Chitosan-coated silver nanoparticles amplify glycopeptide efficacy against Staphylococcus aureus.

The escalating prevalence of antibiotic resistance represents a critical threat to global public health, while the development of new antibiotics remains limited. Vancomycin, a glycopeptide antibiotic, is essential for treating severe Gram-positive infections. However, the emergence of vancomycin-intermediate (VISA) and vancomycin-resistant (VRSA) Staphylococcus aureus strains has raised significant concerns due to their challenging resistance profiles and limited treatment options. In this study, we developed a carboxylic acid-capped silver nanoparticle (AgNPs) platform to enhance the therape

Glycopeptide Heteroresistance Among Coagulase-Negative Staphylococci: A Systematic Review and Meta-analysis.

We identified a high prevalence of glycopeptide heteroresistance amongst coagulase-negative staphylococci (41.4%, 95% confidence interval 30.7-52.9; meta-analysis including n = 1432 isolates). Heteroresistance was associated with methicillin resistance, did not require glycopeptide exposure, and may be more prevalent among isolates from invasive infections. Heteroresistance may represent an under-appreciated reason for treatment failure of CoNS infections.

Comprehensive Immunoglobulin G, A, and M Glycopeptide Profiling for Large-Scale Biomedical Research.

Glycosylation of immunoglobulin G (IgG) is recognized as a key modulator of cellular effector functions. At the same time, an increasing body of evidence underlines the importance of other antibody isotypes, especially IgA and IgM, in pathophysiological conditions. Therefore, methods to efficiently study the complex interplay between isotypes, subclasses, and glycosylation of antibodies during acute and chronic states of inflammation are needed. As a solution, we present an integrated and comprehensive method combining simultaneous affinity enrichment of IgG, IgA, and IgM with a single measure

Lycium barbarum glycopeptide ameliorates motor and visual deficits in autoimmune inflammatory diseases.

BACKGROUND: Lycium barbarum glycopeptide (LbGp), extracted from the traditional Chinese medicine (TCM) of Lycium barbarum (LB), provides a neuroprotective effect against neurodegenerative and neuroimmune disorders contributing to its immunomodulatory and anti-inflammatory roles. Neuromyelitis optica spectrum disorders (NMOSD) is an autoimmune-mediated central nervous system (CNS) demyelinating disease, clinically manifested as transverse myelitis (TM) and optic neuritis. However, no drug has been demonstrated to be effective in relieving limb weakness and visual impairment of NMOSD patients.

Experimental Evaluation of the Hypersensitivity Reactions of a New Glycopeptide Antibiotic Flavancin in Animal Models.

Glycopeptide antibiotics are still in demand in clinical practice for treating infections caused by resistant gram-positive pathogens; however, their use is limited due to severe adverse reactions. Their predominant types of side effects are immunoglobulin E-mediated or nonmediated hypersensitivity reactions. Therefore, the development of new glycopeptide antibiotics with improved toxicity profiles remains an important objective in advancing modern antimicrobial agents. We investigated a new eremomycin aminoalkylamide flavancin, its anaphylactogenic properties, influence on histamine levels in

Genomic Insights into the Distribution and Phylogeny of Glycopeptide Resistance Determinants within the Actinobacteria Phylum.

The spread of antimicrobial resistance (AMR) creates a challenge for global health security, rendering many previously successful classes of antibiotics useless. Unfortunately, this also includes glycopeptide antibiotics (GPAs), such as vancomycin and teicoplanin, which are currently being considered last-resort drugs. Emerging resistance towards GPAs risks limiting the clinical use of this class of antibiotics-our ultimate line of defense against multidrug-resistant (MDR) Gram-positive pathogens. But where does this resistance come from? It is widely recognized that the GPA resistance determi

Factors associated with acquisition of glycopeptide-resistant enterococci during a single-strain outbreak.

The aim of our study was to describe and to investigate the factors associated with glycopeptide-resistant enterococci (GRE) acquisition during a single-strain outbreak which occurred in several wards of hospital from September 2013 to January 2014. We designed a case-control study. Analyses were performed using Bayesian methods. Univariate logistic regressions with informative priors from published studies were conducted. A multivariate model was build including variables with a probability of odd-ratio exceeding one (Pr) >85% or <15%. Thirteen cases and 52 controls were recruited. The

Relationship between glycopeptide production and resistance in the actinomycete Nonomuraea sp. ATCC 39727.

Glycopeptides and β-lactams inhibit bacterial peptidoglycan synthesis in Gram-positive bacteria; resistance to these antibiotics is studied intensively in enterococci and staphylococci because of their relevance to infectious disease. Much less is known about antibiotic resistance in glycopeptide-producing actinomycetes that are likely to represent the evolutionary source of resistance determinants found in bacterial pathogens. Nonomuraea sp. ATCC 39727, the producer of A40926 (the precursor for the semisynthetic dalbavancin), does not harbor the canonical vanHAX genes. Consequently, we invest…

Reduction in glycopeptide resistance in vancomycin-resistant enterococci as a result of vanA cluster rearrangements.

The molecular characterization of five clinical isolates of vanA-containing vancomycin-resistant enterococci with altered resistance to glycopeptides was examined. One strain represented an IS1216V insertion accompanied by partial deletion of the reading frame of vanX following a transposition event. The other four strains represented IS1216V within the vanX-vanY intergenic region associated with deletion of vanY or vanZ.

Glycopeptide specificity of helper T cells obtained in mouse models for rheumatoid arthritis.

Five protected analogues of beta-D-galactosyl-(5R)-5-hydroxy-L-lysine were prepared, in which the galactosyl moiety was modified by monodeoxygenation or inversion of stereochemistry at C-4. The building blocks were used in the solid-phase synthesis of a set of glycopeptides related to the peptide fragment CII256-273 from type II collagen. Evaluation of the glycopeptides revealed that T-cell hybridomas obtained in collagen-induced arthritis (CIA), which is a common mouse model for rheumatoid arthritis, recognized the galactosyl moiety with high specificity for individual hydroxy groups. Moreove…

Use of a glycopeptide antibiotic, teicoplanin, in the treatment of septicaemia caused by gram-positive bacteria.

Teicoplanin, a recently introduced glycopeptide antibiotic, has been used, in combination with other antibiotics, to treat 31 episodes of septicaemia caused by Gram-positive organisms. Teicoplanin has double the activity of vancomycin against many Gram-positive bacteria, but allergic reactions and toxicity appear to be infrequent. A single daily dose is sufficient to maintain therapeutic levels, which is an advantage in conditions requiring long-term treatment. Of the 31 episodes treated, 16 were associated with infective endocarditis, 11 with Hickman catheter infection, two with bone and join

Solitary magnocellular neurons in the homozygous Brattleboro rat have vasopressin and glycopeptide immunoreactivity.

A small but distinctive population (about 1 in 600) of magnocellular neurosecretory neurons in homozygous Brattleboro rats are immunoreactive for vasopressin, and a similar number for the carboxy-terminal glycopeptide of the vasopressin prohormone. These solitary cells are found in all animals and in all parts of the magnocellular system, but not in the suprachiasmatic or other hypothalamic nuclei. The majority of the solitary cells do not differ morphologically from the remainder of the magnocellular neurons. The immunoreactivity is markedly denser in the Nissl bodies than in the Golgi region

Mechanism of action of glycopeptide hormones and cholera toxin: what is the role of ADP-ribosylation?

The cultured murine Leydig tumor cell line MLTC-1 and the normal rat thyroid strain FRTL have adenylate cyclase activities that are stimulated by human chorionic gonadotropin (hCG) and thyrotropin, respectively. Both cell types also respond to choleragen. Activation of adenylate cyclase in membranes by choleragen required NAD whereas stimulation of the enzyme by hormones did not. With [alpha-32P]NAD as a donor, ADP-ribosylation of membrane proteins was determined under the same conditions used to assay adenylate cyclase activity. Under these conditions, choleragen, but not the hormones, caused

Axonal transport of glycoproteins in regenerating olfactory nerve: enhanced glycopeptide concanavalin A-binding.

The size and concanavalin A-binding characteristics of glycopeptides derived from axonally transported glycoproteins were studied in regenerating garfish olfactory nerve. A regeneration related increase was observed in the proportion of total glycopeptide radioactivity associated with the lower molecular weight, dialyzable fraction. There was also a 3-5 fold increase in the axonal transport of low molecular weight concanavalin. A-binding glycopeptides in regenerating nerve. These results suggest a shift to an enhanced synthesis and axonal transport of glycoproteins containing low molecular wei…

Rapid Glass Wool Enrichment of Glycopeptides for MALDI-MS Analysis of Immunoglobulin G Glycosylation in COVID-19 Samples.

Glycosylation is the most common protein post-translational modification, affecting protein properties and functions. Abnormal variations in glycosylation are associated with diseases, e.g., coronavirus disease COVID-19. Matrix-assisted laser desorption/ionization mass spectrometry has been widely utilized for studying protein glycosylation, after proper purification of glycopeptides or glycans using hydrophilic interaction liquid chromatography (HILIC) or laboratory-synthesized hydrophilic materials. Here, glass wool tips were developed to enrich immunoglobulin G glycopeptides and applied in …
