BACKGROUND: Long noncoding RNAs (lncRNAs) have recently received wide attention as key molecules that mediate a variety of physiological and pathological processes by regulating gene expression; however, knowledge of lncRNAs in rheumatoid arthritis (RA) is limited. Thus, we investigated the lncRNA expression profile in fibroblast-like synoviocytes (FLSs) from patients with RA and explored the function of abundantly expressed lncRNAs.
METHODS: LncRNA and mRNA microarrays were performed to identify differentially expressed lncRNAs in RA FLSs compared with normal FLSs. Quantitative polymerase ch

Rheumatoid Arthritis (RA) associated osteopenia is a resultant of multifactorial secondary osteoporosis, including disuse-atrophy, glucocorticoid-induced osteopenia, and RA specific activation of osteolysis. In addition, primary osteoporosis, i.e. postmenopausal and senile osteoporosis may overlap with them. Pursuits for the genetic factors for the causes of RA associated osteopenia could be clarified both from the characterization of RA associated factors and the osteoporosis associated factors. Recent progress in systematic genome-wide analysis for the association studies with multiple gene

BACKGROUND: Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by synovial hyperplasia, oxidative stress, and dysregulated immune responses that culminate in progressive joint destruction. Fibroblast-like synoviocytes (FLS) acquire an aggressive, invasive phenotype, while peripheral blood mononuclear cells (PBMCs) perpetuate inflammatory responses. 6-Bromoindirubin-3′-oxime (6BIO) has been shown to modulate signalling pathways associated with inflammation, oxidative stress, and immune cell activation, processes that are critically dysregulated in RA. This study evaluates

OBJECTIVE: To study the role of HLA class II genes in conferring susceptibility to seronegative vs seropositive rheumatoid arthritis (RA).
METHODS: The distribution of DRB1 and DPB1 alleles in 33 seronegative, 20 weakly seropositive, 79 distinctly seropositive patients with RA, and in 181 healthy controls was determined by genomic typing: IgM rheumatoid factor (RF) seronegativity was ascertained in Waalers' latex, and ELISA tests.
RESULTS: DRB1*0101/2 (encoding variants of DR1) was associated with weakly seropositive RA (and possibly seronegative RA) but not the clearly seropositive disease.

Rheumatoid arthritis (RA) is the most common form of chronic inflammatory arthritis characterized by pain, swelling and destruction of joints, with a resultant disability. Disease-modifying anti-rheumatic drugs (DMARDs) and biological drugs can interfere with the disease process. In this study, the effect of β-d-mannuronic acid (M2000) as a novel non-steroidal anti-inflammatory drug (NSAID) with immunosuppressive and anti-inflammatory effects together with antioxidant effects was evaluated on IL17, RORγt, IL4 and GATA3 gene expression in 12 RA patients. Previously, M2000 driven from sodium alg

Brincidofovir (BCV, commercially known as Tembexa®) is an antiviral drug with broad-spectrum activity against double-stranded DNA viruses, including orthopoxviruses. BCV was developed and approved for treating human smallpox in adults and children, including neonates, under the FDA Animal Rule. The efficacy data for licensure were obtained using two surrogate models of smallpox: the rabbitpox virus (RPXV) in New Zealand White rabbits and ectromelia virus (ECTV) in Balb/c mice. The potential for BCV resistance development was evaluated in RPXV- and ECTV-infected animals treated with BCV from pi

Radioactive iodine (RAI) treatment is an established therapeutic tool for 'differentiated thyroid cancers'. The therapeutic effectiveness is linked to the preservation of the iodine-concentrating ability of the neoplastic tissue, a unique, inherent quality of a normal thyroid gland. Iodine concentration is a function involving the expression of transport proteins and organification. Thyroid differentiation score (TDS) is an integrated quantity, first introduced by The Cancer Genome Atlas (TCGA), conveying the relative expression of proteins involved in histogenesis, morphologic and functional

As one of the most common air pollutants, fine particulate matter (PM2.5) increases the risk of diseases in various systems, including the urinary system. In the present study, we exposed male and female C57BL/6J mice to PM2.5 for 8 weeks. Examination of renal function indices, including creatinine (CRE), blood urea nitrogen (BUN), uric acid (UA), and urinary microalbumin, indicated that the kidneys of female mice, not male mice, underwent early renal injury, exhibiting glomerular hyperfiltration. Meanwhile, pathological staining showed that the kidneys of female mice exhibited enlarged glomer

Primary oral mucosal melanoma is an extremely rare and aggressive tumor arising from melanocytes located in the mucosal epithelium of the oral cavity. Although malignant melanoma of oral mucosa shares some clinical features with its cutaneous counterpart, it has been associated with a worst prognosis; its etiopathogenesis are still only partially unraveled as there is no influence of UV radiation. It is known that the mitogen-activated protein kinase pathway mediates cellular responses to growth signals and its activation is an important phenomenon in melanoma. The aim of this study was to eva

Genetic alterations, including mutations and copy-number alterations, are central to the pathogenesis of myelodysplastic syndromes and related diseases (myelodysplasia), but their roles in allogeneic stem cell transplantation have not fully been studied in a large cohort of patients. We enrolled 797 patients who had been diagnosed with myelodysplasia at initial presentation and received transplantation via the Japan Marrow Donor Program. Targeted-capture sequencing was performed to identify mutations in 69 genes, together with copy-number alterations, whose effects on transplantation outcomes

While restoration of ACE2 activity in the pancreas leads to improvement of glycemia in experimental models of Type 2 diabetes, global deficiency in ACE2 disrupts β-cell function and impairs glucose tolerance in mice, demonstrating the physiological role of ACE2 in glucose homeostasis. Although the contribution of pancreatic ACE2 to glucose regulation has been demonstrated in genetic models of diabetes and in models with overexpression of the renin-angiotensin system (RAS), it is unclear whether islet ACE2 is involved in glycemic control in common models of human Type 2 diabetes. BACKGROUND: Rheumatoid arthritis (RA) is a complex autoimmune disease characterized by clinical and molecular heterogeneity, notably in the presence of anti-cyclic citrullinated peptide antibodies (CCP). CCP positivity in RA patients is associated with more severe disease progression and distinct responses to treatment compared to CCP- patients. Although previous studies have investigated cellular and molecular differences between these RA subtypes, there has been limited exploration of their genetic differences at a systems scale, taking into account underlying molecular networks.

BACKGROUND AND AIMS: The association between rheumatoid arthritis (RA) and blood lipid levels has often been described as paradoxical, despite the strong association between RA and cardiovascular disease (CVD) risk. We aimed to clarify the genetic architecture that would explain the relationship between RA and blood-lipid levels, while considering inflammation as measured by C-reactive protein (CRP).
METHODS: Genome-wide association study (GWAS) summary statistics were collected from the CHARGE Consortium and Global Lipids Genetics Consortium. Blood-lipid levels includes HDL-C, LDL-C, triglyc

Rheumatoid arthritis (RA) is an autoimmune disease characterized by inflammation and pro-inflammatory cytokines production. IL-1Ra is an anti-inflammatory cytokine codified by IL1RN gene that blocks IL-1 signalling. A VNTR polymorphism of 86 bp in IL1RN gene has been associated with RA risk and regulation of IL-1Ra expression. In this study, we determined mRNA and protein expression of IL-1Ra in RA patients and control subjects (CS). This study included 85 RA patients classified according to the ACR/EULAR 2010 criteria and 67 CS. Polymerase chain reaction was used to identify IL1RN VNTR polymo

Difenoconazole (DFZ) is a broad-spectrum triazole fungicide that is widely utilized in agriculture. Although DFZ has been demonstrated to induce reproductive toxicity in aquatic species, its toxic effects on the mammalian reproductive system have yet to be fully elucidated. In vivo, male mice were administered 0, 20 or 40 mg/kg/d of DFZ via oral gavage for 35 days. Consequently, DFZ significantly decreased testicular organ coefficient, sperm count and testosterone levels, augmented sperm malformation rates, and elicited histopathological alterations in testes. TUNEL assay showed increased apop

Nutritional status including vitamin A could explain some of the developmental deformities observed in cultivated teleosts, including Atlantic cod (Gadus morhua). In the present study we aimed to investigate the transcriptional effect of retinoic acid (RA) on bone related genes using Atlantic cod craniofacial explants tissue cultures. Two different osteoblast specific osteocalcin/bone gla protein isoforms were discovered in cod. Transcription of both isoforms was up-regulated following RA treatment of 65 dph cod lower jaw explants. In contrast, transcripts coding for genes related to bone reso

Nitric oxide has been described as a trigger for the synthesis of proinflammatory mediators and as a cytotoxic molecule with a pivotal role in apoptosis at the joints of rheumatoid arthritis (RA) patients. Polymorphisms in the NOS2A gene, which codes for the inducible nitric oxide synthase [(i)NOS], have been tested for association with several autoimmune diseases such as Crohn's disease or type 1 diabetes. Moreover, the existence of correlated levels of (i)NOS protein and synovial cell apoptosis in RA patients, pointed to NOS2A as a good candidate gene involved in RA predisposition. The role
