Emergency nursing care of a patient with acute appendicitis complicated by esomeprazole-induced anaphylactic shock: a case report.

RATIONALE: Esomeprazole-induced anaphylactic shock followed by cardiac arrest is an extremely rare but life-threatening adverse event, particularly in patients with acute appendicitis. Clinical nursing experience in managing such critical conditions remains limited, and this case report provides valuable insights into emergency and intensive nursing care. PATIENT CONCERNS: A 53-year-old male patient with acute appendicitis developed facial flushing, urticaria, and loss of consciousness within 5 min after starting esomeprazole infusion, rapidly progressing to ventricular fibrillation and cardi

Comparative Analysis of the Efficacy of Esomeprazole and Ilaprazole in Patients With Neurological Disorders Using the Gastroesophageal Reflux Disease Questionnaire.

OBJECTIVES: Proton pump inhibitors (PPIs) are widely used to reduce gastric acid levels and are often prescribed with antiplatelet agents in patients with stroke. However, the interactions and differences among various PPIs remain unclear. Therefore, we aimed to compare the differences between esomeprazole and ilaprazole in patients with and without stroke. We also compared the effects of aspirin use in the ilaprazole group. METHODS: We retrospectively analyzed 580 patients with neurological disorders who responded to the Gastroesophageal Reflux Disease Questionnaire at a tertiary hospital be

Deduction of full factorial design of HPLC technique for the simultaneous analysis of meloxicam and esomeprazole in their laboratory prepared tablets.

A reversed phase HPLC method with UV detection was designed for the first time for the simultaneous estimation of meloxicam and esomeprazole in their combined tablet dosage forms. Full factorial design was used for rapid optimization of the proposed method. Chromatographic separation was reached using isocratic elution on C18 column. The mobile phase was a mixture of methanol: acetonitrile: 0.05 M potassium dihydrogen phosphate buffer at pH 5, adjusted using phosphoric acid and/or NaOH when needed. The flow rate was 1 mL/min and the injection volume was 20 µL. The detection wavelength was 230

The effect of esomeprazole on the upper GI tract release and systemic absorption of mesalazine from colon targeted formulations.

The aim of the present study was to investigate the effect of coadministration of the proton pump inhibitor (PPI) esomeprazole on the upper GI tract behavior and systemic exposure of mesalazine from two mechanistically different colon targeted delivery systems: Claversal (pH-dependent release) and Pentasa (prolonged release). To this end, gastric, jejunal and systemic concentrations of mesalazine and its metabolite N-acetyl mesalazine were monitored in 5 healthy volunteers following oral intake of Pentasa or Claversal with or without PPI pre-treatment (cross-over study). Our exploratory study

Esomeprazole covalently interacts with the cardiovascular enzyme dimethylarginine dimethylaminohydrolase: Insights into the cardiovascular risk of proton pump inhibitors.

BACKGROUND: Proton pump inhibitors (PPIs) are widely prescribed drugs for the treatment of gastroesophageal reflux disease (GERD). Several meta-analysis studies have reported associations between prolonged use of PPIs and major adverse cardiovascular events. However, interaction of PPIs with biological molecules involved in cardiovascular health is incompletely characterized. Dimethylarginine dimethylaminohydrolase (DDAH) is a cardiovascular enzyme expressed in cardiomyocytes, and other somatic cell types in one of two isotypes (DDAH1 and DDAH2) to metabolize asymmetric dimethylarginine (ADMA)

Multivariate Model Update Chemometric Methods for Determination of Prednisolone and Esomeprazole in Spiked Human Plasma: A Comparative Study.

BACKGROUND: Prednisolone (PRD) is an immunosuppressant and anti-inflammatory drug, although it may cause peptic ulcers as a side effect. Esomeprazole (ESO) is used for the treatment of peptic ulcers, therefore the two drugs are co-administered in cases of organ transplantation and autoimmune diseases. OBJECTIVE: This work aims to determine the two drugs simultaneously, in bulk, and in spiked human plasma by eliminating the overlap of their spectra and the interference of the plasma matrix. METHODS: Two simple and effective updated chemometric models-principal component analysis (PCA) and par

Combination of esomeprazole and pirfenidone enhances antifibrotic efficacy in vitro and in a mouse model of TGFβ-induced lung fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal lung disease of unknown etiology. Currently, pirfenidone and nintedanib are the only FDA-approved drugs for the treatment of IPF and are now the standard of care. This is a significant step in slowing down the progression of the disease. However, the drugs are unable to stop or reverse established fibrosis. Several retrospective clinical studies indicate that proton pump inhibitors (PPIs; FDA-approved to treat gastroesophageal reflux) are associated with favorable outcomes in patients with IPF, and emerging preclinical studies repo

Evaluation of the effectiveness of esomeprazole treatment strategies in the management of patients with gastroesophageal reflux disease symptoms: a meta-analysis.

Here we aimed to evaluate the effectiveness of esomeprazole treatment strategies comparing with other proton pump inhibitors (PPI) in clinical practice for six months in the management of patients with symptomatic gastroesophageal reflux disease (GERD). An extensive search of the literature focusing on PPI therapeutic evaluation was performed up to December 2014. Risk ratio (RR) with its corresponding 95% confidence intervals (CIs) in each study was chosen as the effect size. Cochrane's Q statistic and I2 test were both conducted to evaluate heterogeneity across individual studies. Meta-regres
