Modelling heme-mediated brain injury associated with cerebral malaria in human brain cortical organoids.

Human cerebral malaria (HCM), a severe encephalopathy associated with Plasmodium falciparum infection, has a 20-30% mortality rate and predo … CR3, brain-derived neurotrophic factor (BDNF) and a receptor tyrosine-protein kinase, ERBB4, in the organoids. We then tested the neuroprotective effect of neuregulin-1 (NRG-1) against heme treatment in organoids. Neural stem and mature cells differentially expressed CXCL-10, CXCR3, BDNF and ERBB4 in the developing organoids and in response to heme-induced …

Chronic Chlamydia infection in human organoids increases stemness and promotes age-dependent CpG methylation.

Chronic infections of the fallopian tubes with Chlamydia trachomatis (Ctr) cause scarring and can lead to infertility. Here we use human fallopian tube organoids and genital Ctr serovars D, K and E for long-term in vitro analysis. The epithelial monolayer responds with active expulsion of the bacteria into the lumen and with compensatory cellular proliferation-demonstrating a role of epithelial homeostasis in the defense against this pathogen. In addition, Ctr infection activates LIF signaling, which we find to be an essential regulator of stemness in the organoids. Infected organoids exhibit …

Dynamic Formation of Microvillus Inclusions During Enterocyte Differentiation in Munc18-2-Deficient Intestinal Organoids.

BACKGROUND & AIMS: Microvillus inclusion disease (MVID) is a congenital intestinal malabsorption disorder caused by defective apical vesicular transport. Existing cellular models do not fully recapitulate this heterogeneous pathology. The aim of this study was to characterize 3-dimensional intestinal organoids that continuously generate polarized absorptive cells as an accessible and relevant model to investigate MVID. METHODS: Intestinal organoids from Munc18-2/Stxbp2-null mice that are deficient for apical vesicular transport were subjected to enterocyte-specific differentiation protocols. …

The Notch and Wnt pathways regulate stemness and differentiation in human fallopian tube organoids.

The epithelial lining of the fallopian tube is of critical importance for human reproduction and has been implicated as a site of origin of high-grade serous ovarian cancer. Here we report on the establishment of long-term, stable 3D organoid cultures from human fallopian tubes, indicative of the presence of adult stem cells. We show that single epithelial stem cells in vitro can give rise to differentiated organoids containing ciliated and secretory cells. Continuous growth and differentiation of organoids depend on both Wnt and Notch paracrine signalling. Microarray analysis reveals that inh…

Cytoskeletal and functional changes in bioreactor assembled thyroid tissue organoids exposed to gamma radiation.

Fischer rat thyroid cells were grown under low-shear stress in a bioreactor to a stage of organization composed of integrated follicles resembling small thyroid glands prior to exposure to 3 Gray-gamma radiation. Bioreactor tissues and controls (both irradiated and non-irradiated) were harvested at 24, 48, 96 and 144 hours post-exposure. Tissue samples were fixed and fluorescently labeled for actin and microtubules. Tissues were assessed for changes in cytoskeletal components induced by radiation and quantified by laser scanning cytometry. ELISA's were used to quantify transforming growth fact

Advances in cellular and structural engineering of brain organoids for disease modeling: A Comprehensive Review.

BACKGROUND: Organoid technology has rapidly evolved as a transformative tool for modeling human physiology and disease. Organoids are three-dimensional, stem cell-derived constructs that self-organize to recapitulate key structural and functional features of native organs. Among known organoids, brain organoids have provided unprecedented insights into human neurodevelopment and pathophysiology, overcoming the limitations of traditional two-dimensional cultures and animal models. These models closely mimic the architecture, cellular diversity, and regional specification of the human brain, off…

Construction and application of pancreatic exocrine organoid and spheroid for drug screening and precision medicine.

The incidence of pancreatic exocrine disorders, particularly pancreatic cancer, has been steadily rising. However, treatment options remain limited, with substantial interindividual variability in therapeutic efficacy. This clinical challenge has accelerated the development of advanced three-dimensional (3D) modeling systems, with patient-derived organoids and multicellular spheroids emerging as transformative tools that faithfully recapitulate tumor pathophysiology. In contrast to 2D cultures, which fail to recapitulate the three-dimensional spatial architecture and cell-cell interactions fou…

Advancing Microplastic and Nanoplastic Toxicity Assessment: Insights from Human Organoid Models.

Evidence has demonstrated that microplastics (MPs) and nanoplastics (NPs) exerted toxic effects on animal models; our understanding of their potential toxicity to human health remains limited due to the lack of human-relevant in vitro models. Human stem cell-derived organoids are sophisticated and multicellular structures that could effectively mimic the key features of native organs and tissues. To date, human organoids have been widely used in toxicology. This study reviews the current application of human organoids in the toxicity assessment of MPs and NPs. Current limitations and future pe…

Adaptation of Enterococcus faecalis to intestinal mucus revealed by a human colonic organoid model.

UNLABELLED: The human gastrointestinal tract hosts a diverse population of microorganisms that have a significant impact on host health. Among this population, Enterococcus faecalis (Ef) represents a common member of intestinal microbiota, colonizing humans early in life but also capable of opportunistically infecting its host. Despite its importance in human health, investigations of its physiological adaptation to the mucosal environment remain limited. Building on recent advances in tissue engineering, we here leverage human colonic organoids (colonoids) to investigate the Ef's mechanisms o…

Structure-Property-Function Relationships in Stimuli-Responsive Hydrogels for Brain Organoid Vascularization.

Human induced pluripotent stem cell (iPSC)-derived brain organoids have emerged as powerful three-dimensional (3D) platforms for modeling human neurodevelopment and neurological disorders. However, the absence of a functional vascular network remains a critical limitation, restricting oxygen and nutrient delivery, impairing metabolic stability, and constraining long-term maturation. Conventional extracellular matrix (ECM) mimetics, such as Matrigel and other static synthetic hydrogels, provide biochemical support but fail to recapitulate the dynamic remodeling that characterizes the developing…

Generation of a Mouse Prostate Organoid-Based Model for Studying Host-Pathogen Interactions.

Aging men are highly affected by prostate diseases, such as benign prostatic hyperplasia, prostatitis, and prostate cancer. Together, these conditions contribute significantly to the global health burden and are a leading cause of illness in men. Organoid models have transformed disease modeling by allowing the replication of key features of epithelial structure and function in vitro. However, the closed lumen structure of organoids limits experimental access to the apical surface, hindering studies of host-pathogen interactions. To overcome these challenges, we generated a physiologically rel…

The rise of bone marrow organoids as next-generation models for blood formation and failure.

Bone marrow organoids (BMOs) are three-dimensional (3D) cell culture models that recapitulate key structural and functional features of the bone marrow (BM) niche. BMOs offer important advantages in hematopoietic research by modeling key aspects of human hematopoiesis compared with classical in vitro 2D and 3D cellular models including bioreactors, BM-on-a-chip platforms, 2D models or BM ossicles by better recreating the 3D architecture, cellular heterogeneity, and spatial organization of the BM microenvironment. They offer a scalable and cost-effective alternative to animal models and reduce …

An organoid-guided roadmap for precision delivery of epigallocatechin gallate in oral submucous fibrosis.

Oral submucous fibrosis is a chronic, exposure linked mucosal scarring disorder that restricts mouth opening and increases cancer risk. Epigallocatechin gallate offers convergent antifibrotic actions across TGF-β signalling, oxidative stress, inflammation and collagen remodelling, yet clinical effects remain inconsistent because instability and rapid clearance prevent sustained intralesional exposure. This Review proposes a precision delivery roadmap that couples engineered local dosage forms and nanocarriers with patient-derived organoids as a functional translational layer. We describe how o…

Organoid transplantation in the adult endometrium restores fertility and uncovers epithelial lineage plasticity.

The adult endometrium regenerates repeatedly, yet the cells and mechanisms that rebuild its epithelium remain poorly defined. To control the cell types available for regeneration, a genetic model to extensively ablate the uterine epithelium was combined with transplantation of lineage-labeled organoids. Ablation without organoid transplantation triggered re-epithelialization, but resulted in infertility. Transplanted endometrial epithelial organoids engrafted into the ablated uterus, reconstructed both the luminal and glandular epithelia, and restored fertility. Depleting organoids of the glan…

Host-virus determinants of Ebola virus persistence in a human cerebral organoid model.

Ebola virus (EBOV) causes Ebola virus disease (EVD), a multisystemic human disease associated with an extraordinarily high case-fatality rate. EVD survivors may experience recrudescent inflammation with viral persistence in immune-privileged tissues, including the central nervous system (CNS). Persistence, defined by ongoing replication of the EBOV genome beyond the acute disease phase, may lead to virion production. Productive persistence has been linked to re-initiation of EVD outbreaks. We developed a human cerebral organoid model to investigate the host and viral determinants of EBOV CNS p
