The efficacy of various classes of anti-emetics in preventing deoxynivalenol-induced vomiting in swine.

The Fusarium mycotoxin deoxynivalenol (DON) is a potent emetic agent. While the basic mechanisms which invoke and mediate emesis are still poorly understood, various neurotransmitters appear to be involved. The action of these transmitters can be blocked by various receptor-specific antagonists. The current study investigated the efficacy of several classes of receptor antagonists to block the emetic effect of DON. Following anti-emetic pretreatment, pigs were administered the toxin (i.v., 80 micrograms/kg, or oral, 300 micrograms/kg) and the onset of emesis was monitored. Certain specific ser

T-2 Toxin Neurotoxicity: Molecular Mechanisms and Emerging Chemoprotective Strategies.

Mycotoxins can contaminate food and raw food materials and are a threat to animal and human health. T-2 toxin is the most toxic secondary metabolite mainly produced by Fusarium species among trichothecenes. T-2 toxin exposure can induce multiple toxic effects, including hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, and reproductive toxicity. Recent studies have reported that T-2 toxin can cross the blood-brain barrier and trigger neurotoxicity. In this review, we summarized the neurotoxic effects caused by T-2 toxin exposure and the underlying molecular mechanisms.…

Revisiting the PCR-Based Molecular Approaches for Mycotoxigenic Fusarium Species Detection and Quantification in Wheat.

Fusarium species cause yield losses in wheat production through fusarium head blight (FHB) and the associated contamination of regulated mycotoxins, such as trichothecenes and zearalenone. Despite the widespread use of PCR-based molecular approaches for Fusarium detection, quantification, and chemotyping, most primers were developed prior to both modern phylogenetic reclassification and the availability of high-quality genome assemblies, leaving their specificity and robustness largely untested. Existing PCR- and qPCR-based assays for Fusarium detection in wheat were reviewed and re-evaluated …

Mycotoxin contamination in food and feed: Occurrence, co-occurrence, combined impacts on human and animal health, climate change implications, risk assessment, and mitigation strategies.

Mycotoxins are toxic secondary metabolites produced by numerous fungi that infect crops during pre- and post-harvest stages. Aflatoxins, ochratoxins, fumonisins, zearalenone, and trichothecenes are of greatest concern due to their widespread occurrence and high toxicity. Consumption of mycotoxin-contaminated food and feed poses serious public health risks, as these compounds are linked to carcinogenic, neurotoxic, nephrotoxic, hepatotoxic, and immunotoxic effects. The frequent co-occurrence of multiple mycotoxins further amplifies health risks through additive or synergistic interactions and c…

FgNup2 regulates nuclear import of the histone H2B monoubiquitination enzyme by stabilizing the FgImpα-FgBre1 complex to mediate pathogenicity in Fusarium graminearum.

Fusarium head blight caused by Fusarium graminearum threatens global wheat production. The pathogenicity of this fungus depends on histone H2B monoubiquitination (H2Bub1), yet the nuclear import mechanism of its key enzyme, FgBre1, remains unclear. In this study, we identified 24 nucleoporins in F. graminearum and found that FgNup2 is essential for fungal growth, toxin biosynthesis, and virulence. FgNup2 positively regulates H2Bub1 levels by facilitating the nuclear import of FgBre1, thereby influencing pathogenic development. The nuclear import of FgBre1 relies on the importin receptor FgImpα

Nutrient regulation targeting macrophage-controlled intestinal mucosal healing: A promising strategy against intestinal mucositis induced by deoxynivalenol.

Deoxynivalenol (DON), one of the most severely polluting mycotoxins globally, contaminates agricultural products and food through environmental pathways, compromising intestinal barrier integrity and homeostasis in humans and animals. This disruption triggers intestinal mucositis and exacerbates inflammatory bowel disease (IBD) progression. Current therapies focusing on immunosuppression face limitations due to drug resistance, shifting research toward mucosal healing strategies for sustained remission. Macrophages, essential mediators of intestinal immunity, exhibit metabolic and phenotypic p

Exposure to Subclinical Doses of Fumonisins, Deoxynivalenol, and Zearalenone Affects Immune Response, Amino Acid Digestibility, and Intestinal Morphology in Broiler Chickens.

Fusarium mycotoxins often co-occur in broiler feed, and their presence negatively impacts health even at subclinical concentrations, so there is a need to identify the concentrations of these toxins that do not adversely affect chickens health and performance. The study was conducted to evaluate the least toxic effects of combined mycotoxins fumonisins (FUM), deoxynivalenol (DON), and zearalenone (ZEA) on the production performance, immune response, intestinal morphology, and nutrient digestibility of broiler chickens. A total of 960 one-day-old broilers were distributed into eight dietary tre

Toxicity and action mechanisms of silver nanoparticles against the mycotoxin-producing fungus Fusarium graminearum.

INTRODUCTION: Fusarium graminearum is a most destructive fungal pathogen that causes Fusarium head blight (FHB) disease in cereal crops, resulting in severe yield loss and mycotoxin contamination in food and feed. Silver nanoparticles (AgNPs) are extensively applied in multiple fields due to their strong antimicrobial activity and are considered alternatives to fungicides. However, the antifungal mechanisms and the effects of AgNPs on mycotoxin production have not been well characterized. OBJECTIVES: This study aimed to investigate the antifungal activity and mechanisms of AgNPs against both

The TFAM-OGG1 axis mediates T-2 toxin-induced chondrocyte mitochondrial dysfunction and cartilage degeneration.

T-2 toxin, a potent trichothecene mycotoxin, exposes humans to its toxicity through chronic ingestion of contaminated food. As a key environmental risk factor for cartilage damage, its precise mechanism of disrupting mitochondrial homeostasis in chondrocytes remains unclear. This study employed transcriptome sequencing to identify core targets and validated findings using both in vitro and in vivo models. We demonstrate that T-2 toxin and its metabolite HT-2 toxin suppress the expression of mitochondrial transcription factor A (TFAM) and 8-oxoguanine DNA glycosylase-1 (OGG1). This suppression

Reshaping Tumor-Lymph Node Immune Axis via Targeted Lymphatic Delivery of Dual-Functional Immune Modulator for Enhanced Cancer Immunotherapy.

The efficacy of cancer immunotherapy is often limited by the immunosuppressive tumor microenvironment (TME) and insufficient immune activation in tumor-draining lymph nodes (TDLN). Since the TME and TDLN form a dynamic axis crucial for tumor metastasis and resistance to immune checkpoint blockade, strategies that effectively modulate both sites are critical. Here, we present a dissolving microneedle (MN) system that generates nanomicelles (NMCs) for localized delivery of a newly identified dual-functional macrocyclic trichothecene, Roridin E (R.E). R.E induces cancer cell-autonomous secretion

Experimental lung mycotoxicosis in mice induced by Stachybotrys atra.

Stachybotrys atra is often isolated from building materials in houses with moisture problems. Spores of S. atra can contain mycotoxins which may lead to various symptoms in exposed residents in damp houses. The pathogenesis of S. atra-induced lung diseases has not been elucidated. The purpose of the present study was to investigate lung mycotoxicosis experimentally in mice after an intranasal exposure to spores of S. atra-fungus. One group of mice received one intranasal injection of spores of a toxic strain of S. atra (1 x 10(6) spores) and the other group spores of a less toxic strain. Spore

Chlorogenic Acid Modulates PINK1-Parkin-Mediated Mitochondrial Quality Control in T-2 Toxin-Induced Thymic Immunotoxicity.

T-2 toxin (T-2), a potent immunotoxic trichothecene, induces severe thymic injury and immune dysfunction. Building on our identification of the ROS-NF-κB-NLRP3 axis, mitochondrial involvement remains poorly understood. Using murine and thymic epithelial cell models, we show that T-2 induces mitochondrial damage, activating PINK1-Parkin-mediated mitophagy that is adaptive but insufficient to maintain mitochondrial homeostasis. Inhibition of mitophagy by 3-methyladenine or Parkin silencing further compromised mitochondrial membrane potential by ∼34% and ∼22% versus T-2, respectively, accompanied

The Effects of Selenium on Wheat Fusarium Head Blight and DON Accumulation Were Selenium Compound-Dependent.

Fusarium head blight (FHB) caused by Fusarium graminearum not only results in severe yield losses, but also contaminates wheat grains with deoxynivalenol (DON) toxins. Prevention and control of FHB and DON contamination rely mainly on resistant varieties and fungicides. Selenium (Se) is an essential element for humans and animals, and also a beneficial element for plants. In this work, four Se compounds, i.e., sodium selenite (Na2SeO3), sodium selenate (Na2SeO4), selenomethionine (SeMet) and selenocysteine (SeCys2), were supplemented in a trichothecene biosynthesis induction (TBI) solid medium

T-2 toxin impairment of murine response to Salmonella typhimurium: a histopathologic assessment.

T-2 toxin and other trichothecene mycotoxins experimentally impair normal immune function and may predispose humans and animals to infectious disease. In this study, the histopathologic effects of Salmonella typhimurium challenge concurrently with sublethal T-2 toxin exposure were examined in the Salmonella-resistant C3H/HeN mouse. Oral administration of T-2 toxin (1 mg/kg) every other day for 10 d had little effect on the tissues examined when compared to control animals. Mice challenged with S. typhimurium and then treated with T-2 toxin every other day for 10 d had markedly larger and more
