Investigating animal models of optic neuropathy: An accurate method for optic nerve and chiasm dissection in mice.

BACKGROUND: Numerous disorders affecting the optic nerve require histological examination of whole length optic nerves and chiasm. Most methods employed to study the histopathology of the optic nerves in animal models of human diseases involve resection of a short retrobulbar section after eye globe exenteration, commonly obtained in mice. This approach might affect the morphology of the optic nerve, thus limiting accurate identification of pathological changes in the tissue. Some histological studies were performed on longer or more posterior parts of the anterior visual pathway included the

ALDH1A3 loss of function causes bilateral anophthalmia/microphthalmia and hypoplasia of the optic nerve and optic chiasm.

The major active retinoid, all-trans retinoic acid, has long been recognized as critical for the development of several organs, including th … bilateral A/M and a simplex case with bilateral anophthalmia and hypoplasia of the optic nerve and optic chiasm. Analysis of novel sequence variants revealed homozygosity for two nonsense mutations in ALDH1A3, c.568A>G, predicting p.Lys190*, in the familial cases, and c.1165A>T, predicting p.Lys389*, in the simplex case. Both mutations predict nonsense-mediat …

Optical coherence tomography and visual field discordance in optic chiasm compression in patients with pituitary adenomas.

PURPOSE: To evaluate whether optical coherence tomography (OCT) structural measurements correlate with visual field (VF) deficits in pituitary adenomas and to determine when OCT abnormalities appear relative to VF defects. METHODS: This retrospective study included 95 patients (190 eyes) who underwent magnetic resonance imaging, automated perimetry, and OCT between 2013 and 2023. Patients were classified as compression (n = 67), abutting (n = 15), or noncontact (n = 13). VF index ([VFI], mean deviation [MD], pattern standard deviation [PSD]) and OCT parameters (retinal nerve fibre layer [RNFL

Congenital aplasia of the optic chiasm presenting as optic nerve hypoplasia with no other ocular or systemic comorbidity.

We introduce a case of an infant girl who presented to our paediatric ophthalmology department with no behavioural response to visual targets and horizontal nystagmus. An ocular assessment revealed isolated bilateral small optic discs prompting further investigation. Her biochemical and endocrinological profiles were normal but a head MRI identified a total absence of the optic chiasm and optic tracts. Electrodiagnostic tests confirmed the diagnosis of aplasia of the optic chiasm. Our patient had no systemic comorbidities and has shown excellent milestone progression. She is now in a mainstrea…

Dimensions of the optic nerves, chiasm, and tracts: MR quantitative comparison between patients with optic atrophy and normals.

OBJECTIVE: The dimensions of the optic nerves, chiasm, and tracts were determined in normal patients and in patients with clinical evidence of optic atrophy to establish whether there was a significant difference between the two groups. MATERIALS AND METHODS: The authors analyzed thin section coronal MRI of 110 patients. Seventy-seven patients were normal, 20 had visual impairment without funduscopic evidence of optic atrophy, and 13 had visual impairment with funduscopic evidence of optic atrophy. RESULTS: The normal mean dimensions were (height x width): optic tracts 2.8 x 5.1 mm; nerves 3

Cavernous Malformation of the Optic Nerve and Chiasm: Prompt Suspicion and Surgery Matter.

BACKGROUND: Cavernous malformations (CMs) of the optic nerve and chiasm are extremely rare, accounting for less than 1% of all intracranial … fects corresponding to a heterogeneously minimally enhancing mass with blood products enlarging the optic chiasm and proximal right optic nerve. Surgical excision of the CM 8 weeks after onset of visual loss improved vision from 20/40 to 20/15 with improved visual fields. A 33-year-old woman with a history of familial multiple CMs, diagnosed at age 18, reporte …

Querectin improves myelin repair of optic chiasm in lyolecithin-induced focal demyelination model.

Although the beneficial effects of quercetin on oligodendrocyte precursor cell (OPCs) population has been evaluated in-vitro, there are few studies about the effects of quercetin on myelin repair in the context of demyelination. The aim of this study was to investigate the effects of querectin on functional recovery and myelin repair of optic chiasm in lysolecithin (LPC)-induced demyelination model. Demyelination was induced by local injection of LPC 1% (2 μl) into rat optic chiasm. Querectin at doses 25 or 50 mg/kg was administrated daily by oral gavage for 7 or 14 days post LPC. Visual evoke…

Diencephalic syndrome and disseminated juvenile pilocytic astrocytomas of the hypothalamic-optic chiasm region.

BACKGROUND: Diencephalic syndrome (DS) is a complex of signs and symptoms related to hypothalamic dysfunction; its main features are emaciation, despite a normal or slightly diminished caloric intake, and an alert appearance. DS has been almost exclusively described in association with space-occupying lesions of the hypothalamic-optic chiasm region, mainly juvenile pilocytic astrocytoma (JPA). A systematic diagnostic approach, including contrast-enhanced magnetic resonance imaging (MRI) of the child's head, is rapidly expanding our knowledge of this syndrome. METHODS: The MRI findings for thr…

Childhood optic chiasm gliomas: radiographic response following radiotherapy and long-term clinical outcome.

PURPOSE: In children with chiasmal gliomas, radiation therapy can arrest progressive visual and neurologic impairment. We examined the radiographic response and clinical outcomes after irradiation. METHODS AND MATERIALS: Forty-two children (median age at diagnosis, 6.6 years) with chiasmal gliomas were managed as follows: 11 asymptomatic patients with neurofibromatosis-1 (NF-1) were observed only; 2 patients, less than 3 years old, underwent surgery and chemotherapy to delay irradiation; and 29 patients with progressive disease received radiation with or without prior surgery or chemotherapy.

Investigation of Demyelination and Remyelination Processes of the Compressed Optic Chiasm: An Experimental Model.

AIM: To demonstrated demyelination and remyelination of the optic nerve histologically by electron microscopy in an experimental model similar to the compression of pituitary adenomas on the optic chiasm. MATERIAL AND METHODS: The rats were fixed to a stereotaxic device under deep anesthesia, and a balloon catheter was placed under the optic chiasm through a burr hole which was in front of the bregma in accordance with the brain atlas of rats. The animals were divided into five groups (n=8): control, mild compression demyelination, severe compression demyelination, mild compression remyelinat…

Compression of the optic chiasm is associated with reduced photoentrainment of the central biological clock.

OBJECTIVE: Pituitary tumours that compress the optic chiasm are associated with long-term alterations in sleep-wake rhythm. This may result from damage to intrinsically photosensitive retinal ganglion cells (ipRGCs) projecting from the retina to the hypothalamic suprachiasmatic nucleus via the optic chiasm to ensure photoentrainment (i.e. synchronisation to the 24-h solar cycle through light). To test this hypothesis, we compared the post-illumination pupil response (PIPR), a direct indicator of ipRGC function, between hypopituitarism patients with and without a history of optic chiasm compres…

Configuration of the optic chiasm in humans with albinism as revealed by magnetic resonance imaging.

PURPOSE: To determine whether the size and configuration of the optic chiasm in humans with albinism is different from that in normal control subjects. METHODS: Seventeen patients and 15 control subjects underwent magnetic resonance imaging of the entire head. Images were reformatted to the chiasm region and analyzed with observer-independent morphometry and measurements by a blinded observer. RESULTS: The albino group showed significantly smaller chiasmatic widths, smaller optic nerves and tracts, and wider angles between nerves and tracts. Statistical morphometry showed a different configu…

CXCL12 promotes the crossing of retinal ganglion cell axons at the optic chiasm.

Binocular vision requires the segregation of retinal ganglion cell (RGC) axons extending from the retina into the ipsilateral and contralateral optic tracts. RGC axon segregation occurs at the optic chiasm, which forms at the ventral diencephalon midline. Using expression analyses, retinal explants and genetically modified mice, we demonstrate that CXCL12 (SDF1) is required for axon segregation at the optic chiasm. CXCL12 is expressed by the meninges bordering the optic pathway, and CXCR4 by both ipsilaterally and contralaterally projecting RGCs. CXCL12 or ventral diencephalon meninges potentl…

Physiological motion of the optic chiasm and its impact on stereotactic radiosurgery dose.

OBJECTIVE: Avoidance of radiation-induced optic neuropathy (RION) from stereotactic radiosurgery (SRS) requires precise anatomical localization; however, no prior studies have characterized the physiologic motion of the optic chiasm. We measured the extent of chiasm motion and its impact on SRS dose. METHODS: In this cross-sectional study, serial MRI was performed in multiple planes in 11 human subjects without optic pathway abnormalities to determine chiasm motion across time. Subsequently, the measured displacement was applied to the hypothetical chiasm dose received in 11 patients treated …

Damage to the optic chiasm in myelin oligodendrocyte glycoprotein-experimental autoimmune encephalomyelitis mice.

Optic chiasm lesions in myelin oligodendrocyte glycoprotein (MOG)-experimental autoimmune encephalomyelitis (EAE) mice were characterized using magnetic resonance imaging (MRI) and validated using electron microscopy (EM). MR images were collected from 3 days after induction to remission, approximately 20 days after induction. Hematoxylin and eosin, solochrome cyanin-stained sections, and EM images were obtained from the optic chiasms of some mice approximately 4 days after disease onset when their scores were thought to be the highest. T2-weighted imaging and apparent diffusion coefficient ma…
