The peptidergic brain-gut axis: influence on gastric ulcer formation.

The existence of a relationship between the brain and the formation of gastric ulcers has been suspected since the last century. The advancement of stereotaxic procedures and the use of electrical lesion or stimulation have allowed localization within the limbic system, hypothalamus and brain stem, of discrete nuclei that influence the formation of gastric ulceration in experimental animals. Recently, further progress in the understanding of how the brain may influence gastric pathogenesis has been made by the demonstration that specific peptides act in the central nervous system to induce or

Investigating the Kidney-Gut-Brain Axis in CKD: Uremic Toxins and Brain Microhemorrhages.

Alterations of gut microbiota are common in chronic kidney disease (CKD) and contribute to increased uremic toxins including indoxyl sulfate (IS), p-cresyl sulfate (pCS) and trimethylamine N-oxide (TMAO), which are linked to cerebrovascular disease risk. This study examined the kidney-gut-brain axis in CKD mice and in dialysis patients. Male and female mice with adenine-induced CKD were fed a high-amino-acid (HAA) diet to increase precursors of gut-derived uremic toxins. A subgroup of mice received antibiotics in drinking water to suppress gut microbiota and evaluate its role in toxin generati

The prairie vole gut-brain-microbiota-axis: a narrative review.

The microbiota-gut-brain axis (MGBA) has garnered considerable attention for its role in health, disease, and higher psychological processes. One area of particular importance is the relationship between the MGBA and stress. Although numerous animal models are suitable for research on stress, the number suitable for research on the impact of social stressors with translatability to humans is limited. The prairie vole is regarded as an ideal organism for probing the impact of social stress, as these animals not only exhibit social behaviours rare in mammals but also lack many drawbacks that com

A Gut-Brain Axis Underlying Succinate-Associated Compulsive-Like Behavior.

Emerging evidence indicates that gut microbiota influences host physiology and behavior through the microbiota-gut-brain axis, potentially contributing to the pathogenesis of obsessive-compulsive disorder (OCD). Our preliminary research demonstrated that mice colonized with fecal microbiota from OCD patients developed compulsive-like behaviors, associated with a significant increase in the microbial metabolite succinate. However, the mechanisms linking succinate to compulsive-like behavior remain to be fully elucidated. Here, we demonstrate that succinate is associated with significant compuls

Lactobacillus acidophilus LA3 Suppresses Neuroinflammation via the Gut-Brain Axis.

The gut-brain axis represents a pivotal pathway through which gut microbiota influence neuroinflammation and neurological health. This study identifies Lactobacillus acidophilus LA3 as a potent probiotic strain capable of mitigating neuroinflammation. LA3 demonstrated superior efficacy in suppressing LPS-induced proinflammatory cytokines in BV-2 microglia. In an LPS-induced murine model, LA3 alleviated systemic and neuroinflammation, depressive-like behaviors, and cognitive impairment, while restoring blood-brain and gut-vascular barrier integrity. LA3 reversed gut microbiota dysbiosis, enrich

Can microbiota gut-brain axis reverse neurodegenerative disorders in human?

The trillions of microbial populations residing in the gut have recently shown that they can be used as a remedy for various diseases. The gut microbiota-brain-axis interface is one unique pathway that the microbiota demonstrates its medicinal value. This medicinal value is further seen when there is a decline in gut microbial diversity (dysbiosis). Dysbiosis leads to neurodegenerative disorders (NDDs). The objective of this review is to ascertain the clinical significance of gut microbiota induced therapeutic strategies. While navigating this important area of interest, we will elucidate the

Gut-Brain Axis in Inflammatory Bowel Disease: Pathogenesis and Therapeutics.

Inflammatory Bowel Disease (IBD), encompassing Crohn's disease and ulcerative colitis, is a chronic inflammatory disorder of the gastrointestinal tract driven by complex interactions between genetic susceptibility, environmental triggers, microbial dysbiosis, and immune dysregulation. The gut microbiome, composed primarily of Firmicutes and Bacteroidetes, plays a crucial role in maintaining intestinal barrier integrity, immune balance, and neuroimmune signaling. Disruption of this microbial ecosystem is characterized by loss of beneficial short chain fatty acid producing bacteria and expansion

The gut microbiota-brain axis in Alzheimer's disease model mice.

The gut microbiota plays a pivotal role in the pathogeneses of Alzheimer's disease (AD), influencing neuroinflammation and disease progression. Understanding the role of the gut microbiota in this process is critical for uncovering novel therapeutic avenues and deepening insights into AD pathogenesis. Nevertheless, the specific gut microbiota alterations under pathogenetic stress in AD remain unclear. In this study, immunofluorescence was performed to detect β-amyloid (Aβ) deposition, hyperphosphorylated tau proteins (P-tau), and the activation of microglia in the brains of APPswe/PSEN1dE9 tra

Gut-brain axis and Parkinson's disease: navigating the microbial enigma.

Parkinson's disease (PD), traditionally seen as a Central Nervous System (CNS) disorder with dopaminergic neuron loss and α-synuclein aggregation, increasingly implicates the gut-brain axis in its pathogenesis. Gut microbiota dysbiosis precedes motor symptoms by years, linking to non-motor issues like constipation and rapid eye movement sleep behavior disorder, with reduced neuroprotective bacteria (e.g., Prevotella), impairing barrier integrity, short-chain fatty acid production, and promoting neuroinflammation. Mechanistically, misfolded α-synuclein may start in the gut, spreading via the va

The gut-brain axis underlying hepatic encephalopathy in liver cirrhosis.

Up to 50-70% of patients with liver cirrhosis develop hepatic encephalopathy (HE), which is closely related to gut microbiota dysbiosis, with an unclear mechanism. Here, by constructing gut-brain modules to assess bacterial neurotoxins from metagenomic datasets, we found that phenylalanine decarboxylase (PDC) genes, mainly from Ruminococcus gnavus, increased approximately tenfold in patients with cirrhosis and higher in patients with HE. Cirrhotic, not healthy, mice colonized with R. gnavus showed brain phenylethylamine (PEA) accumulation, along with memory impairment, symmetrical tremors and

Gut-Brain Axis in Preterm Infants with Surgical Necrotizing Enterocolitis.

Necrotizing enterocolitis (NEC) affects 5 to 10% of very low-birth-weight infants and remains a leading cause of mortality and long-term morbidity. Preterm infants with NEC, especially those requiring surgery, have higher inflammatory markers in the blood, severe white matter abnormalities on brain imaging, and adverse neurodevelopmental outcomes. This review presents current evidence regarding the clinical factors associated with brain injury in preterm infants with NEC needing surgical intervention. Studies that evaluate neuroprotective strategies to prevent brain injury are greatly needed t

Remodeling of microbiota gut-brain axis using psychobiotics in depression.

Depression is a multifaceted psychiatric disorder mainly orchestrated by dysfunction of neuroendocrine, neurochemical, immune, and metabolic systems. The interconnection of gut microbiota perturbation with the central nervous system disorders has been well documented in recent times. Indeed, alteration of commensal intestinal microflora is noted in several psychiatric disorders such as anxiety and depression, which are presumed to be routed through the enteric nervous system, autonomic nervous system, endocrine, and immune system. This review summarises the new mechanisms underlying the crosst

Role of Microbiota-Gut-Brain Axis in Regulating Dopaminergic Signaling.

Dopamine is a neurotransmitter that plays a critical role both peripherally and centrally in vital functions such as cognition, reward, satiety, voluntary motor movements, pleasure, and motivation. Optimal dopamine bioavailability is essential for normal brain functioning and protection against the development of neurological diseases. Emerging evidence shows that gut microbiota have significant roles in maintaining adequate concentrations of dopamine via intricate, bidirectional communication known as the microbiota-gut-brain axis. Aging Microbiota-Gut-Brain Axis in Stroke Risk and Outcome.

The microbiota-gut-brain-axis (MGBA) is a bidirectional communication network between gut microbes and their host. Many environmental and host-related factors affect the gut microbiota. Dysbiosis is defined as compositional and functional alterations of the gut microbiota that contribute to the pathogenesis, progression and treatment responses to disease. Dysbiosis occurs when perturbations of microbiota composition and function exceed the ability of microbiota and its host to restore a symbiotic state. Dysbiosis leads to dysfunctional signaling of the MGBA, which regulates the development and

The synergistic regulation of fat reduction mechanism by tea polyphenols through the gut-microbial-brain axis and gut-liver axis.

The occurrence and development of obesity are closely related to intestinal dysbiosis, metabolic disorders, and neuroendocrine signals. Tea polyphenols can be the key to synergistically regulating energy balance by reshaping the gut microbiota. This review innovatively proposes and systematically demonstrates that tea polyphenols mediate the cross-dialogue and synergy between the gut-brain axis and the gut-liver axis by modulating gut microbiota metabolites (such as short-chain fatty acids and secondary bile acids), thereby constructing a multi-dimensional interaction fat reduction network. Pharmacomicrobiomics and the gut-brain axis: microbial modulation of drug response in brain disorders.

Pharmacomicrobiomics is the study of how the gut microbiota affects the metabolism, efficacy, and safety of drugs. Gut microbes play a key role in neurochemical communication along the gut-brain axis and can alter responses to neuroactive medications. Dysbiosis alters pharmacodynamics by modulating the synthesis of neurotransmitters, immune function and neuroinflammation. Concurrently, it alters pharmacokinetics through microbial biotransformation, which directly changes drug bioavailability, activation, and clearance. Increasing evidence shown an association between variations in microbiome a

Gastrointestinal dysfunction after brain injury: Mechanisms and the role of the brain-gut axis.

Brain injury (BI) encompasses traumatic BI and stroke, and is a leading cause of mortality and morbidity worldwide. In addition to primary brain damage, BI can cause a series of sequelae, of which gastrointestinal (GI) dysfunction is one of the most important affecting patient outcomes. GI dysfunction, including delayed gastric emptying, increased intestinal permeability, and gut dysbiosis, is common among patients with BI. GI dysfunction not only impairs nutrient absorption and increases the risk for infection, but also enhances secondary BI by aggravating the systemic inflammatory response.

Role of the gut-microbiota-metabolite-brain axis in the pathogenesis of preterm brain injury.

Brain injury, a common complication in preterm infants, includes the destruction of the key structural and functional connections of the brain and causes neurodevelopmental disorders; it has high morbidity and mortality rates. The exact mechanism underlying brain injury in preterm infants is unclear. Intestinal flora plays a vital role in brain development and the maturation of the immune system in infants; however, detailed understanding of the gut microbiota-metabolite-brain axis in preterm infants is lacking. In this review, we summarise the key mechanisms by which the intestinal microbiota
