[Lafora disease and founder effect in a small neotropical locality].

The Lafora disease is an uncommon genetic condition. Four cases (two families) were detected in Zarcero, a small town in Costa Rica (population under 2000). They belonged to two separate consanguineous marriages but both families had common ancestors. The diagnosis of Lafora disease was confirmed by liver biopsy in one of the patients. The ages of onset were 13, 14, 16 and 17 years. Patients died after four, nine, six and five years of severe progressive physical and mental deterioration, respectively. The gene for Lafora disease arrive to Zarcero from one of its founders. There are no other c

Implication of molecular analysis of HLA-A*02 subtyping for unrelated bone-marrow donor selection.

The HLA-A*02 allele is the most heterogeneous allele at HLA-A locus with 22 different subtypes so far identified. All of these subtype polymorphisms are located in alpha 1 and alpha 2 domains which are responsible for peptide biding and HLA restricted recognition by T-cell receptor. The aim of the present study was to determine the frequency of different HLA-A*02 alleles in 33 healthy unrelated Croatians. HLA-A*02 subtyping has also been retrospectively performed in 2 recipient-unrelated donor pairs and in 4 recipient-HLA phenotypically identical parent pairs. All subjects, previously typed as

Presence of allele alphaLELY in an Amazonian Indian population.

Allele alphaLELY is a low-expression allele of the erythroid spectrin alpha-chain that is characterized by a C --> G mutation at position alpha1857 in exon 40 and a C --> T (nt -12) mutation in intron 45. This second mutation is probably responsible for the partial skipping of exon 46. This exon is essential for the nucleation of the alpha-chains by the beta-chains during erythropoeisis. Although allele alphaLELY remains asymptomatic in both heterozygotes and homozygotes, it enhances the expression of deleterious alpha-alleles that occur and, as such, has clinical importance. In this study, th

Atypical strains of Candida albicans recovered from AIDS patients.

By using multilocus enzyme electrophoresis (MLEE) we have analyzed the genetic diversity encountered among chlamydospore-positive Candida albicans strains. While the type II strains of the former C. stellatoidea were genetically indistinguishable from those of C. albicans, type I strains constituted a distinct subgroup compared with C. albicans strains. Nevertheless, all these strains remained genetically very closely related compared with other species of Candida (e.g. C. tropicalis, C. krusei and C. glabrata). These results corroborate the synonymy between C. stellatoidea and C. albicans. Ch

Alzheimer's disease and apolipoprotein E in Italy.

Recent studies have provided evidence of association of apolipoprotein E (ApoE) epsilon 4 allele and late onset familial and sporadic Alzheimer's disease (AD). Epidemiological studies have established allelic variation at the ApoE locus. We have analyzed the ApoE gene polymorphism in a sample of 416 Italian subjects. Our data confirm a significant association between epsilon 4 allele and sporadic AD. The frequency of epsilon 4 allele in early onset familial AD patients was comparable to control values suggesting that epsilon 4 allele does not represent a risk factor for early onset familial AD

Comparative analysis of HLA polymorphism at the serologic and molecular level in Moroccan and Ashkenazi Jews.

The Jewish people comprise two major groups, one encompassing the Jews of Ashkenazi (Central and Eastern European) origin and the other including those of Sephardic (Middle Eastern and North African) descent. To the latter belong the Jews of Moroccan stock, who form the largest Jewish subgroup among the non-Ashkenazi population living in Israel. As the members of each of these groups differ in physiognomy and life style, it was of interest to investigate whether these differences are also reflected in their respective HLA compositions. To this end, 132 subjects of Ashkenazi and 113 individuals

Science, medicine and phenylketonuria.

Science addresses ignorance; medicine uses facts. The scientific approach to phenylketonuria (PKU) led to the discovery of its causes, both ultimate (allelic heterogeneity at the PAH locus) and proximate (dietary phenylalanine), the proximal phenotype (phenylalanine hydroxylase deficiency), the associated metabolic phenotype and the major distal phenotype (impaired cognitive development and neuropsychological function) for which the pathogenesis is still being investigated. By applying knowledge through newborn screening, early diagnosis and treatment, the brain disease of PKU has been greatly

Immunoglobulin heavy chain variable region polymorphisms and multiple sclerosis susceptibility.

In addition to the major histocompatibility complex (MHC), genetic susceptibility in multiple sclerosis (MS) appears to be influenced by other loci. A recent study has identified a population association with an immunoglobulin heavy chain variable region polymorphism in the VH2-B5 family, with both familial and sporadic MS patients. We have repeated this association study in a second MS patient group and used two ethnically and geographically matched control groups and the MS patients' unaffected sibs for comparisons. The VH2-B5 polymorphism was found to be over-represented in MS patients when

The spectrum of beta thalassaemia in Burma.

The molecular defects causing beta thalassaemia have been analysed in 85 unrelated Burmese patients. The patients included 14 with homozygous beta thalassaemia, 70 with HbE/beta thalassaemia and one with HbS/beta thalassaemia. Using a combination of allele-specific oligoprobe hybridization and direct sequencing of genomic DNA amplified by the polymerase chain reaction, 95/99 of the beta-thalassaemia alleles have been characterized. Six mutations have been identified of which three, the G-T at IVS-1 position 1, the G-C at IVS-1 position 5 and the deletion of TCTT in codons 41/42, accounted for

Genetic factors in sleep disorders.

Several sleep disorders have a genetic basis. These conditions include the narcoleptic syndrome, sleep walking, periodic movements in sleep, circadian delay syndromes and familial insomnia. These disorders illustrate different control mechanisms involved in sleep and wakefulness, including those determining the prevalence and timing of NREM and REM activity, somatomotor inhibition and excitation, autonomic discharge, and the circadian framework of sleep. The genetic defect in narcolepsy has been localised to the short arm of chromosome 6, but the chromosomal localisations of the genetic basis

Glucose-6-phosphate dehydrogenase deficiency and hereditary ovalocytosis in the Ok Tedi impact region of Papua New Guinea.

We report the distribution of two genetic traits, glucose-6-phosphate dehydrogenase (G-6-PD) deficiency and hereditary ovalocytosis (HO) in a number of populations living in the Ok Tedi impact region of Papua New Guinea. Significant interpopulation heterogeneity in the distributions of G-6-PD deficiency and HO was observed. The highlands populations of the region did not show any G-6-PD deficiency, but in the highlands fringe and lowland populations the trait has achieved polymorphic frequencies. Hereditary ovalocytosis is significantly more common in the region and is present in all the popul

Nonrandom distribution of genotypes among red cell indices.

Venous blood samples were obtained from 25,302 healthy adults in Kentucky, USA. The red cell indices measured on these samples were evaluated by multiple stepwise regression analysis to derive an algorithm capable of discriminating the 138 individuals within this population who had genotypes AA, AC, AS or AA beta-thalassemia. The simple discriminant MCV2 x MCH with a cut-off set at 1530 detected 137 out of 138 of the heterozygotes with a false positive rate in this population of 4.4%. Other discriminants tested produced fewer false positives but also missed a sufficient number of heterozygotes

RFLP analysis in families with sporadic hemophilia A. Estimate of the mutation ratio in male and female gametes.

To investigate the sporadic occurrence of hemophilia A and to estimate the sex ratio of mutation rates directly, 17 families with isolated cases of the disorder were studied by RFLP analysis and by clotting assays. Three RFLPs, one intragenic and two with close linkage to hemophilia A, were used. In eight families the RFLP study excluded the carrier status of the maternal grandmothers. Since hemostatic studies showed that the eight mothers of these propositi were hemophilia carriers, the origin of the newly mutated genes was inferred from the RFLP patterns: six hemophilic genes derived from th

Effect of temperature on mutation in cultured human skin fibroblasts.

Because clothing enhances the gonadal temperature in the human male and because as a consequence of that the spontaneous mutation rate might be increased, a study was undertaken to determine the effect of temperature on mutation towards HPRT deficiency in human diploid skin fibroblasts. Culturing of the cells in HAT medium containing azaserine, to remove pre-existing mutants, was highly mutagenic. Some results suggested that azaserine acts as an indirect mutagen. The mutational process in the presence of azaserine was influenced by temperature, a rise in temperature from 33 to 37 degrees C lea

Enzyme polymorphism in Ascaris suum (Nematoda).

The potential of multilocus electrophoretic studies for providing insight into the population biology of parasitic organisms was studied using the swine parasite Ascaris suum suum. Thirty-eight loci encoding enzymatic or nonenzymatic proteins have been resolved in extracts of adult worms by starch-gel electrophoresis. A preliminary study of variation in Ascaris from eastern Iowa revealed an average heterozygosity of 6.6%. Allele frequencies at six polymorphic loci were similar in males and females and genotypic frequencies were in accord with those expected in a single, randomly mating populat

Behçet's disease: lack of correlation of clinical manifestations with HLA antigens.

It has been suggested that certain tissue types are associated with different clinical manifestations of Behçet's disease in Japan and England. Regional differences in the associations between HLA antigens and Behçet's disease have been established. We tissue type 119 patients with Behçet's disease in Istanbul and attempted to correlate various clinical manifestations with HLA antigens. The frequency of only one tissue antigen, HLA-B5, was found to be increased in these series. HLA-B5 was present in 94 of 119 patients (77%) compared to 89 of 268 controls (33%), P less than 0.0001, ARR = 6.79.
