Inferred spoligoforest topology unravels spatially bimodal distribution of mutations in the DR region.

Biomarkers of Mycobacterium tuberculosis complex (MTBC) mutate over time. Among the biomarkers of MTBC, spacer oligonucleotide type (spoligotype) and mycobacterium interspersed repetitive unit (MIRU) patterns are commonly used to genotype clinical MTBC strains. In this study, we present an evolution model of spoligotype rearrangements using MIRU patterns to disambiguate the ancestors of spoligotypes. We use a large patient dataset from the United States Centers for Disease Control and Prevention (CDC) to generate this model. Based on the contiguous deletion assumption and rare observation of c

A novel point variant in NTRK3, R645C, suggests a role of this gene in the pathogenesis of Hirschsprung disease.

Hirschsprung disease (HSCR) is a developmental disorder characterized by the absence of ganglion cells in the myenteric and submucosal plexuses due to a defect in the migration process of neural crest neuroblasts. Manifestation of the disease has been linked to the dysfunction of two principal signalling pathways involved in the enteric nervous system (ENS) formation: the RET-GDNF and the EDN3-EDNRB receptor systems. However, the NTF3/NTRK3 signalling pathway plays an essential role in the development of the ENS suggesting a potential role for those genes in the pathogenesis of HSCR. We have s

Mitochondrial DNA-related disorders.

Mitochondrial diseases are a group of disorders due to a mitochondrial respiratory chain deficiency. They may depend on mitochondrial genome (mtDNA-related disorders) as well as on a nuclear genome defect (nDNA-related disorders). mtDNA-related disorders encompass an increasing number of clinical pictures associated with more than 250 different provisional or confirmed pathogenic changes in mtDNA. Although some clinical syndromes are nosologically defined, most of the cases present with polymorphous phenotypes ranging from pure myopathy to multi-system involvement. Complexity of mitochondrial

Segregation of a missense mutation in the amyloid beta-protein precursor gene with familial Alzheimer's disease.

In 1991 we described a missense mutation in the amyloid beta-protein precursor (AbetaPP) gene in two familial Alzheimer's disease (FAD) kindreds. This gene encodes the amyloid beta peptide deposited in senile plaques in AD. We made four predictions based upon these results: 1. Other FAD kindreds would be identified with AbetaPP mutations; 2. FAD is genetically heterogeneous; 3. Abeta deposition is central to the pathogenesis of AD and 4, Regulatory variants in the AbetaPP gene lead to late onset AD. In the ensuing years substantial evidence has accrued in support of these predictions. Nineteen

[Cadasil: a case with molecular diagnosis].

CADASIL disease (Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy) was described in 1991 by Tourmier-Lasserve. Two years later the same authors described its association with chromosome 19; nonetheless, the mutations in gene Notch3 were not described until 1996. Clinical findings depend on the age at onset. The early form of the illness is found in young patients, generally less than 30 years old, and the main clinical manifestation is a migraine headache with subcortical lesions in the white matter, while in the later form ischemic events and behavior

[Screening for mutations in the PAH gene in Chinese: a new splice and a novel polymorphic mutation].

OBJECTIVE: to screening for novel mutations in the phenylalanine hydroxylase (PAH) genes in Chines as to investigate the molecular basis of phenylketonuria and to raise its diagnosis rate. METHODS: We applied the polymorase chain reaction (PCR) on amplified DNA fragment harboring exon 4 of the PAH gene. For novel mutations detected, the amplified fragments were sequenced by using ds-DNA cycle-sequencing method. RESULTS: Seven out of 24 cases of PKU had different kinds of band-shifting, which are subjected to two patterns of polymorphism. The first one, only in one case, has one mutation band

[Anaemia caused by a congenital haemoglobin defect can also occur in native Dutch patients].

An 11-year-old boy of Dutch descent had suffered from tiredness, pallor and poor appetite for many years. There was no indication of chronic infection. Iron supplements did not produce sufficient improvement in the symptoms. Following analysis using fast protein liquid chromatography and DNA sequencing analysis, it appeared that this was a case of β-thalassaemia intermedia, caused by a point mutation in the promotor region of the β-globulin gene. The boy's parents both proved to be heterozygotes for this point mutation. A second patient, a 5-year-old girl of Dutch descent, suffered from an ict…

Analysis and expression of Rab38 in oculocutaneous lung disease.

Rab38 is a low-molecular-weight G-protein highly expressed in melanocytes of the skin and alveolar type II cells in the lung. A point mutation in the postulated GTP/GDP-interacting domain of Rab38 has been identified as the genetic lesion responsible for oculocutaneous albinism (OCA) in chocolate (cht) mice. Another point mutation that prevents translation of Rab38 mRNA is the molecular basis of the Ruby gene mutation causing the phenotype of OCA and prolonged bleeding time in Fawn-Hooded and Tester-Moriyama rats. Cht mice show conspicuously enlarged lamellar bodies in alveolar type II cells a…

A mouse model for achondroplasia produced by targeting fibroblast growth factor receptor 3.

Achondroplasia, the most common form of dwarfism in man, is a dominant genetic disorder caused by a point mutation (G380R) in the transmembrane region of fibroblast growth factor receptor 3 (FGFR3). We used gene targeting to introduce the human achondroplasia mutation into the murine FGFR3 gene. Heterozygotes for this point mutation that carried the neo cassette were normal whereas neo+ homozygotes had a phenotype similar to FGFR3-deficient mice, exhibiting bone overgrowth. This was because of interference with mRNA processing in the presence of the neo cassette. Removal of the neo selection m…

Homology among tet determinants in conjugative elements of streptococci.

A mutation to tetracycline sensitivity in a resistant strain of Streptococcus pneumoniae was shown by several criteria to be due to a point mutation in the conjugative omega (cat-tet) element found in the chromosomes of strains derived from BM6001, a clinical strain resistant to tetracycline and chloramphenicol. Strains carrying the mutation were transformed back to tetracycline resistance with the high efficiency of a point marker by donor deoxyribonucleic acids from its ancestral strain and from nine other clinical isolates of pneumococcus and by deoxyribonucleic acids from group D Streptoco…

Computational introspection of impact of mutation A90V and A90E in DNA gyrase a of Mycobacterium tuberculosis in developing resistance against Delafloxacin.

Quinolone group antibiotics have proven to be an effective drug against Mycobacterium tuberculosis. However, point mutation in DNA gyrase, one of the primary targets for quinolone antibiotics, has led to conformational alterations in the binding sites, leading to a change in drug pharmacodynamics. This has contributed to the development of drug-resistant strains such as multidrug-resistant strains (MDR) and an extreme drug-resistant strains (XDR). In this study, our objective was to find the impact of mutation A90V and A90E on DNA Gyrase A of Mycobacterium tuberculosis. Ten quinolone antibioti…

Synthesis and properties of the anticodon stem-loop of human mitochondrial tRNAMet containing the disease-related G or m1G nucleosides at position 37.

A single point mutation (A4435G) in the human mitochondrial tRNAMet (hmt-tRNAMet) gene causes severe mitochondrial disorders associated with hypertension, type 2 diabetes and LHON. This mutation leads to the exchange of A37 in the anticodon loop of hmt-tRNAMet for G37 and 1-methylguanosine (m1G37). Here we present the first synthesis and structural/biophysical studies of the anticodon stem and loop of pathogenic hmt-tRNAsMet.

A5814G mutation in mitochondrial DNA can cause mitochondrial myopathy and cardiomyopathy.

We describe a 5-year-old child with hypertrophic cardiomyopathy, mitochondrial myopathy, and lactic acidosis. Mitochondrial DNA analysis showed a heteroplasmic A5814G point mutation in the tRNA(Cys) gene. The mutational load was extremely high (>95%) in muscle, fibroblasts, and blood. This report expands the clinical heterogeneity of the A5814G mutation, which should be considered in the differential diagnosis of hypertrophic cardiomyopathy in childhood.
