L-2-Oxothiazolidine-4-carboxylate and N-acetylcysteine as precursors of intracellular glutathione in human peritoneal mesothelial cells.

L-2-Oxothiazolidine-4-carboxylate and N-acetylcysteine as substrates for intracellular glutathione in human peritoneal mesothelial cells were tested. Both substances at concentrations of 0.01 mM and higher augmented the level of glutathione in mesothelial cells. L-2-Oxothiazolidine-4-carboxylate had a milder but more stable effect than N-acetylcysteine. Cells with increased concentrations of the intracellular glutathione were more resistant to injury by free radicals. When used at higher concentrations (> 1 mM), both substances became cytostatic to mesothelial cells as evidenced by growth inhi

[Clinical administration of platelet-rich plasma and growth factors to the musculoskeletal system].

Platelet-rich plasma (PRP) and growth factors have been increasing in popularity for the orthopedic treatment of degenerative and traumatic diseases. The treatment concept is based on the substitution of growth-inducing substances in tissues with low or absent regeneration capacity (cartilage, tendons) as well as for the induction or further acceleration of growth and regeneration (bone, muscle). This review article provides an overview on the clinical feasibility of usage and a summary of the current study situation.

Functional status of vascular endothelium in diabetes mellitus.

Normal endothelial cells synthesize and release biologically active substances. These substances maintain homeostasis through adequate blood flow, delivery of nutrients, activation and inhibition of coagulation proteins, prevention of thrombosis and diapedesis of leukocyte. Endothelial dysfunction implies failure of vascular endothelium to perform its normal functions of vasodilatation and vasoconstriction. It results from an imbalance between endothelium derived constricting and relaxing factors. Altered endothelial cell activity predisposes to increased production of vasoconstrictors, i.e.,

Cytokinin and growth of excised roots of Bryophyllum calycinum.

Excised roots of Bryophyllum calycinum require for growth both auxin and cytokinin. This is demonstrated by the poor growth of 2-mm root tips in a basal medium of mineral salts, sucrose, and vitamins supplemented with either an auxin or a cytokinin, and much better growth when the basal medium is supplemented with both auxin and cytokinin. However, both substances are synthesized by the root, as is demonstrated by the growth of large inocula (dry wt 6-7 mg) through many successive passages in a medium limited to mineral salts, sugar, and vitamins.

Partial purification of (a) low molecular weight ovine pineal compound(s) with an inhibiting effect on the growth of human melanoma cells in vitro.

An in vitro human melanoma cell assay was used to work up the partial purification of (a) low molecular weight (MW) substance(s) from aqueous extracts of ovine pineal tissue shown to contain a growth-inhibiting activity. A combination of paper chromatography, ion-exchange and reverse-phase high performance liquid chromatography with post-column antitumor assay has been developed. This allows a specific identification of an ovine pineal factor (MW less than 500) which inhibits the growth of human melanoma cells in vitro. The substance was partially purified to about 1,000 times as compared to t

Excessive growth in a child with craniopharyngioma and growth hormone deficiency.

In a 5-year-old boy presenting with clumsiness and excessive growth, a large craniopharyngioma was diagnosed. Biochemically, there was a deficiency of growth hormone, a hypothalamic hypothyroidism and hypocorticalism, a thyroxine binding globulin elevation, an abnormal gonadotropin secretion and a mild hyperprolactinaemia. After removal of the tumour growth stopped almost completely. Plasma insulin-like growth factor (IGF)-I was in the lower normal range. Plasma IGF-II decreased after tumour removal. It is speculated that the tumour produced a growth factor causing excessive growth.

Blood components for topical use in tissue regeneration: evaluation of corneal lesions treated with platelet lysate and considerations on repair mechanisms.

BACKGROUND: The fields of application of topically administered platelet derivatives are numerous and increasing. The use of this blood component is based on the fact that it contains growth factors and proteins of the clotting system. Studies carried out so far have been aimed at identifying these substances, assaying their content in the various types of platelet concentrate used, determining the in vivo and in vitro mechanisms of action, and trying to standardise the production methods. However, much still remains to be discovered, not only about the growth factors, but also about all those

Prenatal substance exposure and offspring development: Does DNA methylation play a role?

The period of in utero development is one of the most critical windows during which adverse conditions and exposures may influence the growth and development of the fetus as well as its future postnatal health and behavior. Maternal substance use during pregnancy remains a relatively common but nonetheless hazardous in utero exposure. For example, previous epidemiological studies have associated prenatal substance exposure with reduced birth weight, poor developmental and psychological outcomes, and increased risk for diseases and behavioral disorders (e.g., externalizing behaviors like ADHD,

Admitting powerlessness empowers recovery from substance use disorder: a conceptual model.

Some individuals use substances to cope with their post-traumatic stress disorder symptoms. The emotional, behavioral, and spiritual experience of both their post-traumatic stress disorder and substance use disorder is manifested in a sense of powerlessness, which is caused by the inability to stop using despite negative consequences related to continued use. Thus, powerlessness is a prominent factor in recovery from substance use disorder. The aims of the present theoretical article were to (a) present the consequences of the co-morbid post-traumatic stress disorder and substance use disorder

Palatal microbiota associated with membranous substances in older Japanese individuals undergoing tube feeding in long-term care: A cross-sectional study.

OBJECTIVE: Tube feeders are prone to membranous substance formation on the palate, and those with membranous substances have a risk of fever, with the probable involvement of their oral bacteria. However, the palatal microbiota of those with membranous substances has not been elucidated. Therefore, we evaluated the differences in palatal microbiota between tube-fed individuals with and without membranous substances to clarify the microbiota. MATERIALS AND METHODS: This study included 19 participants aged 65 years who required tube feeding. The participants' characteristics were collected from

Investigating Mechanisms of Stillbirth in the Setting of Prenatal Substance Use.

INTRODUCTION: Intrauterine fetal demise affects between 0.4-0.8% of pregnancies worldwide. This significant adverse pregnancy outcome continues to be poorly understood. In utero exposure to substances increases the risk of stillbirth to varying degrees according to the type of substance and degree of exposure. The aim of this qualitative narrative review is to investigate common biologic relationships between stillbirth and maternal substance use. METHODS: A PubMed literature search was conducted to query the most commonly used substances and biologic mechanisms of stillbirth. Search terms in

Cerebral reserve capacity: implications for alcohol and drug abuse.

Cerebral reserve capacity (or functional reserve) refers to the brain's ability to maintain function when confronted by degenerative processes. Functional reserve can be estimated by several associated measures, including premorbid brain size, premorbid IQ, and level of education attained. There is accumulating evidence that the magnitude of reserve capacity is important in determining the onset and progression of the clinical manifestations of neurodegenerative brain diseases. Normal aging also whittles away at this cerebral reserve, and there may be a consequent unmasking of morbid effects t

Extracellular matrix and growth factors in the pathogenesis of some craniofacial malformations.

The normal development of cranial primordia and orofacial structures involves fundamental processes in which growth, morphogenesis, and cell differentiation take place and interactions between extracellular matrix (ECM) components, growth factors and embryonic tissues are involved. Biochemical and molecular aspects of craniofacial development, such as the biological regulation of normal or premature cranial suture fusion, has just begun to be understood, thanks mainly to studies performed in the last decade. Several mutations has been identified in both syndromic and non-syndromic craniosynost

Immunotherapy for invasive fungal infections: from bench to bedside.

Immunotherapy for invasive fungal infections has been an area of significant research and clinical interest since the first-half of the twentieth century when hyperimmune equine immunoglobulin was first successfully administered to patients with acute and chronic cryptococcal meningitis. Since that time, effective antifungal compounds have been developed, but a much more complex array of host disorders have also emerged creating an even greater need for immunotherapy in conjunction with conventional antifungal therapy. In this review, the scientific foundation supporting the use of various imm

[Thyroid and growth factors].

Although TSH is the major regulator of thyroid growth and function, the role of TSH in thyroid cell growth in vitro is controversial. Recent studies have shown that a number of growth factors, cytokines, vitamins, hormones and other reagents including iodide, lithium or retinoids modulate proliferation of cultured thyroid cells. Insulin-like growth factors (IGFs) stimulate growth of thyroid cells from a variety of species and also enhance expression of differentiated functions stimulated by TSH. Epidermal growth factor (EGF), fibroblast growth factor (FGF), and Transforming growth factor (TGF)

Therapeutic modulation of growth factors and cytokines in regenerative medicine.

Regeneration that takes place in the human body is limited throughout life. Therefore, when organs are irreparably damaged, they are usually replaced with an artificial device or donor organ. The term "regenerative medicine" covers the restoration or replacement of cells, tissues, and organs. Stem cells play a major role in regenerative medicine by providing the way to repopulate organs damaged by disease. Stem cells have the ability to self renew and to regenerate cells of diverse lineages within the tissue in which they reside. Stem cells could originate from embryos or adult tissues. Growth

[The role of locally synthesized growth factors and cytokines in pathogenesis of osteoporosis].

Bone remodelling is a complex process involving a number of cellular functions directed toward the coordinated resorption and formation of new bone. Bone remodelling is regulated by systemic hormones and by local factors. The local factors are synthetized by skeletal cells and include growth factors and cytokines. The end product of remodeling is the maintenance of a mineralized bone matrix, and the major organic component is the collagen. Local factors play an important role in pathophysiology of osteoporosis as systemic hormones.
