multimodal Neuroimaging After Traumatic Brain Injury.

Traumatic brain injury is a major cause of long-term cognitive impairment, yet the mechanisms underlying recovery remain poorly understood. Neuroimaging methods such as diffusion magnetic resonance imaging (MRI), functional MRI (fMRI), and positron emission tomography (PET) provide insight into micro- and macro-scale changes post-traumatic brain injury (TBI), but the relationships between regional cellular and functional alterations remain unclear. In this exploratory study, we conducted a longitudinal, multimodal neuroimaging analysis quantifying TBI-related pathologies in four biomarkers, na

The anatomy of conscious vision: an fMRI study of visual hallucinations.

Despite recent advances in functional neuroimaging, the apparently simple question of how and where we see--the neurobiology of visual consciousness--continues to challenge neuroscientists. Without a method to differentiate neural processing specific to consciousness from unconscious afferent sensory signals, the issue has been difficult to resolve experimentally. Here we use functional magnetic resonance imaging (fMRI) to study patients with the Charles Bonnet syndrome, for whom visual perception and sensory input have become dissociated. We found that hallucinations of color, faces, textures…

Different functional connectivity gradients reflect aging and Alzheimer's disease.

Aging and Alzheimer's disease (AD) are accompanied by alterations to large-scale communication patterns in the brain, which can be tracked in vivo using functional connectivity. The location, direction and relevance of these changes remain widely debated, although they are rarely studied in the context of whole-cortex communication dynamics. Here, in two independent cohorts (BioFINDER-2, N = 973; Alzheimer's Disease Neuroimaging Initiative, N = 129), we show that functional connectivity changes associated with aging and AD are strongly aligned with separate fundamental axes of hierarchical bra

Propofol disrupts the functional core-matrix architecture of the thalamus in humans.

Research into the role of thalamocortical circuits in anesthesia-induced unconsciousness is difficult due to anatomical and functional complexity. Prior neuroimaging studies have examined either the thalamus as a whole or focused on specific subregions, overlooking the distinct neuronal subtypes like core and matrix cells. We conducted a study of heathy volunteers and functional magnetic resonance imaging during conscious baseline, deep sedation, and recovery. We advanced the functional gradient mapping technique to delineate the functional geometry of thalamocortical circuits, within a framew

Cerebellar functional connectivity alteration in individuals with lower limb amputation.

Limb amputation triggers a reorganization of brain structure and function. Previous research using neuroimaging techniques, such as resting state functional magnetic resonance imaging, indicates reduced functional connectivity in the sensorimotor network in amputees compared to non-amputees. Yet, data on lower limb amputees remains limited. We investigated functional connectivity differences within the sensorimotor network (S1M1) using an analysis of 10 regions of interest in a group of 26 lower limb amputees compared to non-amputees. The statistical analysis revealed a decreased connectivity
