A comparison of indirect immunofluorescence and electron microscopy for the diagnosis of some haemorrhagic viruses in cell cultures.

Yellow fever, dengue (types 1, 2 and 4), Chikungunya, Rift Valley fever, Ebola, Marburg, and Lassa viruses were inoculated into susceptible cell cultures and daily investigated by indirect immunofluorescence (IFA) and electron microscopy (EM) with a view to achieve an early detection-identification of these agents. Compared to the other cell lines tested (Vero, BHK-21 and Aedes albopictus), CV-1 cells were found to be more sensitive. Viral antigens were detected by IFA from a few hours post inoculation (CHIK and RVF) to a maximum of 3 days (YF and EBO). For most of the viruses studied, the cyt

Scanning electron microscopy of the wall of the third ventricle in the brain of Rana temporaria. Part IV.

The surface specializations of the wall of the third cerebral ventricle of Rana temporaria were investigated with the scanning electron microscope. These specializations can be divided into three types: cilia, large bulbous protrusions, and microvillus-like protrusions. Most parts of the ventricular surface are densely ciliated. In contrast, other regions are either scantily ciliated or devoid of cilia. Four areas of the ventricular surface are studded with numerous large bulbous protrusions. These large protrusions can be divided into two types: One type consists of intraventricular end bulbs

[Angio-architecture of the colon in Crohn disease and ulcerative colitis. Light microscopy and scanning electron microscopy studies with reference to the morphology of the healthy large intestine].

The etiology and the pathogenesis of the chronic inflammatory bowel diseases known as Crohn's disease and ulcerative colitis have not been defined. Therefore, in this study the main emphasis was placed on description of the pathologic anatomy. Disturbed blood supply and vascular disorders have been discussed as etiopathogenetic factors. The results in the literature are frequently contradictory. For this reason, the vascular system of the colon in Crohn's disease and ulcerative colitis was systematically examined by means of various morphological methods in this study. Microvascular corrosion

Sample preparation of some plant tissues for transmission electron microscopy using high pressure freezing and freeze substitution.

For sample preparation of biological block sectioning, high pressure freezing and freeze substitution (HPF-FS) is a better choice than chemical fixation. Freezing fixation can retain the original structures and preserve the soluble materials, and avoid the disadvantage of chemical fixation. Using high-pressure freezing, plant tissues are more difficult to freeze successfully than animal tissues and microbes because they have cell walls and varying degrees of vacuolation. For plant tissues, including the roots of Arabidopsis thaliana, 1-hexadecene is an ideal cryoprotectant/filler. The ability

Mechanism of beta-arrestin 1 mediated Src activation via Src SH3 domain revealed by cryo-electron microscopy.

Beta-arrestins (βarrs) are key regulators and transducers of G-protein coupled receptor signaling; however, little is known of how βarrs communicate with their downstream effectors. Here, we delineate structural mechanisms underlying βarr-mediated signal transduction. Using cryo-electron microscopy, we elucidate how βarr1 recruits and activates the non-receptor tyrosine kinase Src, a well-established signaling partner of βarrs. βarr1 engages Src SH3 through two distinct sites, each employing a different recognition mechanism: a polyproline motif in the N-domain and a non-proline-based interact

Exploring placental ultrastructure: A review of electron microscopy techniques and emerging methods for resolving 3D organelle architecture.

Trophoblast cells line the surface of placental villi, facilitating the exchange of nutrients, gases, and wastes between the maternal and fetal circulations. The fusion of cytotrophoblast (CTB) cells into the surrounding multinucleated syncytiotrophoblast (STB), is accompanied by a shift in cellular ultrastructure (subcellular architecture). Mitochondria undergo a remarkable decrease in size and alteration in morphology following trophoblast differentiation, and have thus been the subject of investigations due to their crucial role in producing energy for placental development. Observing this

A block staining method using ethanolic phosphotungstic acid for the visualisation of collagens in transmission electron microscopy.

Conventional transmission electron microscopic imaging of biological samples requires contrast enhancement by staining with heavy metal salts. The most widely used stains are uranyl acetate and its non-radioactive lanthanoid replacements, and lead citrate. However, these substances proved of limited use for the visualisation of small fibrillar collagens. We therefore developed a preparation of ethanolic phosphotungstic acid (E-PTA) as an improvement to overcome this deficiency. We were able to establish a highly effective and time saving block staining procedure that can be integrated in the d

Effects of quercetin in male rats' testis after consumption of permethrin: a transmission electron microscopy (TEM) study.

Permethrin, a widely used pyrethroid insecticide, impairs male reproductive function through oxidative stress and endocrine disruption. This study evaluated the protective effects of quercetin against permethrin-induced testicular toxicity in adult male Wistar rats. Thirty rats were divided into five groups (n = 6): control, DMSO vehicle, permethrin (35 mg/kg/day orally), permethrin + quercetin (15 mg/kg/day orally), and quercetin alone, for 60 days. Permethrin exposure induced severe ultrastructural damage including mitochondrial swelling, nuclear chromatin condensation, and Sertoli cell deta

Visual Adaptations of the Dicentrarchus punctatus: New Insights Into Its Retina Adaptations With Transmission Electron Microscopy Analysis.

This study was designed to illustrate the light and electron microscopic characterisations of the eye of Dicentrarchus punctatus with new insight into transmission electron microscopic features of the retina. The eye is composed of three layers: the external sclera-cornea layer, the middle layer, and the internal layer. The cornea has three laminae: stratified epithelial cuboidal cells anteriorly, Bowman (middle), and the dermato-stromal lamina posteriorly. The mucoidal lamina was observed between the cranial part of the sclera-corneal and the derma-stromal, and established by the fibre bundle

Largescale semithin sectioning method supported by a stainless steel ring enables multiscale analysis in transmission electron microscopy.

In transmission electron microscopy, semi-thin sections are prepared and stained with 2% toluidine blue to assess sample integrity and accurately identify regions of interest under light microscopy prior to ultrathin sectioning. This procedure can be challenging for large specimens, especially in attaching semi-thin sections to glass slides without wrinkles. Consequently, semi-thin sections are limited to lengths between 0.2 and 2.0 mm along their longest side, which reduces the amount of structural detail that can be observed. To address this limitation, we have implemented a specialized stai

Ex situ and in situ demonstration of amyloid fibrils for confirmation of amyloidosis using transmission electron microscopy.

Confirmation of extracellular amyloid deposition across various animal species and tissue types has been a long-standing challenge in veterinary diagnostic pathology. Transmission electron microscopy (TEM) has historically been used to advance the understanding of amyloid fibril morphology and confirm amyloid fibril deposition when histologic methods provide unclear results. We assessed the feasibility of utilizing TEM for routine confirmation of amyloidosis as an addition to histology. We analyzed ex situ amyloid fibrils with direct, negative-contrast TEM and in situ amyloid fibrils with alde

Real-time visualisation of fast nanoscale processes during liquid reagent mixing by liquid cell transmission electron microscopy.

Liquid cell transmission electron microscopy (LCTEM) is a powerful technique for investigating crystallisation dynamics with nanometre spatial resolution. However, probing phenomena occurring in liquids while mixing two precursor solutions has proven extremely challenging, requiring sophisticated liquid cell designs. Here, we demonstrate that introducing and withdrawing solvents in sequence makes it possible to maintain optimal imaging conditions while mixing liquids in a commercial liquid cell. We succeeded in visualising a fast nanoscale crystallisation mechanism when an organic molecule of
