BACKGROUND: Heme-incorporating peroxygenases are responsible for electron transport in a multitude of organisms. Yet their application in biocatalysis is hindered due to their challenging recombinant production. Previous studies suggest Komagataella phaffi to be a suitable production host for heme-containing enzymes. In addition, co-expression of helper proteins has been shown to aid protein folding in yeast. In order to facilitate recombinant protein expression for an unspecific peroxygenase (AnoUPO), we aimed to apply a bi-directionalized expression strategy with Komagataella phaffii.

Split transketolases (split TKs) are an underexplored class of carboligases with potential for application in biocatalysis. Here, we investigate ten split TKs and reveal substantial functional heterogeneity, with marked differences in substrate scope, activity, enantioselectivity, and robustness. The enzymes accepted diverse acceptors, aliphatic and aromatic, while retaining the canonical (S)-selectivity of full-length TKs. Pyruvate acceptance further revealed distinct catalytic behaviours, including cross-coupling and self-coupling specificity. Selected enzymes also displayed high organic sol

The asymmetric construction of highly congested polycyclic architectures remains a formidable challenge in chemical synthesis. Biocatalysis offers exquisite stereocontrol, yet the repertoire of enzyme-catalyzed cycloadditions remains notably limited. Here, we elucidate the intriguing oxidative [3 + 2] cycloaddition capability of a flavoenzyme CpaO and develop it as a robust biocatalyst for asymmetric construction of the pentacyclic scaffold of (-)-α-Cyclopiazonic acid ((-)-α-CPA), a nanomolar inhibitor of sarco/endoplasmic reticulum calcium(II)-dependent ATPase. This work presents not only the

Imine reductases (IREDs) provide promising opportunities for the synthesis of various chiral amines. Initially, asymmetric imine reduction was reported, followed by reductive aminations of aldehydes and ketones via imines. Herein we present the reductive amination of structurally diverse carbonyls and dicarbonyls with hydrazines (reductive hydrazination), catalyzed by the IRED from Myxococcus stipitatus. In analogy to IRED-catalyzed reductive aminations, various carbonyls and dicarbonyls could react with simple hydrazines to produce substituted acyclic and cyclic N-alkylhydrazines. By incorpor

Enzymatic carbon-phosphorus bond formation is extremely rare in nature, limiting biocatalytic access to phosphorus-containing compounds that are widely used in pharmaceuticals and agrochemicals. Here, we report an asymmetric enzymatic hydrophosphorylation through oxygen activation using a repurposed flavin-dependent enzyme. Mechanistic studies revealed that reactive oxygen species are converted into productive phosphorus-centered radicals, followed by radical addition and enzymatic hydrogen atom transfer, achieving high enantioselectivity. The enzyme accommodates diverse phosphorus-hydrogen do

Carboligation reactions are highly valuable transformations in modern applied biocatalysis. Tertiary α-hydroxy ketones have broad relevance, yet their synthesis remains challenging. This 1,2-functionality can be introduced by thiamine diphosphate (ThDP)-dependent enzymes. However, most ThDP-dependent enzymes form new C-C bonds using aldehydes as acceptor substrates, yielding secondary α-hydroxy ketones. Only a few enzymes accept ketones as substrates, enabling access to chiral tertiary alcohols. Here, we describe a bioinformatic strategy to identify ThDP-dependent enzymes producing tertiary al

Polyketide synthases (PKSs) usually employ a ketoreductase (KR) to catalyze the reduction of a β-keto group, followed by a dehydratase (DH) that drives the dehydration to form a double bond between the α- and β-carbon atoms. Herein, a DH*-KR* involved in FR901464 biosynthesis was characterized: DH* acts on glyceryl-S-acyl carrier protein (ACP) to yield ACP-linked pyruvate; subsequently KR* reduces α-ketone that yields L-lactyl-S-ACP as starter unit for polyketide biosynthesis. Genetic and biochemical evidence was found to support a similar pathway that is involved in the biosynthesis of lankac

Glucose-6-phosphate dehydrogenase (G6PD) is the first enzyme in the pentose phosphate pathway. It has been extensively studied by biochemical and structural techniques. 13 X-ray crystal structures and five electron cryo-microscopy structures in the PDB are focused on in this topical review. Two F420-dependent glucose-6-phosphate dehydrogenase (FGD) structures are also reported. The significant differences between human and parasite G6PDs can be exploited to find selective drugs against infections such as malaria and leishmaniasis. Furthermore, G6PD is a prognostic marker in several cancer type

Escherichia coli strain PC09 (DeltaxylB, cAMP-independent CRP (crp*) mutant) expressing an NADPH-dependent xylose reductase from Candida boidinii (CbXR) was previously reported to produce xylitol from xylose while metabolizing glucose [Cirino et al. (2006) Biotechnol Bioeng 95(6): 1167-1176]. This study aims to understand the role of NADPH supply in xylitol yield and the contribution of key central carbon metabolism enzymes toward xylitol production. Studies in which the expression of CbXR or a xylose transporter was increased suggest that enzyme activity and xylose transport are not limiting

Formaldehyde is a toxic metabolite generated in the bacterial catabolism of lignin-derived aromatic compounds (LDACs) through the oxidative O-demethylation of methoxylated aromatics. Microbial cell factories designed to convert LDACs to commodity chemicals should ideally utilize formaldehyde to improve carbon efficiency and strain fitness. Methylotrophs and natural degraders of LDACs have evolved various strategies for utilizing formaldehyde. These range from cofactor-dependent detoxification systems that extract reducing equivalents to cyclical pathways that synthesize building blocks. In add

Alcohol dehydrogenases (ADHs) mediated biocatalytic asymmetric reduction of ketones have been widely applied in the synthesis of optically active secondary alcohols with highly reactive hydroxyl groups ligated to the stereogenic carbon and divided into (R)- and (S)-configurations. Stereocomplementary ADHs could be applied in the synthesis of both enantiomers and are increasingly accepted as the "first of choice" in green chemistry due to the high atomic economy, low environmental factor, 100 % theoretical yield, and high environmentally friendliness. Due to the equal importance of complementar

A biocatalytic approach for the synthesis of metaxalone and its analogues was developed based on the reaction of epoxides and cyanate catalyzed by halohydrin dehalogenase. Gram-scale synthesis of chiral and racemic metaxalone was achieved with 44% (98% ee) and 81% yields, respectively, by protein engineering of the halohydrin dehalogenase HHDHamb from Acidimicrobiia bacterium. Additionally, various metaxalone analogues were synthesized at 28-40% yields (90-99% ee) for chiral forms and 77-92% yields for racemic forms.

Engineering of reaction media is an exciting alternative for modulating kinetic properties of biocatalytic reactions. We addressed the combined effect of an aqueous two-phase system (ATPS) and high hydrostatic pressure on the kinetics of the Candida boidinii formate dehydrogenase-catalyzed oxidation of formate to CO2. Pressurization was found to lead to an increase of the binding affinity (decrease of KM, respectively) and a decrease of the turnover number, kcat. The experimental approach was supported using thermodynamic modeling with the electrolyte Perturbed-Chain Statistical Associating Fl

Devising synthetic strategies to construct a covalent bond is a common research topic among synthetic chemists. A key driver of success is the high tunability of the conditions, including catalysts, reagents, solvents, and reaction temperature. Such flexibility of synthetic operations has allowed for the rapid exploration of a myriad of artificial synthetic transformations in recent decades. However, if we turn our attention to chemical reactions controlled in living cells, the situation is quite different; the number of hit substrates for the reaction-type is relatively small, while the crowd
