POTEF upregulation drives hepatocellular carcinoma progression via oncogenic signaling and immune modulation.

Members of the POTE gene family have been implicated in tumorigenesis across multiple malignancies. However, the biological and clinical significance of POTEF in hepatocellular carcinoma (HCC) remains unclear. Differential expression analyses of POTE family genes were performed using transcriptomic sequencing of 12 paired HCC and adjacent tissues and validated using TCGA and GEO datasets. POTEF expression was further confirmed by qRT-PCR, western blotting, and immunohistochemistry in clinical specimens. Functional assays, including CCK-8, wound-healing, apoptosis, and xenograft experiments, we

Immunoelectroanalytical multiplexing of RNA methylation signatures to decode oncogenic point mutations in cancer cells.

Epigenomic and epitranscriptomic alterations-particularly RNA methylations such as N6-methyladenosine (m6A), N5-methylcytosine (m5C), N1-methyladenosine (m1A), and N7-methylguanosine (m7G)-are increasingly recognized as pivotal regulators in the pathobiology of colorectal cancer (CRC) and other highly prevalent disorders. Despite their growing clinical and biological relevance, rapid and robust electrochemical strategies enabling the single (m1A, m7G) or multiplexed quantification of these RNA modifications remain unavailable. Herein, we report an indirect competitive, four-analyte amperometri

Hedgehog signaling drives glial cell plasticity and oncogenic reprogramming in gastroenteropancreatic neuroendocrine neoplasms.

BACKGROUND: Disruption of the Men1 locus in epithelial and endocrine tissues fails to generate the full spectrum of gastroenteropancreatic neuroendocrine tumors (GEP-NETs), raising the possibility of a potential stromal source for these cancers. Neural crest-derived glial cells were previously implicated in neuroendocrine tumors arising in the pituitary and pancreas, yet these studies lacked a clear mechanism for these events. Here, we investigated the hypothesis that Men1-driven Hedgehog (HH) signaling redirects the glial cell fate to give rise to neuroendocrine tumors in the gastrointestinal

The Oncogenic LncRNA-PVT1 in colorectal cancer: mechanisms of ceRNA-mediated progression and clinical implications.

Colorectal cancer (CRC) remains one of the leading causes of cancer-related mortality worldwide, with both genetic and epigenetic factors contributing to its initiation and progression. Long non-coding RNAs (lncRNAs) have emerged as pivotal regulators of tumorigenesis. Among these, plasmacytoma variant translocation 1 (PVT1) has attracted increasing attention due to its oncogenic functions. PVT1 primarily acts as a competing endogenous RNA (ceRNA), sequestering tumor-suppressive microRNAs to promote cancer progression. Additionally, polymorphisms within the PVT1 locus have been associated with

Covalent Targeting and Thermostabilization of Oncogenic R280K and R273H Mutants p53 by Small Molecule RVJB59.

Breast cancer is one of the most common cancers worldwide. Approximately 30%-40% of breast cancers harbor mutations in the TP53 gene, leading to structural and functional alterations in the p53 protein. These changes result in mutant proteins that are unable to perform their canonical tumor suppressor functions and, in many cases, exhibit gain-of-function properties, making mutant p53 a highly attractive therapeutic target. Among these mutations, R280K and R273H are two clinically relevant DNA-binding mutations. In this study, we extended our investigation of the (R)-tryptophanol isoindolinone

Oncogenic role of LncRNA DLEU2 in thyroid cancer through a NAT10/USP39 regulatory axis.

Thyroid cancer incidence is rapidly increasing worldwide, with many patients’ developing radioiodine-refractory disease that leads to a poor prognosis. Long non-coding RNAs (lncRNAs) are critical regulators of tumor progression. While DLEU2 has been implicated in several cancers, its specific function in thyroid cancer remained undefined. This study aimed to investigate the role of DLEU2 in thyroid cancer progression through its interaction with NAT10 and USP39. We examined DLEU2 expression in 70 paired thyroid cancer and adjacent tissues, and in multiple cell lines using qRT-PCR, finding mark

Design, Synthesis and Biophysical Characterisation of CPP-PNA Conjugates for Targeting Oncogenic microRNA-221.

Peptide nucleic acids (PNAs) are a promising platform targeting disease-associated microRNAs due to their high binding affinity, excellent sequence specificity and resistance to enzymatic degradation. However, their therapeutic application is often limited by inefficient cellular uptake. In this study, we present the design, synthesis and physicochemical characterisation of cell-penetrating peptide (CPP)-PNA conjugates that target the oncogenic microRNA miRNA-221-5p, as well as their preliminary biological evaluation. Anti-miRNA-221-5p PNAs functionalised for conjugation with CPPs via Cu(I)-ca

Lineage identity governs oncogene dependence in mouse NSCLC models of KRAS inhibitor resistance.

Primary and acquired resistance to targeted therapy represent significant challenges to durable treatment responses in oncology. In lung adenocarcinoma, KRAS inhibitors are hampered by incomplete clinical responses and acquired resistance. Here we treated genetically engineered mouse models of Kras-driven lung adenocarcinoma with KRAS inhibitors to model these conditions. Initial treatment response was rapid but incomplete, with residual tumor burden transcriptionally resembling alveolar epithelial cells. With continued treatment, we invariably observed genetic Kras amplification as a resistan

NOTCH3 Internal Tandem Duplication Defines a Novel Oncogenic Activation Mechanism of NOTCH Signaling.

NOTCH signaling is activated in tumors through multiple mechanisms, including mutations, gene rearrangements, and gene amplification. We report a novel activation mechanism, an internal tandem duplication (ITD) near the NOTCH3 negative regulatory region (NRR), found in a myogenic mesenchymal neoplasm. This 17-amino acid residue duplication disrupts the tightly autoinhibited structure surrounding the S2 cleavage site, resulting in ligand-independent S2 cleavage and constitutive pathway activation, as demonstrated by increased expression of the NOTCH3 target gene HES1. Cells expressing NOTCH3-IT

Sweetened Beverages and Pancreatic Cancer: Insights From Recent Epidemiological Data and Oncogenic Mechanisms.

PURPOSE OF REVIEW: Hyperglycemia and altered carbohydrate metabolism may promote pancreatic cancer cell survival and proliferation. This narrative review critically evaluates epidemiological evidence on whether sugar-sweetened beverage (SSB) consumption is associated with pancreatic cancer risk and examines potential biological mechanisms involving glucose and fructose metabolism, oncogenic KRAS signaling, and metabolic reprogramming. RECENT FINDINGS: Earlier pooled analyses and a 2019 meta-analysis found no significant overall association between SSB consumption and pancreatic cancer risk. H

Oncogenic KRAS-driven type I interferon signalling primes pancreatic cancer for necroptosis.

Pancreatic ductal adenocarcinoma (PDAC) is projected to become the second leading cause of cancer-related death within this decade. Here, we show that its major driver oncogene KRAS activates the cGAS-STING-TBK1 axis, inducing a type I interferon (IFN) response that primes PDAC cells for necroptosis. Using genetically engineered mouse models, we find that cancer cell-specific deletion of caspase-8 is sufficient to trigger necroptotic cell death, eliminating most pancreatic precursor lesions. Mechanistically, KRAS-driven IFN signalling induces ISGF3-dependent expression of necroptosis-related i

Conditional dependency of oncogenic KRAS in driving ketone body catabolism and pancreatic cancer growth.

Pancreatic ductal adenocarcinoma (PDAC) is a highly lethal malignancy driven predominantly by oncogenic KRAS mutations, which enforce extensive metabolic reprogramming to support tumor progression. Although ketone body catabolism has emerged as a critical metabolic adaptation in PDAC, the signaling mechanisms that link mutant KRAS to the ketolytic machinery remain largely unexplored. Here, we identify a previously unrecognized, context-dependent role of oncogenic KRAS in driving ketone body utilization. We show that KRAS mutation alone is insufficient to fully activate ketolysis; instead, it p

CRISPR activation screens identify oncogenic lncRNAs that are susceptible to CDK4/6 inhibitor treatment.

The roles of long non-coding RNAs (lncRNAs) in tumorigenesis and therapeutic response remain largely unknown. Here we perform genome-wide and focused CRISPR activation screens to identify lncRNAs regulating palbociclib response in breast cancer cells. A synchronized two-stage proliferation screen not only characterizes tumor growth-regulating lncRNAs, but also reveals a strong negative correlation between lncRNA-mediated regulation of tumor proliferation and CDK4/6 inhibitor sensitivity. By integrating CRISPRa screen results with drug response data from 815 cancer cell lines, we identify and f

Co-infection of Oncogenic Human Papillomavirus 16 Genotype in Genital Warts: A Case Report.

Human papillomavirus (HPV) is globally one of the most common sexually transmitted infections (STIs). HPV 6 and 11, which are low-risk subtypes, are commonly associated with condyloma acuminatum. These benign lesions are typically described as papillary or cauliflower-like. We present the case of a 28-year-old uncircumcised male patient with these lesions on his foreskin. They were surgically removed, and upon histopathologic evaluation, the diagnosis of condyloma acuminatum was made. Real-time polymerase chain reaction (RT-PCR) was performed on the excised tissue for investigational purposes,

Glutathione peroxidase 4 (GPX4) as an oncogenic factor in human tumors: a pan-cancer analysis.

BACKGROUND: Tracheostomy is one of the most common procedures done in intensive care unit (ICU) patients. Decannulation is the weaning off from tracheostomy to maintain spontaneous respiration and/or airway protection. However, this step needs a near perfect coordination of brain, swallowing, coughing, phonation, and respiratory muscles. However, despite its perceived importance, there is no universally accepted protocol for this vital transition. In this systematic review of decannulation we focus attention to this important aspect of tracheostomy care. AIM: To compare the two methods of dec

Oncogenic and immunological roles of LAMP5 across cancers and its potential utility in bladder cancer.

Lysosomal associated membrane protein family member 5 (LAMP5), a recently identified member of the LAMP family, has been associated with poor prognosis in multiple cancer types; however, its precise oncogenic mechanisms remain unclear. This study systematically investigated the oncogenic and immunological functions of LAMP5 using multiple datasets. LAMP5 expression was significantly dysregulated in various cancers, highlighting its potential as a diagnostic and prognostic biomarker. GSVA indicated that LAMP5 expression is likely associated with enhanced cell proliferation and tumor invasive po

Epigenetic and oncogenic inhibitors converge to drive a metabolic catastrophe in castration-resistant prostate cancer.

Men with advanced prostate cancer are typically treated with androgen deprivation therapy, but most ultimately develop resistance and incurable disease (e.g., castration-resistant prostate cancer, CRPC). The majority of CRPCs overexpress the epigenetic enzyme EZH2 and harbor alterations in the PI3K pathway, providing 2 targetable pathways outside of the androgen receptor. Here, we show that EZH2 inhibitors synergize with PI3K, AKT, or mTORC1 inhibitors to kill CRPC in vitro and promote tumor regression in vivo. Strikingly, these agents trigger a catastrophic energy crisis by cooperatively supp

A ligandable PNT domain establishes ERG as a directly targetable oncogenic driver in prostate cancer.

The TMPRSS2:ERG gene fusion, present in approximately 50% of prostate cancers in patients of European ancestry, drives oncogenesis through aberrant overexpression of the ERG transcription factor. Despite its role as a truncal oncogenic driver, ERG has been considered undruggable due to the absence of enzymatic activity and apparent lack of ligandable pockets. Here, we demonstrate continued dependency on ERG in metastatic prostate cancer and identify a druggable pocket within its N-terminal Pointed (PNT) domain. Using an inducible shRNA system in TMPRSS2:ERG-positive VCaP cells, we show that ER…

Tumor-targeted bispecific antibodies effectively inhibit oncogenic pathways while minimizing toxicity.

Enhancing the specificity of oncogenic pathway inhibition in cancer cells improves both efficacy and tolerability of anticancer therapies. Hyperactivated Wnt signaling is a key driver in cancers such as colorectal cancer and pancreatic ductal adenocarcinoma, but its direct inhibition is limited by severe toxicities due to the importance of Wnt signaling in normal tissues, particularly the gut and bone. To address this challenge, we designed bispecific antibodies that selectively target surface proteins of oncogenic signaling pathways in tumor cells while sparing normal cells. Using single-cell
