Iron deficiency triggered transcriptome changes in bread wheat.

A series of complex transport, storage and regulation mechanisms control iron metabolism and thereby maintain iron homeostasis in plants. Despite several studies on iron deficiency responses in different plant species, these mechanisms remain unclear in the allohexaploid wheat, which is the most widely cultivated commercial crop. We used RNA sequencing to reveal transcriptomic changes in the wheat flag leaves and roots, when subjected to iron limited conditions. We identified 5969 and 2591 differentially expressed genes (DEGs) in the flag leaves and roots, respectively. Genes involved in the s

[Comparative analysis of gene expression in tea plant (Camellia sinensis (L.) Kuntze) under low-temperature stress].

Low-temperature stress is one of the main factors limiting the distribution and reducing the yield of many subtropical crops, including the tea crop. Efficient breeding to develop frost-tolerant cultivars requires a reliable set of genetic markers for identifying resistance donors, and that is why it is necessary to reveal the specific genetic response in frost-tolerant genotypes in comparison with frost- susceptible ones. In this work, we performed a comparative analysis of the expression of 18 tea genes (ICE1, CBF1, DHN1, DHN2, DHN3, NAC17, NAC26, NAC30, bHLH7, bHLH43, P5CS, WRKY2, LOX1, LOX

Comparative transcriptomics identifies patterns of selection in roses.

BACKGROUND: Roses are important plants for human beings with pivotal economical and biological traits like continuous flowering, flower architecture, color and scent. Due to frequent hybridization and high genome heterozygosity, classification of roses and their relatives remains a big challenge. RESULTS: Here, to identify potential markers for phylogenetic reconstruction and to reveal the patterns of natural selection in roses, we generated sets of high quality and comprehensive reference transcriptomes for Rosa chinensis 'Old Blush' (OB) and R. wichuriana 'Basye's Thornless' (BT), two speci

Functional validation of EXLB1 in leaf and stomatal development in tobacco.

Leaf morphological traits are the key determinants of tobacco yield and stress responses, and cell wall synthesis and metabolism play an important role in leaf development. However, the underlying regulatory mechanism of cell wall on leaf morphology remains largely unknown in tobacco. Here, we compared two pairs of tobacco cultivars (DMC vs. DX3 and DMC vs. WM3) with distinct leaf traits, and identified a cell wall-related expansin gene, NtEXLB1, based on transcriptome data analysis. The homolog NbEXLB1 from N. benthamiana is highly expressed in the mature leaf, and the NbEXLB1 protein is loca

Feeding preference of Rhynchophorus ferrugineus (Oliver) (Coleoptera: Curculionidae) on different date palm cultivars and host biochemical responses to its infestation.

To counter the insect infestation, plants respond with wide-ranging and highly dynamic biochemical reactions. Of these, the anti-oxidative activity is poorly understood. The red palm weevil (RPW) Rhynchophorus ferrugineus (Oliver), one of the most widespread pests in Pakistan, prefers to infest date palm Phoenix dactylifera. Our present study investigated the feeding preference of RPW to 11 different date palm cultivars and the results suggested that the Hillawi cultivar was most preferred. Greater infestation rate, fecundity and hatching rate were also recorded from Hillawi and Mozawati than

Triploidy in Citrus Genotypes Improves Leaf Gas Exchange and Antioxidant Recovery From Water Deficit.

The triploidy has proved to be a powerful approach breeding programs, especially in Citrus since seedlessness is one of the main consumer expectations. Citrus plants face numerous abiotic stresses including water deficit, which negatively impact growth and crop yield. In this study, we evaluated the physiological and biochemical responses to water deficit and recovery capacity of new triploid hybrids, in comparison with diploid hybrids, their parents ("Fortune" mandarin and "Ellendale" tangor) and one clementine tree used as reference. The water deficit significantly decreased the relative wat

lpa1-5525: A New lpa1 Mutant Isolated in a Mutagenized Population by a Novel Non-Disrupting Screening Method.

Phytic acid, or myo-inositol 1,2,3,4,5,6-hexakisphosphate, is the main storage form of phosphorus in plants. It is localized in seeds, deposited as mixed salts of mineral cations in protein storage vacuoles; during germination, it is hydrolyzed by phytases to make available P together with all the other cations needed for seed germination. When seeds are used as food or feed, phytic acid and the bound cations are poorly bioavailable for human and monogastric livestock due to their lack of phytase activity. Therefore, reducing the amount of phytic acid is one strategy in breeding programs aimed

Response of wheat germplasm to infestation of English grain aphid (Hemiptera: Aphididae).

The English grain aphid, Sitobion avenae (F.), is a destructive insect pest of wheat. In this study, wheat germplasm was evaluated for resistance and tolerance to English grain aphid infestation. Except for spikelet number/spike, the 1,000-kernel weight, spike weight, grain weight/ spike, and sterile spikelet number/spike of infested plants were all significantly impacted by English grain aphid infestation. Of 292 wheat accessions, only one genotype showed English grain aphid high resistance, and most genotypes showed intermediate resistance. Tolerance index values ranged widely among all acce

[The occurrence of aflatoxins in some components and complete feed mixtures for pigs].

The feed components and complete feed mixtures used in pig breeding and fattening in June 1978 were examined qualitatively by the silica-gel thin-layer chromatographic method, and quantitatively by the method after Velasco. The object of the examination was the occurrence of aflatoxins. On the whole, 104 feed samples were taken and subjected to the laboratory examination; this total number included 77 samples (74.0%) coming from 24 farms and 27 samples (26.0%) from eight feed plants in the South Moravian region. The presence of aflatoxins was detected in 14 samples of the examined feeds, i. e.

Whole-genome analysis uncovers four novel Streptomyces species linked to sweet potato soil rot in China.

UNLABELLED: Sweet potato soil rot, caused by pathogenic Streptomyces, is an emerging threat to sweet potato production in China. However, the diversity and pathogenic mechanisms of the associated species remain poorly understood. In this study, symptomatic storage roots exhibiting typical pox-like lesions were collected from major production regions in Henan and Guangdong provinces. A total of 225 actinomycete strains were isolated using optimized methods, and six highly pathogenic strains were identified through tuber slice, hydroponic, and pot inoculation assays. Dissecting the genetics of forage quality traits in soft red winter wheat in the U.S. southeast region.

Winter wheat plays a viable role in agriculture, not only as a primary grain crop but also as a valuable forage source that bridges fall-spring forage gaps in many regions, including the southeastern (SE) U.S. Despite its nutritive potential, genetic basis of forage-quality traits remains insufficiently understood, limiting breeding efforts for dual-purpose cultivars. This study aimed to dissect the genetic architecture of forage quality in 182 soft red winter wheat (SRWW) genotypes adapted to the SE U.S. using genome-wide association study (GWAS). Field experiments were carried out in randomi

A two-stage screening approach integrated with GWAS for rare phenotypes such as spontaneous haploid genome doubling in maize.

Spontaneous haploid genome doubling (SHGD) is a valuable trait in maize breeding, enabling the development of doubled haploid (DH) lines without chemical chromosome doubling. However, SHGD is a rare phenotype, expressed in only a small fraction of maize germplasm, making its identification resource-intensive. This study evaluated the efficiency of a two-stage field screening approach designed to identify maize genotypes with high haploid male fertility (HMF), a key indicator of SHGD potential. Simulation analyses showed that evaluating 50 haploid plants per genotype, combined with a 25% HMF th

Assembly and maturation of calf gut microbiome from neonate to post-puberty.

With the help of rumen bacteria, ruminants can feed on indigestible plant materials and produce over 70% of their energy as fatty acids. However, during lactation, ruminants exhibit characteristics of monogastric animals due to an undeveloped rumen; therefore, understanding gut microbiome changes in growing calves is essential. Our understanding of the gut microbiome in growing calves remains limited in large populations with the same diet, breed, and period. Here, we describe 16S rRNA gene amplicon sequencing data from 420 faecal samples, 20 rumen contents, 17 small intestine contents, and 18

Jasmonic acid and nitric oxide orchestrate a hierarchical melatonin cascade for Botrytis cinerea resistance in tomato.

Tomato gray mold, caused by Botrytis cinerea (B. cinerea), poses a major challenge to tomato production. In plants, B. cinerea resistance is positively regulated by melatonin; however, the regulatory mechanism of melatonin biosynthesis during B. cinerea infection is not known. Here, we established the working model of jasmonic acid (JA) and nitric oxide (NO) on melatonin biosynthesis in the state of B. cinerea infection. NO responded to B. cinerea infection earlier than JA. In the early stage of infection, tomato caffeic acid O-methyltransferase 2 (SlCOMT2) was S-nitrosylated by NO at Cys344,

Performance of the Methods Used to Reflect the High-Temperature Stress in Crops.

High-temperature stress is one of the most important factors threatening crop productivity in the world. Breeding thermo-tolerant crops and developing cultural practices that can alleviate stress is the most effective and fundamental approach to tackling warmer temperatures. In order to test stress-tolerant practices, creating a realistic stress environment during the period when plants are most sensitive affects success. High-temperature stress trials are carried out in plastic tunnels, sowing time regulation, infrared heating (Free Air Temperature Elevation), greenhouse applications, and cli

AsAP2 transcriptionally activates ferulate 5-hydroxylase, diverting ferulic acid metabolism toward lignin biosynthesis in Angelica sinensis.

Premature flowering in Angelica sinensis (Danggui) triggers severe declines in root yield and medicinal quality5 by reducing bioactive ferulic acid and promoting lignification. Ferulate 5-hydroxylase (F5H), a cytochrome P450 enzyme, drives metabolic flux toward lignin biosynthesis, but its regulatory network and functional dynamics during early flowering remain unresolved. Transcriptomic and functional analyses identified AsF5H (As09G05225) as a key gene upregulated in early-flowering plants, correlating with root lignin accumulation and ferulic acid depletion. Phylogenetic studies confirmed F

Eleven biosynthetic genes explain the majority of natural variation in carotenoid levels in maize grain.

Vitamin A deficiency remains prevalent in parts of Asia, Latin America, and sub-Saharan Africa where maize (Zea mays) is a food staple. Extensive natural variation exists for carotenoids in maize grain. Here, to understand its genetic basis, we conducted a joint linkage and genome-wide association study of the US maize nested association mapping panel. Eleven of the 44 detected quantitative trait loci (QTL) were resolved to individual genes. Six of these were correlated expression and effect QTL (ceeQTL), showing strong correlations between RNA-seq expression abundances and QTL allelic effect
