Effects of phytoestrogens and other plant-derived compounds on mesenchymal stem cells, bone maintenance and regeneration.

Phytoestrogens and other plant-derived compounds and extracts have been developed for the treatment of menopause-related complaints and diso … osteogenic versus adipogenic TFs. Preventing the age- and hormone deficiency-related shift towards adipogenesis without provoking adverse estrogenic effects represents a very promising strategy for treating bone loss and other metabolic diseases beyond bone. Research on plant-derived compounds will have to be pursued in vitro as well as in preclinical studies …

SPSB1 Promotes Subcutaneous Adipose Hyperplasia in Facial Port-Wine Stains by Controlling HDAC1 Degradation and Stability Through Two Distinct Proteolytic Pathways.

Facial port-wine stains (PWS) are often complicated by subcutaneous adipose hyperplasia, yet the underlying cellular and molecular mechanisms remain unclear. Here, using single-cell RNA sequencing of facial adipose tissues, we identified a distinct subpopulation of adipose stem and progenitor cells that expressed high levels of SPSB1 in hypertrophic PWS. SPSB1 expression was transcriptionally upregulated by FOSL1. Functional assays revealed that SPSB1 potently promoted adipogenesis both in vitro and in vivo. Mechanistically, SPSB1, acting as an adapter for the Cullin-RING E3 ligase complex, di…

Role of endogenous incretin hormones, GLP-1 and GIP, in cardiovascular physiology.

Obesity, type 2 diabetes (T2D), and cardiovascular disease are closely related conditions contributing to the global rise in cardiometabolic … utilization during ischemia. Endogenous GIP is involved in insulin secretion, β-cell survival, and adipogenesis. In obesity and T2D, incretin secretion and insulinotropic effects are altered. The therapeutic potential of GLP-1 receptor agonists and emerging dual GLP-1/GIP agonists has been shown to aid in managing metabolic dysfunction and, more recently, in …

Dual-energy computed tomography quantification of creeping fat in Crohn's disease: A new radiologic biomarker for transmural disease monitoring.

Accurate, noninvasive assessment of disease activity remains one of the central challenges in the management of Crohn's disease (CD). The st … ression of peroxisome proliferator-activated receptor gamma 2 and occludin, implicating CrF in both adipogenesis and intestinal barrier dysfunction. Crucially, CrF remained elevated during clinical remission, underscoring its potential as a more sensitive marker of subclinical disease persistence than conventional activity indices alone. Navigating the Complexity: A Comprehensive Review of GSK-3 Inhibition in Regenerative Medicine.

Glycogen synthase kinase-3 (GSK-3) is a central regulator of numerous cellular signaling pathways, with critical roles in metabolism, proliferation, differentiation, and tissue regeneration. This review explores the multifaceted effects of pharmacological GSK-3 inhibition across multiple body districts, focusing on its highly context-dependent impact. It has been proven that blocking GSK-3 promotes embryonic stem cell self-renewal, neuronal differentiation, osteogenesis, and dentinogenesis, while it has an inhibitory effect on adipogenesis. GSK-3 inhibition effects on muscle, heart, liver, and…

The Adipose-Sympathetic Nerve Crosstalk: FSTL1 as an Adipocyte-Derived Neurotrophic Factor for WAT Browning.

Follistatin-like 1 (FSTL1) has been considered a vital adipokine. Several lines of evidence have shown that FSTL1 is essential for white adipose tissue (WAT) adipogenesis, brown adipose tissue involution, and thermogenesis. However, the roles of FSTL1 in WAT browning and its underlying mechanisms are not fully understood. Our data show that FSTL1 overexpression induces WAT browning, enhances energy metabolism, and promotes glucose uptake in male mice at ambient room temperature. Overexpression of FSTL1 specifically in WAT prevents metabolic dysfunction in mice with diet-induced obesity. Mice w…

SETD6-mediated methylation of PPARγ establishes a transcriptional feedback circuit promoting lipid accumulation in liver-derived cells.

Peroxisome proliferator-activated receptor gamma (PPARγ) is a key transcriptional regulator of genes mediating adipogenesis (fat-cell differentiation), and lipid storage in several cell types like hepatocytes. As such, its regulation is crucial for cell and organismal physiology. Indeed, PPARγ's activity is regulated by multiple mechanisms, including post-transcriptional modifications, which, when dys-coordinated, may contribute to the pathogenesis of various states, including obesity, insulin resistance, and fatty liver disease. Here, we demonstrate that SETD6 binds to and methylates PPARγ at…

Targeting the STK39/ARID2 Axis to Inhibit NF-κB Signaling: A Novel Pathway for Mesenchymal Stem Cell Osteogenic Differentiation in Osteoporosis Management.

OBJECTIVES: Osteoporosis (OP) is a prevalent bone disease characterized by reduced bone mass and increased fracture risk, in part due to imp … induction. ARID2 silencing reduced BMSC proliferation and osteogenic differentiation while favoring adipogenesis. Conversely, STK39 overexpression enhanced proliferation and osteogenic differentiation and attenuated nuclear factor κB (NF-κB) signaling during osteogenic induction; these effects were reversed by ARID2 silencing. CONCLUSION: The STK39-ARID2 axis …

Activation of taf7l promotes sensory nerve regeneration by enhancing conditioning effect.

Peripheral nervous system (PNS) neurons can regenerate after injury, yet functional recovery is often limited, especially following proximal … and axonal length. These findings identify Taf7l, previously known for roles in spermatogenesis and adipogenesis, as a novel transcriptional regulator in neurons. Taf7l is induced by nerve injury and modulates neurite outgrowth, suggesting it as a potential therapeutic target to enhance PNS regenerative capacity.

Tazarotene ameliorates steroid-induced osteonecrosis of the femoral head by activating autophagy.

BACKGROUND: Steroid-induced osteonecrosis of the femoral head (SIONFH) is a debilitating condition characterized by lipid accumulation and bone structural collapse. A key pathogenic mechanism involves downregulated osteogenesis and upregulated adipogenesis of bone marrow mesenchymal stem cells (BMSCs). Autophagy has been implicated in lipid metabolism and bone homeostasis. This study investigated whether activating autophagy could ameliorate SIONFH. METHODS: Lipid droplet based high-throughput screening of an active compound library was performed on dexamethasone (DEX)-treated rat BMSCs. Furt…

Zfp217 Regulates Hepatic de Novo Lipogenesis via METTL3-Mediated N6-Methyladenosine Modification of SREBF1.

Zinc finger protein 217 (Zfp217) mediates adipogenesis via an N6-methyladenosine (m6A)-dependent mechanism; however, its role in hepatic lipid metabolism is unexplored. Nonalcoholic fatty liver disease (NAFLD), characterized by hepatic triglyceride (TG) accumulation resulting from disrupted lipid homeostasis, lacks well-defined epigenetic regulatory mechanisms. Here, we report that global Zfp217 heterozygous knockout alleviates high-fat diet (HFD)-induced hepatic steatosis in mice, as evidenced by reduced liver weight, decreased hepatic and serum TG and total cholesterol (T-CHO) levels, and he…

Targeting mineralocorticoid receptors to treat metabolic diseases via the adipocyte.

The mineralocorticoid receptor (MR), classically known for its role in electrolyte homeostasis, has emerged since the early 2000s as a key regulator of adipocyte biology. MR activation induces oxidative stress and inflammation, promotes white adipogenesis, and suppresses brown fat differentiation, potentially contributing to obesity and metabolic syndrome (MetS). In mice, MR antagonists (MRAs) have shown beneficial metabolic effects by reversing adipose tissue dysfunctions, counteracting excessive expansion of fat depots, and enhancing brown adipose tissue (BAT) thermogenesis. Clinical transla…
