Direct Addition of Grignard Reagents to Aliphatic Carboxylic Acids Enabled by Bulky turbo-Organomagnesium Anilides.

The synthesis of ketones through addition of organometallic reagents to aliphatic carboxylic acids is a straightforward strategy that is limited to organolithium reagents. More desirable Grignard reagents can be activated and controlled with a bulky aniline-derived turbo-Hauser base. This operationally simple procedure allows the straightforward preparation of a variety of aliphatic and perfluoroalkyl ketones alike from functionalized alkyl, aryl and heteroaryl Grignard reagents.

CuH-Catalyzed Regio- and Enantioselective Hydrocarboxylation of Allenes: Toward Carboxylic Acids with Acyclic Quaternary Centers.

We report a method to prepare α-chiral carboxylic acid derivatives, including those bearing all-carbon quaternary centers, through an enantioselective CuH-catalyzed hydrocarboxylation of allenes with a commercially available fluoroformate. A broad range of heterocycles and functional groups on the allenes were tolerated in this protocol, giving enantioenriched α-quaternary and tertiary carboxylic acid derivatives in good yields with exclusive branched regioselectivity. The synthetic utility of this approach was further demonstrated by derivatization of the products to afford biologically impor

Omega-3-carboxylic acids provide efficacious anti-inflammatory activity in models of crystal-mediated inflammation.

This study assesses the efficacy and exposure-response relationship of omega-3-carboxylic acids (OM-3 CA) in models of crystal-based inflammation. Human THP-1 macrophages and primary peripheral blood mononuclear cells exposed to multiple inflammatory crystal types were used to determine the anti-inflammatory potential of omega-3 (OM-3) fatty acids in vitro. Anti-inflammatory effects of OM-3 CA in vivo were tested in rat monosodium urate (MSU) crystal air pouch and rat knee intra-articular MSU injection models. Acute treatment with the OM-3 fatty acid docosahexaenoic acid suppressed MSU-, chole

Omega-3 carboxylic acids (Epanova): a review of its use in patients with severe hypertriglyceridemia.

Omega-3 carboxylic acids (Epanova) [OM3-CA] is the first free fatty acid form of long-chain marine omega-3 fatty acids (eicosapentaenoic acid and docosahexaenoic acid being the most abundant) to be approved by the US FDA as an adjunct to diet to lower triglyceride levels in patients with severe hypertriglyceridemia (≥ 500 mg/dL). Oral OM3-CA has greater bioavailability than ethyl ester forms of omega-3 and, unlike omega-3 acid ethyl esters, does not require co-ingestion of a high-fat meal, as it does not need pancreatic enzyme activity for absorption. Parahydrogen-induced polarization of carboxylic acids: a pilot study of valproic acid and related structures.

Parahydrogen-induced polarization (PHIP) is a promising new tool for medical applications of MR, including MRI. The PHIP technique can be used to transfer high non-Boltzmann polarization, derived from parahydrogen, to isotopes with a low natural abundance or low gyromagnetic ratio (e.g. (13)C), thus improving the signal-to-noise ratio by several orders of magnitude. A few molecules acting as metabolic sensors have already been hyperpolarized with PHIP, but the direct hyperpolarization of drugs used to treat neurological disorders has not been accomplished until now. Here, we report on the firs

Thermodynamics of the distribution of some carboxylic acids between organic solvents and a perchlorate solution.

The distribution of formic, benzoic and 4-methylbenzoic acids between a 0.1 mol dm(-3) (Na+, H+)ClO4- solution and octanol and carbon tetrachloride was studied at five different temperatures of 10 - 30 degrees C. The thermodynamic parameters for the transfer process of monomeric acid between two phases and for the dimerization of monomeric acid in carbon tetrachloride (deltaH and deltaS) were evaluated from the van't Hoff isochore. The free-energy change for the transfer of a methylene group from water to organic solvents was entropically controlled, irrespective of the organic solvents. The d

Inhibition of a medium chain acyl-CoA synthetase involved in glycine conjugation by carboxylic acids.

Molecular characteristics of carboxylic acids were investigated for the ability to inhibit a purified medium chain acyl-CoA synthetase, using hexanoic acid as a substrate. Salicylic acid, 4-methylsalicylic acid, 2-hydroxynaphtoic acid, and 2-hydroxyoctanoic acid, which do not act as substrates for the medium chain acyl-CoA synthetase, were potent as inhibitors. Valproic acid was not an inhibitor. Salicylic acid, 2-hydroxynaphthoic acid, and 2-hydroxyoctanoic acid inhibited the medium chain acyl-CoA synthetase with Ki values of 37, 5.2, and 500 microM, respectively. 4-Methylsalicylic acid was m

Well-Defined Molecular Alkali Metal Catalysts for Mild and Selective Reduction of Carboxylic Acids by Hydrosilylation.

The selective transformation of carboxylic acids into alcohols remains a challenge, although many catalytic methods have been developed recently. Hydrosilylative reduction appeared to be an attractive strategy in the presence of a metallic catalyst. In those processes, transition metals were usually employed for the catalyst design, and the reactions often suffered from poor selectivity, limited functional group tolerance, and the requirement of elevated temperature. Here, we report inexpensive and easy-to-make alkali metal catalysts that effectively promote the hydrosilylation of carboxylic a

Repurposing TPP-dependent enzymes with anodic oxidation for the conversion of aldehydes to enantioenriched carboxylic acids.

Thiamine diphosphate (ThDP)-dependent enzymes are highly promiscuous and robust biocatalysts in nature, orchestrating precise C-C bond formation and cleavage reactions. Combining organophotoredox catalysis with protein engineering, our group has repurposed ThDP-dependent enzymes for stereoselective dual photo-/enzymatic radical transformations. Inspired by the carboxylic acid byproducts in photo/thiamine biocatalytic systems, we postulated that the input of external electricity could offer an alternative method to expand the catalytic repertoire of ThDP-dependent enzymes. Recently, we integrat

Fixed-bed biofilm reactor for single-stage bioconversion of organic waste to medium-chain carboxylic acids.

Biofilm-based systems can enhance medium-chain carboxylic acids (MCCAs) production by retaining slow-growing microorganisms and increasing biomass density. This study investigated a novel single-stage fixed-bed biofilm system for the co-fermentation of food waste and sewage sludge. Significant enhancements in stability and metabolic efficiency were achieved without external pH control or the addition of external electron donors, as the process relied on the endogenous in-situ production of lactate and ethanol. Using a 1:1 support-to-inoculum ratio with porous sintered glass as solid support, t

Copper-Catalyzed Domino Reaction of Isocyanides with Carboxylic Acids and PhSO2SeCH3 to Access Se-CH3 Selenocarbamates.

We described a new path for the preparation of Se-CH3 selenocarbamates from readily accessible carboxylic acids, isocyanides, and PhSO2SeCH3 with the formation of new C-Se and C-N bonds. This protocol is not only suitable for late-stage derivatization of carboxylic acid drugs but also permits double reactions of three-component reactions. Most importantly, compound 4t possesses potential antioxidant activity and may alleviate oxidative stress damage by regulating the Nrf2 signaling pathway.

A Highly Selective Cobalt Catalyst for Primary Amine Synthesis from Carboxylic Acids, Esters, and Vegetable Oils.

The prevalence of carboxylic acids and esters in nature renders them diverse and conveniently accessible building blocks for chemical synthesis. In this Letter, we present a straightforward cobalt-based catalytic system for the selective synthesis of primary amines from carboxylic acid derivatives and ammonia. The current system has been demonstrated to provide an efficient synthesis route for various amine derivatives and exhibits excellent chemoselectivity despite the complex reaction network. The optimal catalyst offers broad functional group tolerance, with both (hetero)aromatic and alipha

Levels of perfluoroalkyl carboxylic acids in recycled plastic pellets: A small input into the plastics cycle.

Perfluoroalkyl carboxylic acids (PFCAs) are used in electrical products and food packaging, and could enter the circular plastic economy. In this study, the concentrations of PFCAs (C6 to C15) were measured in 99 recycled plastic pellet samples collected in Japan and nine selected countries from 2021 to 2024. The concentration distribution of PFCAs in recycled plastic pellets (n = 99) was 59% below 50 ng/kg for total PFCAs. The average total PFCA concentration in the samples from Japan (n = 68) was 73 ng/kg (16 ng/kg from perfluorooctanoic acid, PFOA) and the maximum concentration was 540 ng/k

Development of novel anilinoquinazoline-based carboxylic acids as non-classical carbonic anhydrase IX and XII inhibitors.

As part of our ongoing endeavour to identify novel inhibitors of cancer-associated CA isoforms IX and XII as possible anticancer candidates, here we describe the design and synthesis of small library of 2-aryl-quinazolin-4-yl aminobenzoic acid derivatives (6a-c, 7a-c, and 8a-c) as new non-classical CA inhibitors. On account of its significance in the anticancer drug discovery and in the development of effective CAIs, the 4-anilinoquinazoline privileged scaffold was exploited in this study. Thereafter, the free carboxylic acid functionality was appended in the ortho (6a-c), meta (7a-c), or para

Carbene and photocatalyst-catalyzed decarboxylative radical coupling of carboxylic acids and acyl imidazoles to form ketones.

The carbene and photocatalyst co-catalyzed radical coupling of acyl electrophile and a radical precursor is emerging as attractive method for ketone synthesis. However, previous reports mainly limited to prefunctionalized radical precursors and two-component coupling. Herein, an N-heterocyclic carbene and photocatalyst catalyzed decarboxylative radical coupling of carboxylic acids and acyl imidazoles is disclosed, in which the carboxylic acids are directly used as radical precursors. The acyl imidazoles could also be generated in situ by reaction of a carboxylic acid with CDI thus furnishing a
