Differential effects of a cafeteria diet and GSPE preventive treatments on the enterohormone secretions of aged vs. young female rats.

Grape seed derived procyanidins (GSPE) have been shown to effectively prevent intestinal disarrangements induced by a cafeteria diet in young rats. However, little is known about the effects of procyanidins and cafeteria diet on enterohormone secretion in aged rats, as the ageing processes modify these effects. To study these effects in aged rats, we subjected 21-month-old and young 2-month-old female rats to two sub-chronic preventive GSPE treatments. After three months of cafeteria diet administration, we analysed the basal and stimulated secretion and mRNA expression of CCK, PYY and GLP-1,

Glycoprotein 2 in health and disease: lifting the veil.

In 2020, we discovered glycoprotein 2 (GP2) variants associated with pancreatic cancer susceptibility in a genome-wide association study involving the Japanese population. Individuals carrying a missense coding variant (rs78193826) in the GP2 gene resulting in a p.V432M substitution had an approximately 1.5-fold higher risk of developing pancreatic cancer than those without this variant. GP2 is expressed on the inner surface of zymogen granules in pancreatic acinar cells, which are responsible for the sorting, storage and secretion of digestive enzymes. Upon neuronal, hormonal, or other stimul

Effect of magnesium on phosphorus and calcium metabolism.

Magnesium is an element that occurs ubiquitously in nature. Magnesium and calcium metabolism are closely related. The intestinal absorption and the renal excretion of the two ions are interdependent. The relationship between phosphorus and magnesium metabolism is more difficult to demonstrate. The most frequent causes of hypomagnesemia in children are reduced intake, impaired intestinal absorption, renal loss and genetic diseases. Hypomagnesemia is reflected clinically in the nervous system, and there are neurophysiological and metabolic changes. Severe hypomagnesemia induces secondary hypocal

Enterocyte-specific ATGL overexpression affects intestinal and systemic cholesterol homeostasis.

Enterocytes of the small intestine (SI) play an important role in maintaining systemic lipid levels by regulating dietary lipid absorption and postprandial lipoprotein secretion. An excessive amount of dietary-derived triglycerides (TGs) taken up by the apical side of enterocytes or basolaterally internalized lipoprotein remnants can be transiently stored in cytosolic lipid droplets (cLDs). As mice lacking adipose TG lipase (ATGL) in the SI display massive accumulation of cLDs but also delayed cholesterol absorption, we hypothesized that SI-specific overexpression of ATGL (Atgl iTg) might have

Oral Delivery of Pentameric Glucagon-Like Peptide-1 by Recombinant Lactobacillus in Diabetic Rats.

Glucagon-like peptide-1 (GLP-1) is an incretin hormone produced by intestinal cells and stimulates insulin secretion from the pancreas in a glucose-dependent manner. Exogenously supplied GLP-1 analogues are used in the treatment of type 2 diabetes. An anti-diabetic effect of Lactobacillus in lowering plasma glucose levels and its use as a vehicle for delivery of protein and antibody fragments has been shown previously. The aim of this study was to employ lactobacilli as a vehicle for in situ production and delivery of GLP-1 analogue to normalize blood glucose level in diabetic GK (Goto-Kakizak

Transcriptional regulatory factor X6 (Rfx6) increases gastric inhibitory polypeptide (GIP) expression in enteroendocrine K-cells and is involved in GIP hypersecretion in high fat diet-induced obesity.

Gastric inhibitory polypeptide (GIP) is an incretin released from enteroendocrine K-cells in response to nutrient ingestion. GIP potentiates glucose-stimulated insulin secretion and induces energy accumulation into adipose tissue, resulting in obesity. Plasma GIP levels are reported to be increased in the obese state. However, the molecular mechanisms of GIP secretion and high fat diet (HFD)-induced GIP hypersecretion remain unclear, primarily due to difficulties in separating K-cells from other intestinal epithelial cells in vivo. In this study, GIP-GFP knock-in mice that enable us to visuali

Effects of novel sulfated-rhamnoglucuronan isolated from Korean seaweed Ulva pertusa on IgA-mediated intestinal-immune system activation in naïve C3H/HeN mice.

A previous study identified a novel sulfated-rhamnoglucuronan structure in Ulva pertusa polysaccharide (UPP). The present study evaluates the intestinal immunostimulatory effects of UPP in naïve C3H/HeN mice. Oral administration of UPP significantly enhanced the secretion of IgA and associated cytokines, including interleukin (IL)-6, IL-10, and transforming growth factor (TGF)-β, from Peyer's patch (PP) cells in vivo. Additionally, the supernatant from PP cell cultures stimulated the proliferation of bone marrow cells. Histological analysis revealed strong stimulation of PP by UPP, as evidence

Milk peptides found in human jejunum induce enteroendocrine hormone secretion and inhibit DPP-IV.

The strong effect of protein digestion products on gastrointestinal hormone release is well recognized. However, little is known about the specific characteristics of hormone inducing peptides. A detailed analysis of food-derived peptides remaining in the human intestinal lumen after protein ingestion would constitute a practical strategy for the targeted identification of hormone inducing and DPP-IV inhibitory peptides. In this study, in vivo gastrointestinal resistant peptides derived from casein and whey protein were evaluated in epithelial intestinal cells. The secretion of cholecystokinin

Giardia duodenalis: Role of secreted molecules as virulent factors in the cytotoxic effect on epithelial cells.

During the course of giardiasis in humans and experimental models, G. duodenalis trophozoites express and secrete several proteins (ESPs) affecting structural, cellular and soluble components of the host intestinal milieu. These include the toxin-like molecules CRP136 and ESP58 that induce intestinal hyper-peristalsis. After the completion of the Giardia genome database and using up-to date transcriptomic and proteomic approaches, secreted 'virulence factors' have also been identified and experimentally characterized. This repertoire includes arginine deiminase (ADI) that competes for arginine

Inhibition of gastric acid secretion by the aqueous extract and purified extracts of Stachytarpheta cayennensis.

Stachytarpheta cayennensis Schauer (Verbenaceae) is used in folk medicine to treat gastric and intestinal disturbances. The freeze-dried aqueous extract of the whole plant tested to rodents up to the dose of 2 g kg-1, p.o., did not produce signs of toxicity. The extract (0.5-2 g kg-1, p.o.) increased the intestinal motility and protected mice against ulcers induced by restraintin-cold, ethanol or indomethacin. Injected into the duodenal lumen the extract inhibited the basal acid secretion as well as that induced by histamine and bethanecol in pylorus-ligated mice. Partition of the aqueous extr

[Study of Strongyloides stercoralis in the duodenal juice obtained by the examination of a string capsule or the Enterotest].

We present our experience in the investigation of Strongyloides stercoralis in the duodenal juice obtained by the "string capsule" o Enterotest, in 1,511 patients from the Gastroenterology Service of the National Hospital "Edgardo Rebagliati Martins" IPSS, between January 1985 to July 1990. We found 36 patients parasitized with Strongyloides stercoralis, an incidence of 2.4%. This method was good tolerated and have proved to be simple, fast, effective and allowing to examine simultaneously many patients at a very low cost.

Multiomics Dissection of the Mechanisms Underlying Eicosapentaenoic Acid Astaxanthin Ester-Mediated Protection Against High-Fat Diet-Induced Gut Barrier Injury.

The intestinal barrier proves exquisitely sensitive to dietary cues and is intimately linked to the progression of intestinal, metabolic, and systemic disorders. This study explored the effects of eicosapentaenoic acid astaxanthin ester (EA) on high-fat-diet (HFD)-induced intestinal barrier damage based on the modification of gut microecology. The results showed that EA inhibited obesity development, dyslipidemia, and colon inflammation, suppressed colon pathological damage, and restored the intestinal barrier by upregulating ZO-1 and Occludin, but these improvement effects were lost in pseudo

IFN-γ-dependent regulation of intestinal epithelial homeostasis by NKT cells.

Intestinal homeostasis is maintained through the combined functions of epithelial and immune cells that collaborate to preserve the integrity of the intestinal barrier. However, the mechanisms by which immune cell populations regulate intestinal epithelial cell (IEC) homeostasis remain unclear. Here, we use a multi-omics approach to study the immune-epithelial crosstalk and identify CD1d-restricted natural killer T (NKT) cells as key regulators of IEC biology. We find that NKT cells are abundant in the proximal small intestine and show hallmarks of activation at steady state. Subsequently, NKT

Chitosan Oligosaccharide Attenuates Lipopolysaccharide-Induced Intestinal Barrier Dysfunction through Suppressing the Inflammatory Response and Oxidative Stress in Mice.

This study was conducted to investigate the protective effect of chitosan oligosaccharide (COS) against lipopolysaccharide (LPS)-induced intestinal injury. The results demonstrated that COS improved the mucosal morphology of the jejunum and colon in LPS-challenged mice. COS alleviated the LPS-induced down-regulation of tight junction protein expressions and reduction of goblet cells number and mucin expression. The mRNA expressions of anti-microbial peptides secreted by the intestinal cells were also up-regulated by COS. Additionally, COS decreased pro-inflammatory cytokine production and neut

Characterization of a Lactobacillus gasseri strain as a probiotic for female vaginitis.

Vaginitis, a prevalent gynecological condition in women, is mainly caused by an imbalance in the vaginal micro-ecology. The two most common types of vaginitis are vaginal bacteriosis and vulvovaginal candidiasis, triggered by the virulent Gardnerella vaginalis and Candida albicans, respectively. In this study, a strain capable of inhibiting G. vaginalis and C. albicans was screened from vaginal secretions and identified as Lactobacillus gasseri based on 16S rRNA sequences. The strain, named L. gasseri VHProbi E09, could inhibit the growth of G. vaginalis and C. albicans under co-culture condit

High-protein compound yogurt with quinoa improved clinical features and metabolism of high-fat diet-induced nonalcoholic fatty liver disease in mice.

Gut microbiota dysbiosis plays a crucial role in the occurrence and progression of nonalcoholic fatty liver disease (NAFLD), which may be in … gut microbiota was disturbed by HFD consumption. Particularly, intestinal probiotics and beneficial intestinal secretions were increased, leading to the expression of glucagon-like peptide-1 in the colon, contributing to NAFLD. Furthermore, endotoxemia and systemic inflammation in HFD-fed mice were restored to the level of control mice when they were fed yogurt and qui …

A comparison of intragastric and intrajejunal feedings in neonatal piglets.

Intrajejunal feedings by continuous infusion of nutrients into the proximal jejunum have been advocated for premature and low-birthweight infants. This technique eliminates the risks of gastric distention and aspiration associated with intragastric feedings. However, intrajejunal feedings disregard the normal physiologic responses to gastric feedings and completely bypass the stomach and duodenum, thus eliminating the important contributions to intestinal function of salivary enzymes, pancreatic secretions, and duodenal osmoregulation and absorption. The objective of this investigation is the

Bacteroides Fragilis-Derived Outer Membrane Vesicles Deliver MiR-5119 and Alleviate Colitis by Targeting PD-L1 to Inhibit GSDMD-Mediated Neutrophil Extracellular Trap Formation.

Inflammatory bowel disease (IBD) results from a breakdown in the symbiotic relationship between the intestinal commensal microflora and the mucosal immune system. Non-toxigenic Bacteroides fragilis, a common human colon symbiote, has been shown to alleviate colitis. However, the underlying mechanisms of this alleviation remain incompletely understood. Herein, it is demonstrated that promoting the secretion of B. fragilis outer membrane vesicles (BfOMVs+) enhances its ability to alleviate dextran sodium sulfate (DSS)-induced colitis, while inhibiting B. fragilis OMV secretion (BfOMVs-) reduces

ATP depletion in anthrax edema toxin pathogenesis.

Anthrax lethal toxin (LT) and edema toxin (ET) are two of the major virulence factors of Bacillus anthracis, the causative pathogen of anthrax disease. While the roles of LT in anthrax pathogenesis have been extensively studied, the pathogenic mechanism of ET remains poorly understood. ET is a calmodulin-dependent adenylate cyclase that elevates intracellular cAMP by converting ATP to cAMP. Thus, it was postulated that the ET-induced in vivo toxicity is mediated by certain cAMP-dependent events. However, mechanisms linking cAMP elevation and ET-induced damage have not been established.
