Alveolar macrophage procoagulant activity is increased in acute hyperoxic lung injury.

Alveolar macrophage accumulation and interstitial fibrin deposition are prominent in adult respiratory distress syndrome and chronic interst … lammatory cells was quantified by bronchoalveolar lavage. Pulmonary injury, manifested by increased capillary permeability, developed progressively during exposure to hyperoxia and was associated with significant augmentation of the procoagulant activity of alveolar macrophages early in the disease. This increase preceded the accumulation of polymorphonuclear leukocytes …

Albumin lung/heart ratio change. A simple clinical means of documenting increased pulmonary endothelial permeability to protein.

We investigated a simple method that can be used at the bedside for documenting the net accumulation of albumin in the lung. The technique employs measurement with a computer-linked gamma camera of the activity ratio in an area of the right lung compared with the same-sized area in the heart at 20 minutes and three hours following intravenous injection of technetium Tc 99m albumin. We applied this measurement to three groups of patients: a control group and patients with roentgenographic evidence of edema classified according to clinically available criteria as either hydrostatic edema or perm

Measurements of pulmonary vascular permeability with PET and gallium-68 transferrin.

We quantified pulmonary vascular permeability with positron emission tomography (PET) and gallium-68-(68Ga) labeled transferrin. Six dogs with oleic acid-induced lung injury confined to the left lower lobe, two normal human volunteers, and two patients with the adult respiratory distress syndrome (ARDS) were evaluated. Lung tissue-activity measurements were obtained from sequential 1-5 min PET scans collected over 60 min, after in vivo labeling of transferrin through intravenous administration of [68Ga]citrate. Blood-activity measurements were measured from simultaneously obtained peripheral b

[Anti-inflammatory, analgesic and anti-pyretic activities of a new anti-inflammatory compound, 2-[4-(3-methyl-2-butenyl)phenyl] propionic acid (TA), in experimental animals].

Anti-inflammatory, analgesic and anti-pyretic activities of orally administered TA were investigated in experimental animals. Against acetic acid-induced vascular permeability in mice, carrageenin-induced hind paw edema in rats and ultra-violet ray-induced erythema in guinea pigs, TA produced a dose related inhibition at doses of 40-160 mg/kg, 10-40 mg/kg and 10-40 mg/kg, respectively. TA produced no inhibition against histamine-induced vascular permeability even at a dose of 200 mg/kg in rats. Cotton pellet-induced granuloma and adjuvant-induced arthritis in rats were significantly inhibited

Inhibition of the vascular actions of IgG aggregates by BN 52021, a highly specific antagonist of paf-acether.

The effect of BN 52021, a selective antagonist of paf-acether (Braquet GB patent 8, 418, 424 July 19, 1984), was studied in normotensive rats challenged with different doses of paf-acether. Sudden death was observed in animals receiving an i.v. dose of 10 micrograms/kg of paf-acether and this was prevented by prior treatment with BN 52021 (5 mg/kg, i.v.). Animals receiving 2.5 micrograms/kg of paf-acether had a fall of mean arterial pressure of 92.5 +/- 4.7 mmHg which recovered to the prechallenge level 20.5 +/- 0.2 min thereafter. Previous treatment with BN 52021 (5 mg/kg, i.v.) reduced the m

Vasogenic edema with intraparenchymatous expanding mass lesions: a theory on its pathophysiology and mode of action of hyperventilation and corticosteroids.

Vasogenic edema with expanding mass brain lesions is hypothesized to be due to an increased intracapillary pressure. The latter may be due to preferential occlusion of the venous system by the growth of the lesion but endothelila proliferation and biogenic amines may also play a part. Endocytosis appears to be a mechanical response to the increased intraluminal pressure. This is a poorly selective process which can explain the proteinaceous nature of vasogenic edema. Steroids may act by forming hydrophobic bonds in the endothelial cell membrane and making it more difficult for any membrane fis

Reduced oncotic pressure after shock: a proposed mechanism.

Hemorrhagic shock plus resuscitation leads to expansion of both interstitial fluid space (IFS) and intracellular fluid (ICF). THe IFS expansion is thought to reflect reduced serum colloid oncotic pressure (COP) from capillary leak of serum albumin (SA). This hypothesis was analyzed in 138 injured patients who received an average of 13.6 units of blood and 10.7 L of saline solution for correction of shock. Plasma volume (PV), extracellular fluid (ECF) by inulin space technique, SA level, percent albumin leak (PAL), total intravascular albumin content (TIAC), COP, IFS, weight gain, and time of i

Quantitation of phototoxic hyperemia and permeability to protein: II. Inhibition by histamine (H1 and H2) receptor antagonists in mouse skin.

Exposure of anthracene treated skin to long-wave ultraviolet radiation (UVA) will produce hyperemia, increased vascular permeability, burning and itching. This photosensitized inflammation was produced in the skin of hairless mice by the topical application of a 0.025% (w/v) solution of anthracene followed by exposure to UVA. The resultant hyperemia was quantified by the increase in 51Cr-erythrocyte content (cpm/wet weight) of skin. After 1000 sec (1.3 x 10(4)J/M2) of UVA, the blood content of the skin had increased nine-fold. Pyrilamine (H1 antagonist), administered orally, did not inhibit th

Properties and production characteristics of vomiting, diarrheal, and necrotizing toxins of Bacillus cereus.

Evidence is provided that the enterotoxin of Bacillus cereus variously described in the literature as diarrheagenic toxin, diarrheal agent, fluid accumulation factor, vascular permeability factor, dermonecrotic toxin, and intestinonecrotic toxin is a single relatively unstable protein of molecular weight approximately 50,000 and isoelectric point of the order of 4.9. It is presumed to be the enterotoxin responsible for the diarrheal-type B. cereus food poisoning syndrome and it may also be the pyogenic and pyrogenic factor in nongastrointestinal B. cereus infections of man and animals. The ent

Cardiopulmonary effects of intravenous fluid overload: radiologic manifestations.

Large volumes of normal saline were infused intravenously in 6 dogs until obvious pulmonary edema was observed radiographically. Following volume overload, statistically significant increases occurred in the size of the heart, left atrium, pulmonary arteries and veins, and systemic veins, without the development of congestive heart failure (CHF). The left ventricular end-diastolic pressure remained normal, and cardiac output and stroke volume increased. The results suggest that, in the absence of left ventricular failure, acute volume overload may simulate the radiographic changes produced by

"Pseudocardiogenic" pulmonary edema.

Fifteen postoperative surgical patients, in whom noncardiac pulmonary edema developed were studied. A presumptive diagnosis of left ventricle failure would have been based on historical evidence of heart disease (80%), electrocardiographic changes of ischemia or arrythmia (87%), or cardiogenic shock (20%). (see article) Fig. 6. PAEDP-PCW gradient. Note that arterial oxygen tension had an inverse relationship to this pressure differential. Roentgenographic findings included pulmonary edema (73%), pulmonary vascular congestion (60%), cardiomegaly or congestive heart failure (40%). Mean increase

Vascular permeability to horseradish peroxidase in brainstem lesions of thiamine-deficient rats.

In the early brainstem lesions of acute dietary thiamine deficiency in rats, an outstanding feature is the occurrence of edema. This study, using horseradish peroxidase as a marker, confirms and extends our previous observations using fluorescent dye-labeled albumin that vascular permeability to proteins remains essentially intact during this phase, and becomes altered only when necrosis and hemorrhage supervene. In addition, however, although a minor degree of pinocytotic transendothelial transport and pericytic uptake of horseradish peroxidase was found in both normal animals and in the earl

The effects of histamine in malaria.

1. Extracts of the blood of monkeys (Macaca mulatta) infected with Plasmodium knowlesi contain histamine. A mean total concentration of 0.15 mug/ml was present in the circulating blood.2. No histamine was detected in the blood of healthy monkeys.3. Vasodilatation and increased vascular permeability was observed at the site of injection of the extract into the skin and brain of guinea-pigs.4. Infiltration of the dermal layer by leucocytes was observed after intradermal injection of the histamine extract. A similar response was obtained in the brain.5. The extract of the blood from the infected

Microvascular barrier - Transdermal and Topical drug delivery in Raynaud's Phenomenon.

Raynaud Phenomenon (RP) is a peripheral vascular disease that results in episodes of vasospasm of the microcirculation of hands and feet, which causes ischemia, pain and functional deficiency. Traditional systemic treatments of RP, such as calcium channel blockers and phosphodiesterase type-5 inhibitors, often have limitations due to perfusion-rate-based drug delivery, lack of consistency in local effect and dose-related adverse effects. Microvascular dysfunction plays a central role in determining drug distribution and therapeutic response in Raynaud's Phenomenon. These considerations have le

Faricimab Reverts VEGF-A165-Induced Impairment of the Barrier Formed by Retinal Endothelial Cells.

VEGF-A165-induced persistent dysfunction of the barrier formed by immortalized bovine retinal endothelial cells (iBREC) is only transiently reverted by inhibition of VEGF-A-driven signaling. As angiopoietin-2 (Ang-2) enhances the detrimental action of VEGF-A165, we studied if binding of both growth factors by the bi-specific antibody faricimab sustainably reverts barrier impairment. Confluent monolayers of iBREC were exposed to VEGF-A165 for one day before 10-1000 µg/mL faricimab was added for additional five days. To assess barrier function, we performed continuous electric cell-substrate imp
