West Nile virus infection decreases fecundity of Culex tarsalis females.

West Nile virus (family Flaviviridae, genus Flavivirus, WNV) persistently infects many mosquito tissues, and it has been associated with cytopathological changes in midgut muscles and salivary glands. However, the effects of WNV infection on mosquito fitness (survival and reproduction) are not known. We conducted a life table study of individually housed female Culex tarsalis Coquillett. After an initial bloodmeal from a WNV-infected or uninfected chicken, mosquitoes were provided sucrose and offered weekly opportunities to feed on a hanging blood drop. WNV transmission status was determined b

Lipid metabolism, viral infection, and antiviral immunity: a new host-pathogen interface.

Viral infections pose significant challenges to global health. Lipid metabolism plays a crucial role in various biological processes, including cell membrane structure, signaling, and energy homeostasis. Recent studies have highlighted the intricate relationship between lipid metabolism and viral infections, revealing how viruses exploit host lipid pathways to facilitate their replication and assembly. This review aims to elucidate the mechanisms by which viruses manipulate lipid metabolism and the subsequent impact on antiviral immunity. We systematically analyze the biological basis of lipid

Targeting Host Dependency Factors: A Paradigm Shift in Antiviral Strategy Against RNA Viruses.

RNA viruses, such as SARS-CoV-2 and influenza, pose a persistent threat to global public health. Their high mutation rates undermine the effectiveness of conventional direct-acting antivirals (DAAs) and facilitate drug resistance. As obligate intracellular parasites, RNA viruses rely extensively on host cellular machinery and metabolic pathways throughout their life cycle. This dependency has prompted a strategic shift in antiviral research-from targeting the mutable virus to targeting relatively conserved host dependency factors (HDFs). In this review, we systematically analyze how RNA viruse

Histopathology of fungal diseases of the lung.

Fungal pneumonias can be a diagnostic problem. However, their recognition is important as they can pose a significant health risk, especially in the immunocompromised host. While many of these infections are accompanied by necrotizing or non-necrotizing granulomas, some might be characterized by cellular interstitial pneumonia, intra-alveolar frothy material or only minimal inflammatory change. Much of the tissue reaction is dependent on the immune status of the patient and the type of fungal organism. While many of the fungi can be identified in tissue, especially if using histochemical stain

Poxviruses and the evolution of host range and virulence.

Poxviruses as a group can infect a large number of animals. However, at the level of individual viruses, even closely related poxviruses display highly diverse host ranges and virulence. For example, variola virus, the causative agent of smallpox, is human-specific and highly virulent only to humans, whereas related cowpox viruses naturally infect a broad spectrum of animals and only cause relatively mild disease in humans. The successful replication of poxviruses depends on their effective manipulation of the host antiviral responses, at the cellular-, tissue- and species-specific levels, whi

High-Definition DIC Imaging Uncovers Transient Stages of Pathogen Infection Cycles on the Surface of Human Adult Stem Cell-Derived Intestinal Epithelium.

Interactions between individual pathogenic microbes and host tissues involve fast and dynamic processes that ultimately impact the outcome of infection. Using live-cell microscopy, these dynamics can be visualized to study, e.g., microbe motility, binding and invasion of host cells, and intrahost-cell survival. Such methodology typically employs confocal imaging of fluorescent tags in tumor-derived cell line infections on glass. This allows high-definition imaging but poorly reflects the host tissue's physiological architecture and may result in artifacts. We developed a method for live-cell i

Investigating Bacterial Invasion of Highly Differentiated Human Bronchial Epithelial Cell Barriers.

Confocal microscopy provides information on host-pathogen interactions. To examine infection at the respiratory epithelial barrier, primary bronchial cells differentiated at an air-liquid interface (ALI) can be imaged and reconstructed as a 3D model. In this chapter, we describe a method for immunostaining and fluorescence imaging of well-differentiated bronchial epithelial tissue that has been exposed to the cystic fibrosis respiratory pathogen Achromobacter xylosoxidans.

Pathogenesis versus symbiosis: a dual perspective on plant-microbe interactions.

Plants function as integral components of soil ecosystems, embedded within diverse microbial consortia that engage in continuous chemical and informational exchange. This molecular dialog governs how plants distinguish harmful, neutral, and beneficial microbes, transforming the rhizosphere into a dynamic signaling interface. Microbial interactions operate along a chemical continuum in which metabolites, volatiles, peptides, and other small molecules act across aqueous, lipidic, and chemically heterogeneous microenvironments to shape recognition and response. The physicochemical organization of

Hos2-mediated KatG1 decrotonylation orchestrates ROS scavenging in rice blast infection.

In plant-pathogen interaction, plants trigger reactive oxygen species (ROS) bursts to defend against pathogen invasion, while pathogens evolved antioxidant systems to scavenge ROS for infection. Our study reveals a regulatory mechanism of ROS adaptation and scavenging in the rice blast fungus Magnaporthe oryzae. We found that KatG1, a bifunctional catalase-peroxidase, acts as a ROS-responsive regulatory enzyme. During infection, the deacetylase Hos2 mediates the decrotonylation of KatG1 at K422, initiating the ROS clearance process. Decrotonylated KatG1 depolymerizes into monomers, interacts w

The hijacking of a wheat Exo70-like protein by a fungal pathogen triggers disease.

Tan spot is an economically important fungal disease of wheat that occurs globally. In this necrotrophic pathosystem, pathogen effectors are recognized by wheat genes in an inverse gene-for-gene manner, leading to the hijacking of host defense mechanisms and ultimately disease susceptibility. Here, we identified the tan spot susceptibility gene Tsc2 through positional cloning, mutagenesis, and transgenic complementation. Tsc2 encodes a diverged form of the exocyst subunit Exo70FX15 that localizes to the nucleus and cytoplasm and does not interact directly with the fungal effector. A 21‑nucleot

Enterovirus 2B protein: From viroporin to membrane stress sensor, mechanistic principles of multi-pathway regulation.

Enterovirus 2B is a highly conserved, functionally versatile, nonstructural protein. The classic viroporin model, which restricts its role to ion channel formation, cannot account for its interactions with host factors such as Bcl-2-associated X protein, interleukin enhancer-binding factor 2, and karyopherin subunit alpha 1, nor its functional divergence among enteroviruses. We propose a novel, testable membrane stress sensor hypothesis: the transmembrane helices of 2B sense lipid composition and pH changes, and through conformational dynamics, spatiotemporally coordinate calcium signaling, ce

Light wavelength-specific regulation of hypersensitive response dynamics in tobacco plants during non-host interaction with Pseudomonas syringae pv. tomato.

Light quality is a key environmental signal that integrates plant immune pathways, yet how specific wavelengths regulate the execution, timing, and efficiency of the hypersensitive response (HR) remains incompletely resolved. This study investigated how light spectral composition modulates HR dynamics and resistance outcomes during a non-host interaction. Tobacco leaves were inoculated with Pseudomonas syringae pv. tomato DC3000 (PstDC3000) and incubated under red (RL), green (GL), blue (BL), white light (WL), or darkness. Monochromatic light treatments distinctly reshaped HR progression. BL a

Altered juvenile hormone dynamics mediate earlier foraging in Nosema-infected honey bees.

Parasites can profoundly alter host behavior, yet the physiological mechanisms underlying these changes are often unclear. In honey bees (Apis mellifera), microsporidian infection has long been associated with precocious foraging, a life-history shift that can destabilize colony function. Here, we identify the endocrine mechanism linking infection to behavioral maturation. Using Nosema apis, we show that infected workers initiate foraging significantly earlier than controls across four colonies (hazard ratio=2.69, P<0.001). This behavioral acceleration was accompanied by a ∼1.7-fold increase i

The multifaceted roles of fibronectin in viral pathogenesis.

Fibronectin is a high molecular weight glycoprotein critical to numerous physiological and pathological processes, including extracellular matrix organization, wound healing, immune responses, and interactions with pathogens. Evidence from various viral systems, including dengue, influenza, hepatitis B, and HIV-1, indicates that targeting fibronectin or disrupting its interactions can notably affect outcomes of viral infections in vitro. Alterations in the expression of fibronectin are associated with immune evasion mechanisms, including the inhibition of interferon signaling, thereby enabling

Insect cypoviruses: advances in structural characterization, evolutionary genomics, and host interaction dynamics.

Cypoviruses are insect-specific, double-stranded RNA viruses belonging to the genus Cypovirus within the family Spinareoviridae. Cypoviruses primarily infect insects of the orders Lepidoptera, Diptera, and Hymenoptera. These viruses replicate in midgut epithelial cells, forming polyhedrin-based occlusion bodies. Cypovirus genomes typically consist of 10-16 linear double-stranded RNA (dsRNA) segments that encodes distinct viral proteins; however, the number of genomic segments may vary among species. Each genomic segment encodes a functionally specialized distinct viral protein, with high intra

Pro- and anti-viral functions of lipid droplets in infection at a glance.

Lipid droplets (LDs) were long regarded as passive storage organelles for neutral lipids as this is the most efficient way to store energy. Recently, however, they have emerged as highly dynamic and heterogenous organelles that act as central hubs of lipid metabolic pathways. LDs provide lipids for energy production, supply precursor metabolites for many lipid classes, incorporate lipids under lipotoxic insults and are interconnected with other organelles through membrane contact sites. Viruses and all other obligate intracellular pathogens rely on metabolic networks in the host cell - includi

Bacterial extracellular vesicles as multifaceted regulators of human diseases and emerging therapeutic platforms.

Outer membrane vesicles (OMVs) are nanoscale vesicles actively released by Gram-negative bacteria, and have become key mediators in bacterial physiology, host-pathogen interactions, and disease pathogenesis. The biogenesis of OMVs is a dynamic process, which results from membrane homeostasis imbalance, peptidoglycan remodeling, and stress responses. These mechanisms enable virulence factors, immune-regulatory molecules, and nucleic acids to be selectively or randomly loaded into vesicles. With their ability to penetrate barriers and spread throughout the body, OMVs are widely involved in the o…

Rasputin/G3BP mediates subversion of antiviral immunity by o'nyong-nyong virus in Anopheles coluzzii.

Cellular G3BP proteins are essential for alphavirus infection in both vertebrate and mosquito hosts, but the underlying mechanism of their proviral activity is poorly understood in any host. Whether the mosquito G3BP ortholog, Rasputin (Rin), interacts with host immunity to influence alphavirus infection has not been investigated, and anopheline mosquito interactions with arboviruses have been little studied. Here, we find that Rin silencing in Anopheles mosquitoes results in decreased ONNV infection levels, indicating a proviral activity for Anopheles Rin. We find that Rin function is require
