Single-cell analysis of the epigenome and 3D chromatin architecture in the human retina.

Most genetic risk variants linked to ocular diseases are non-protein coding and presumably contribute to disease through dysregulation of gene expression, however, deeper understanding of their mechanisms of action has been impeded by an incomplete annotation of the transcriptional regulatory elements across different retinal cell types. To address this knowledge gap, we carried out single-cell multiomics assays to investigate gene expression, chromatin accessibility, DNA methylome and 3D chromatin architecture in human retina, macula, and retinal pigment epithelium (RPE)/choroid. We identifie…

The Effect of Tonsillectomy on Sleep Architecture in Pediatric Patients With Obstructive Sleep Apnea.

OBJECTIVES: To evaluate the impact of pediatric adenotonsillectomy (T&A) on sleep architecture, related sleep factors, and respiratory parameters in children with obstructive sleep apnea (OSA), stratified by disease severity. METHODS: We analyzed 300 consecutive children who underwent full-night polysomnography (PSG) before and after T&A between January 2021 and March 2022. Patients were stratified into severe (apnea-hypopnea index [AHI] > 10 events/h or SpO2  < 80%) and very severe (AHI > 30 events/h) OSA groups. Correlations were assessed using Spearman analysis. RESULTS: The cohort includ…

Uncovering the genetic architecture and evolutionary roots of androgenetic alopecia in African men.

Androgenetic alopecia is a highly heritable trait. However, much of our understanding about the genetics of male-pattern baldness comes from … e limits of statistical power, we did not find evidence that continental differences in the genetic architecture of baldness are due to Neanderthal introgression. While most loci that are associated with androgenetic alopecia do not have large integrative haplotype scores or fixation index statistics, multiple baldness-associated SNPs near the EDA2R and AR gen …

A novel strategy for enhanced schizophrenia detection using established CNN architectures.

Schizophrenia, a complex psychotic condition, is challenging to diagnose due to its reliance on clinical assessments and behavioral evaluations. Neuroimaging studies, particularly structural MRI, have revealed reductions in grey matter volume in brain regions such as the temporal lobe and insula. This study combines deep learning and neuroimaging for precise automatic detection of schizophrenia. Using T1-weighted coronal MRI data from three publicly accessible datasets (MCICShare, COBRE, UCLA), a DeepLabv3+ model with ResNet50-based segmentation was employed to isolate the temporal lobe and in

Thick or Thin? Implications of Cartilage Architecture for Osteoarthritis Risk in Sedentary Lifestyles.

Osteoarthritis (OA) is a leading cause of disability worldwide and is characterized by the gradual degradation of articular cartilage in weight-bearing joints, notably the knees and hips. However, the primary morphological and anatomical determinants of the disease onset and progression remain unclear. This narrative overview examines how variations in cartilage thickness-traditionally viewed as a biomechanical protective feature-can paradoxically compromise metabolic homeostasis during prolonged sedentary behavior. Intriguingly, compelling evidence suggests that despite its superior load-bear

Effect of Internal Architecture on the Elasticity of Microgel Monolayers at the Air/Water Interface.

Soft microgels can deform and adsorb at liquid interfaces, forming monolayers with tunable compressibility and elasticity. Their deformabilityor softnessis governed by the internal architecture and inhomogeneity of the polymeric network. However, establishing a direct correlation between single-particle properties and their collective interfacial behaviorssuch as self-assembly and mechanical responseremains a fundamental challenge. In this work, we employ core-shell microgels with controllable internal architectures to investigate how single-particle softness influences the elasticity of m…

Polygenic Scores for Dizygotic Twinning: Insights into the Genetic Architecture of Female Fertility.

Natural dizygotic twinning (DZT) results from hyper-ovulation and is considered an indicator of female fertility. DZT has low polygenicity, with only 0.20% of SNPs estimated to have a nonzero effect. A polygenic score (PGS) for DZT explains 1.6% of variance in DZT liability and we observe an odds ratio of 2.29 between the 1st and the 10th PGS decile. The PGS is higher in mothers of naturally conceived twins compared to mothers who received fertility (MAR) treatments in both the Netherlands and Norway. The largest differences were observed for mothers who received hormonal ovulation induction,

Polygenic scores for dizygotic twinning: insights into the genetic architecture of female fertility.

PURPOSE: Natural dizygotic twinning (DZT) results from hyper-ovulation and is an indicator of female fertility. However, some traits linked to DZ twinning are also associated with infertility. We examined the relationship between DZT and female (in)fertility using recent GWAS findings. METHODS: We investigated the genetic architecture of DZT and compared polygenic scores (PGS) for DZT between mothers of naturally conceived DZ twin pregnancies and mothers who required fertility treatments (MAR) in the Netherlands Twin Register (NTR) and the Norwegian Mother, Father, and Child Cohort Study (MoB…

Asymmetries in the Architecture of ON and OFF Arbors in ON-OFF Direction-Selective Ganglion Cells.

Direction selectivity is a fundamental feature in the visual system. In the retina, direction selectivity is independently computed by ON and OFF circuits. However, the advantages of extracting directional information from these two independent circuits are unclear. To gain insights, we examined the ON-OFF direction-selective ganglion cells (DSGCs), which recombine signals from both circuits. Specifically, we investigated the dendritic architecture of these neurons with the premise that asymmetries in architecture will provide insights into function. Scrutinizing the dendrites of dye-filled ON…

Alterations in Inflammatory Markers and Tissue Architecture in the Gerbil Prostate Following Castration.

SummaryOrchiectomy induces atrophy of epithelial cells in the prostate gland, stimulated by androgen deprivation. The objective of the current study was to assess the changes arising from tissue remodeling during short periods of androgen deprivation in the ventral prostate of Mongolian gerbils. Adult male gerbils were divided into groups: control and castrated, euthanized on the 3rd (Ca3d), 7th (Ca7d), and 14th (Ca14d) days post-orchiectomy (n = 7/group). The ventral lobe of the prostate was submitted for histological, stereological, morphometric, serological, and immunohistochemical analyses

Stromal architecture and fibroblast subpopulations with opposing effects on outcomes in hepatocellular carcinoma.

Dissecting the spatial heterogeneity of cancer-associated fibroblasts (CAFs) is vital for understanding tumor biology and therapeutic design. By combining pathological image analysis with spatial proteomics, we revealed two stromal archetypes in hepatocellular carcinoma (HCC) with different biological functions and extracellular matrix compositions. Using paired single-cell RNA and epigenomic sequencing with Stereo-seq, we revealed two fibroblast subsets CAF-FAP and CAF-C7, whose spatial enrichment strongly correlated with the two stromal archetypes and opposing patient prognosis. We discovere

Engineering Two-Dimensional Nanobody-Origami Architectures for Enhanced Antiviral Activity.

The nanoscale organization of binding ligands offers a powerful strategy to combat viral infections. Herein, we report a programmable 2D DNA origami platform that enables nanoscale control of multivalent nanobody (Nb) spatial patterns for a broad-spectrum antiviral application. By site-selective Nb-DNA conjugation and spatial positioning on 2D origami, we synthesized origami-Nb nanoarchitectures with Nb patterns rationally designed to approximately match the geometric presentation of viral surface proteins. We demonstrate that Nb configurations greatly enhanced viral binding affinity and neutr…

Investigating the common genetic architecture and causality of metabolic disorders with neurodegenerative diseases.

BACKGROUND: The co-occurrence of metabolic dysfunction and neurodegenerative diseases suggests a genetic link, yet the shared genetic architecture and causality remain unclear. We aimed to comprehensively characterise these genetic relationships. METHODS: We investigated genetic correlations among four neurodegenerative diseases and seven metabolic dysfunctions, followed by bidirectional Mendelian randomisation (MR) to assess potential causal relationships. Pleiotropy analysis (PLACO) was used to detect the pleiotropic effects of genetic variants. Significant pleiotropic loci were refined and…

Insights into the genetic architecture of cerebellar lobules derived from the UK Biobank.

In this work we endeavor to further understand the genetic architecture of the cerebellum by examining the genetic underpinnings of the different cerebellar lob(ul)es, identifying their genetic relation to cortical and subcortical regions, as well as to psychiatric disorders, as well as traces of their evolutionary trajectories. We confirm the moderate heritability of cerebellar volumes, and reveal genetic clustering and variability across their different substructures, which warranted a detailed analysis using this higher structural resolution. We replicated known genetic correlations with se…

The Relationship between White Matter Architecture and Language Lateralization in the Healthy Brain.

Interhemispheric anatomical differences have long been thought to be related to language lateralization. Previous studies have explored whet … s (IFOF) and forceps minor tracts. These results suggest that diffusion measures of microstructural architecture as well as geometrical features of reconstructed white matter tracts play a role in language lateralization. People with increased dependence on the right or both frontal hemispheres for language processing may have more developed commissural fibers …

SOLID: minimizing tissue distortion for brain-wide profiling of diverse architectures.

Brain-wide profiling of diverse biological components is fundamental for understanding complex brain pathology. Despite the availability in whole-brain imaging, it is still challenging to conduct multiplexed, brain-wide analysis with current tissue clearing techniques. Here, we propose SOLID, a hydrophobic tissue clearing method that can minimize tissue distortion while offering impressive clearing performance. SOLID achieves high-quality imaging of multi-color labeled mouse brain, and the acquired datasets can be effectively registered to the Allen Brain Atlas via commonly-used algorithms. SO

Brain metabolites are associated with sleep architecture and cognitive functioning in older adults.

Sleep deficits are a possible risk factor for development of cognitive decline and dementia in older age. Research suggests that neuroinflammation may be a link between the two. This observational, cross-sectional study evaluated relationships between sleep architecture, neuroinflammation and cognitive functioning in healthy older adults. Twenty-two adults aged ≥60 years underwent whole-brain magnetic resonance spectroscopic imaging (in vivo method of visualizing increased brain temperatures as a proxy for neuroinflammation), supervised laboratory-based polysomnography, and comprehensive neuro…
