[The programming effects of the placenta on offspring neurobehavioral development].

Neurobehavioral development in offspring is shaped by multiple interacting determinants. As a critical organ during gestation and fetal development, the placenta exerts programming effects on offspring neurobehavioral outcomes through diverse mechanisms. This article systematically examines the programming role of placental factors from the following perspectives: placental barrier function, placental pathological alterations, neuroendocrine pathways, epigenetic regulation, and immunomodulatory mechanisms. In addition, it explores potential mechanisms underlying sex-specific influences on offs…

The POHaD paradigm: role of the placenta in paternal programming.

The Paternal Origins of Health and Disease (POHaD) paradigm emerges from the well-known Developmental Origins of Health and Disease (DOHaD) … th of future generations. The significant role of the paternal genome in the fetal component of the placenta and the recognition of the placenta's involvement in postnatal disease programming underscore the importance of studying the placenta in paternal programming research. The aim of this work is to review what we know so far about paternal programming …

Microplastics and the Placenta: A Call to Action for Perinatal Research.

Microplastics have been detected in human placental and neural tissues, raising urgent concerns about their potential effects on maternal and fetal health. Emerging evidence links microplastics to systemic inflammation, neurotoxicity, and endocrine disruption, yet their impact on pregnancy outcomes and fetal development remains poorly understood. Given the placenta's central role in early-life health, perinatal researchers are uniquely positioned to lead investigations into this environmental threat. We call for collaborative, multidisciplinary research to better understand and mitigate the ef…

A Narrative Review on the Effect of Valproic Acid on the Placenta.

BACKGROUND: Valproic acid (VPA) is an antiepileptic and mood-stabilizing drug with well-established teratogenic risks when taken during pregnancy. While its harmful effects on fetal development are well known, less attention has been given to its impact on placental development and function, despite the placenta's critical role in pregnancy. AIM: This narrative review examines how VPA exposure affects placental growth, morphology, nutrient transport, and epigenetic modifications. It also considers whether placental dysfunction may contribute VPA's teratogenic effects. RESULTS: Evidence sugge…

Maternal probiotic supplementation and effects on the fetal placenta.

Increasing number of pregnant women are consumi ng probiotics to promote their own health and that of their unborn fetuses. Such supplements … by maternal probiotic supplementation through microorganism production of bioactive compounds. The placenta is in direct communication with the underlying uterine tissue. Thus, compounds in the maternal blood can easily transfer across the placenta and impact this hormonally sensitive organ. Select studies suggest that disruptions to the maternal microbio …

Effects of selective serotonin reuptake inhibitors on the placenta†.

The rate of depression in pregnant mothers has dramatically risen in the past few decades. One of the well-studied causes of depression is a … ins active within the synaptic cleft. While such treatments might be beneficial for the mother, the placenta and fetal brain can be inadvertently affected. SERT has been proposed to be the primary means by which the placenta internalizes maternal 5-HT and then transfers this neurotransmitter to the emerging brain, where it guides initial brain development. …

mRNA-1273 is placenta-permeable and immunogenic in the fetus.

COVID-19 mRNA vaccines are generally recognized as safe for gestational administration. However, their transplacental pharmacokinetics remain obscure. In this study, mRNA-1273 intramuscularly given to pregnant mice rapidly circulated in maternal blood and crossed the placenta within 1 h to spread in the fetal circulation. Although spike mRNA in fetal circulation faded away within 4-6 h, it could accumulate in fetal tissues, mainly the liver and get translated into spike protein. Transplacental mRNA-1273 proved immunogenic in the fetuses, as postnatally equipped with anti-spike immunoglobulin (…

Characterization of senescence-associated transcripts in the human placenta.

INTRODUCTION: Fusion of mononucleated cytotrophoblasts into syncytium leads to trophoblast senescence. Yet, premature senescence is associated with preeclampsia, fetal growth restriction (FGR), and related obstetrical syndromes. A set of 28 transcripts that comprise senescence-associated secretory phenotype (SASP) was recently described in placentas from women with preeclampsia. We posited that this transcript set is uniquely regulated in late-term placentas or in placentas derived from participants with major obstetrical syndromes. METHODS: Using our large placental RNAseq bank, we analyzed …

Role for MRI in Secondary Screening for Placenta Accreta.

Aims/Background Rates of both placenta praevia and placenta accreta spectrum (PAS) are rising. There is significant morbidity associated with PAS and antenatal detection reduces morbidity at delivery. In the UK there is a move to centralise care of these cases in dedicated units. Incidental diagnosis is more common, in the absence of a regular screening program. In non-specialist hospitals, there is a need to consider adjunct confirmatory diagnostics prior to referral to specialist centres. This is important to ensure capacity management in specialist centres. Ultrasound (US) and Magnetic Reso…

We hypothesize that the immune status of mothers during pregnancy impacts upon the placenta, in turn affecting infant immunity. We explore this hypothesis by examining data from key studies and clinical cohorts, aiming to 'join the dots' to form a body of knowledge to catalyse development of more comprehensive studies. Arsenic and the placenta: A review with emphasis on the immune system.

Chronic arsenic exposure affects over 140 million people globally. While arsenic easily crosses the placenta, the specific mechanisms impacting placental immune cell populations and fetal health are unclear. Maternal arsenic exposure is epidemiologically linked to increased infection risk, mortality, and cancer susceptibility in offspring, emphasizing the importance of understanding placentally-mediated immune effects. This review explores the potential role of the placenta, a key organ for immune transfer to the developing fetus, in mediating chronic low-dose arsenic exposure effects. Examini…
