The concept of the Land of Not-Unhappiness refers to the potential achievement of eliminating the pathologies of the aging process. To inform of how close we are to settling in the land, we summarize and review the achievements of research on anti-aging interventions over the last hundred years with a specific focus on strategies that slow down metabolism, compensate for aging-related losses, and target a broad range of age-related diseases. We critically evaluate the existing interventions labeled as "anti-aging," such as calorie restriction, exercise, stem cell administration, and senolytics

Community engagement in biomedical trials is to ensure ethical conduct in research, yet it has been criticised regarding power and exploitation of vulnerable communities where trials take place. This makes community engagement processes complex. We report on one example of how the global politics of biomedical research and local issues of contemporary politics and identities intertwine in a community engagement process. These issues emerged during observations in staff training at a biomedical HIV prevention trial centre in South Africa from September to November 2015. Within the practices of …

BACKGROUND: Prospective study protocols and registrations can play a significant role in reducing incomplete or selective reporting of primary biomedical research, because they are pre-specified blueprints which are available for the evaluation of, and comparison with, full reports. However, inconsistencies between protocols or registrations and full reports have been frequently documented. In this systematic review, which forms part of our series on the state of reporting of primary biomedical, we aimed to survey the existing evidence of inconsistencies between protocols or registrations (i.e…

We present evidence that the dog hookworm (Ancylostoma caninum) is underutilised in the study of host-parasite interactions, particularly as a proxy for the human-hookworm relationship. The inability to passage hookworms through all life stages in vitro means that adult stage hookworms have to be harvested from the gut of their definitive hosts for ex vivo research. This makes study of the human-hookworm interface difficult for technical and ethical reasons. The historical association of humans, dogs and hookworms presents a unique triad of positive evolutionary pressure to drive the A. caninu

Social and biomedical scientists, journal editors, and public health officials continue to debate the merits of the use of race and ethnicity in health-related research. As biomedical research focuses on issues of racial or ethnic health disparities, it remains unclear how biomedical scientists investigate race or ethnicity and health. This paper examines how biomedical researchers construct and analyze race or ethnicity in their studies and what conclusions they make about difference and health. Using content analysis of 204 biomedical research journal publications, which were supported by gr…

OBJECTIVES: The ability of large language models (LLMs) to answer targeted scientific questions by synthesizing information from research articles remains an open research challenge.
METHODS: We evaluated the effects of fine-tuning and a question-specific prompting strategy to answer 16 pre-defined questions about HIV drug resistance studies, including whether viral genetic sequences were reported and the demographics and antiviral treatments of the individuals from whom sequences were obtained. For fine-tuning, we constructed an instruction set comprising 250 HIV drug resistance studies, wit

BACKGROUND: The health and welfare of pigs used in biomedical research is essential to research quality and compliance with the 3Rs (replacement, reduction, and refinement). Hematological variables are objective markers to quantitatively determine health issues and evaluate physiological differences before and after experimental procedures. There are no recent validated hematologic reference intervals (RIs) published for Danish crossbred Landrace Yorkshire Duroc (LYD) pigs to aid researchers and veterinarians in their decision-making. The objective of this study was to establish hematologic RI…

There are many reasons why novel therapeutics fail in clinical trials but these failures are often attributed to lacking quality of preclinical data. These problems are not limited to any specific therapeutic area, academic or industrial research and are due in large part to several generic factors influencing research quality (e.g., related to definition of pre-specified endpoints, principles of study design and analysis, biased reporting, and lack of proper training). Yet, neuroscience drug discovery is often said to be affected more than the other fields. Within neuroscience, behavioral stu

LCAT converts free cholesterol to cholesteryl esters in the process of reverse cholesterol transport. Familial LCAT deficiency (FLD) is a genetic disease that was first described by Kaare R. Norum and Egil Gjone in 1967. This report is a summary from a 2017 symposium where Dr. Norum recounted the history of FLD and leading experts on LCAT shared their results. The Tesmer laboratory shared structural findings on LCAT and the close homolog, lysosomal phospholipase A2. Results from studies of FLD patients in Finland, Brazil, Norway, and Italy were presented, as well as the status of a patient reg

In the field of biomedical HIV prevention, researchers have meaningfully incorporated behavioral and social sciences research (BSSR) into numerous clinical trials, though the timing and degree of integration have been highly variable. The literature offers few frameworks that systematically characterize these collaborations. To fill this gap, we developed a typology of BSSR approaches within biomedical HIV prevention research. Focusing on trials that had safety and/or efficacy endpoints, we identified five approaches for combining BSSR and clinical research: formative, embedded, parallel, expl

The quality criteria of biomedical journals--and of 'Magnesium Research' as such--are being given a new insight. This has practical implications for the contributors and the editors. General considerations on the patterns of evaluation of scientific papers highlight five types of errors, that may be incurred in submitted manuscripts. 1. The information conveyed may remain ambiguous: for example, lack of discrimination between acute and chronic patterns, or confusion between the clinical and toxicological consequences of pharmacological and physiological studies: for example it is a real scient

The emergence of super-resolution microscopy (SRM) has had a significant impact on the research field of diffraction-limited optical fluorescence microscopy, which has a span of applications from fundamental biophotonics to advanced materials science. SRM is a powerful technique capable of producing ultra-high-resolution images of organic, semiconductor materials and live cells by overcoming the conventional diffraction limit. In this manuscript, we discuss various strategies for improving the spatial and temporal resolution of SRM techniques without causing photodamage to the sample, which is

Alzheimer's disease (AD) is a significant global health issue, impacting 50 million individuals with dementia worldwide, a number projected to triple by 2050. Although research has been conducted for decades, there is no effective prevention, treatment, or early diagnostic method for AD. Research on AD in Latin America (LatAm) faces unique challenges compared to developed countries due to socioeconomic, cultural, and infrastructural factors. While the prevalence of dementia is rapidly increasing in LatAm due to demographic shifts, the region is underrepresented in research, diagnostics and car

This article examines how biomedical research articles construct sex, gender, sexuality and sexual orientation in relation to intersex variations. We analyse 368 biomedical articles (approximately 1.3 million words) from PubMed spanning 1950-2020, comparing publications before and after the 2006 'Chicago Consensus' statement that introduced the terminology 'disorders of sex development' (DSD). Using multidimensional analysis and corpus-assisted discourse analysis, we identify key semantic domains and trace collocational patterns around terms including sex, gender, identity, sexual, and psychos…
