Plasma prolactin levels in maturing intact and cryptorchid male rats: development of stress response.

In order to determine the possible role of the seminiferous tubules in the regulation of prolactin secretion during sexual development, male rats were rendered cryptorchid at 22 days of age, and thereafter different groups of animals were decapitated at 8-10 day intervals between Day 32 and Day 70. Cryptorchid rats showed destruction of the germinal epithelium accompanied by increased plasma FSH and, to a much lesser extent, increased plasma LH titers. Nevertheless, plasma prolactin levels were similar to those of intact controls throughout the entire period studied. Plasma prolactin titers in…

The usefulness of cannulated prolactin test in mild hyperprolactinemia: case series and systematic review of the literature.

PURPOSE: Mild hyperprolactinemia can occur due to venipuncture-related stress leading to unnecessary diagnostic investigations/treatment. We aimed to assess the cannulated prolactin testing (CPT) usefulness in the investigation of mild hyperprolactinemia by evaluating our case series and conducting a literature review. METHODS: We conducted a retrospective study including hyperprolactinemia patients on an initial measurement (referral prolactin (rPRL)), who underwent a CPT between 2018–2025. During CPT a catheter was inserted and samples for prolactin were drawn at baseline (PRL0’), 30 min and…

The Emerging Role of Prolactin as a Biomarker for Reproduction, Health, and Welfare in Wildlife Species: Elephants as a Model.

Prolactin is a key hormone that plays significant roles in reproduction, extending far beyond its well-known function in mammalian lactation. It is vital for reproductive success across diverse taxa, including birds, fish, reptiles, and amphibians, where it regulates critical reproductive behaviors and physiological processes like parental care, pregnancy maintenance, embryonic diapause, immunity, osmoregulation, metamorphosis, and molting. Environmental cues often fine-tune its actions to ensure reproduction occurs under optimal conditions. Conversely, dysregulation of prolactin can severely …

Effect and mechanism of maternal prolactin levels on depressive-like behavior in prenatally stressed offspring.

Prenatal stress (PS) has an adverse impact on the neurodevelopment of the fetus and increases the risk of depression in offspring. Prolactin (PRL), a polypeptide hormone released by the pituitary gland, has a promising neuroprotective effect. However, it is not clear whether the decline in PRL levels during gestation is related to the depressive-like behavior of offspring caused by PS. This study aimed to assess the role and mechanism of PRL during gestation on depressive-like behavior in PS offspring rats. We found that PS led to a marked decrease in serum PRL levels of the maternal rats, acc…

Emerging Function of Prolactin-Inducible Protein-Is This Important Tear Protein Found in Alzheimer's Disease?

Alzheimer's disease is characterized by a chronic, long-term neurodegenerative process and an increasing need for easily accessible biomarkers that enable early diagnosis and disease monitoring. For this reason, tears have attracted growing interest as a potential source of such biomarkers, and prolactin-inducible protein is a candidate tear protein of mechanistic interest whose clinical value remains to be established as a biomarker of Alzheimer's disease. The literature indicates that prolactin-inducible protein is physiologically present in the lacrimal apparatus. Proteomic studies in patie…

AQP5 trafficking is regulated by its C-terminal tail and interaction with prolactin-inducible protein.

BACKGROUND: Aquaporin-5 (AQP5) is a crucial membrane protein involved in water transport across cellular membranes, particularly within exocrine glands such as salivary glands. Dysregulation of AQP5, including its mislocalization, has been associated with various diseases, emphasizing the need to understand the molecular mechanisms governing its trafficking. This study investigates the multifaceted regulatory mechanisms of AQP5 trafficking, with specific emphasis on the role of the carboxyl-terminal (C-terminal) tail and the functional involvement of prolactin-inducible protein (PIP) as an int…

Macrophages and iNOS contribute to the effects of dural prolactin and repeated stress in mouse migraine models.

BACKGROUND: Migraine impacts 15% of the global population, predominantly women. Previous studies have shown a role for prolactin in animal migraine models induced by either stimulation of the dura mater or repeated stress exposure. However, the site of prolactin action is not fully known nor are its downstream mechanisms. This study investigated the potential downstream mechanisms and the cell types involved in prolactin- and repeated stress-induced migraine-like responses. METHODS: Two preclinical migraine models were used in this study, dural stimulation and repeated restraint stress. Dural…

Inverse dose-dependent effects of aripiprazole on sexual dysfunction and prolactin levels in patients with schizophrenia.

Aripiprazole, with its distinct pharmacodynamic profile, acts as a partial agonist at dopamine D2 and serotonin 5-HT1A receptors and an antagonist at 5-HT2A receptors, offering a superior safety profile compared to other antipsychotic drugs. The aim of this study was to assess the dose-dependent effects of aripiprazole on sexual dysfunction and prolactin levels in patients with schizophrenia. Patients receiving aripiprazole were recruited for this study. Psychopathology was assessed using the Positive and Negative Syndrome Scale (PANSS), and sexual dysfunction was evaluated with the Arizona Se…

The contribution of prolactin in the occurrence of premature cardiovascular disease predominantly through modulating the inflammatory cascade.

Cardiovascular disease, mainly coronary atherothrombosis, is an essential contributor to all-cause mortality, accounting for approximately 30% of all global deaths. Imbalances in hormone profiles may play a role in the evolution of atherothrombosis. In the last decade, our clinical research focused on the hormone prolactin (PRL) and its contribution to premature cardiovascular disease. In this narrative review, we aimed to give current insights into how PRL could modulate the inflammatory cascade within the microenvironment of the atherosclerotic plaque, which could lead to increased cardiovas…

Macroprolactin over time: Is there any point in rechecking it in people with a persistently elevated serum prolactin?

OBJECTIVE AND DESIGN: Macroprolactinemia may influence the interpretation of serum prolactin levels-a recognised phenomenon since 1981. The degree of macroprolactinaemia over time is less well described. We determined how macroprolactin status (based on polyethylene glycol (PEG) precipitation) varied by analysing serial measurements in hyperprolactinaemic individuals over a period of 9 years. PATIENTS AND MEASUREMENTS: Results from 1810 individuals were included. All serum total prolactin results (measured using Roche Cobas 8000 analyser) were extracted from the laboratory information system …

Prolactin Secreting Pituitary Carcinoma and the Role of Peptide Receptor Radionuclide Therapy: A Brief Report.

Pituitary carcinoma is a rare entity comprising 0.1-0.2% of all pituitary tumors and presents significant diagnostic and therapeutic challenges. Intraspinal drop metastasis in these tumors is even rarer. We report a case of a prolactin secreting pituitary carcinoma with intracranial metastasis and multiple intraspinal drop metastasis. This is the first case where 68Gallium labelled [1,4,7,10 - tetraazacyclododecane - 1,4,7,10 - tetraacetic acid] -1- NaI3 - octreotide (68Ga-DOTANOC) whole-body positron emission tomography-computed tomography (PET-CT) has been used in a case of malignant prolact…

The Ser434Phe Androgen Receptor Gene Mutation Does Not Affect Fertility but is Associated with Increased Prolactin.

INTRODUCTION: Prolactin is a hormone secreted by the anterior pituitary gland essential for lactation. Non-physiological hyperprolactinemia characterized by serum prolactin levels exceeding 20 ng/mL in men and 25 ng/mL in women, often results from medication use or pituitary gland tumors. In a minority of cases, the cause of hyperprolactinemia remains unknown despite clinical investigations. Familial idiopathic hyperprolactinemia may stem from mutations in genes encoding prolactin (PRL) and its receptor (PRLR). METHODS: This study investigated genetic polymorphisms in PRL and PRLR genes using…

The Hook Effect: A Case Study of a Giant Invasive Prolactinoma With Falsely Low Serum Prolactin.

Prolactinomas are benign pituitary tumors also known as prolactin-secreting adenomas (PSA). These tumors cause excessive secretion of prolactin (hyperprolactinemia), a hormone responsible for lactation. Diagnosing hyperprolactinemia relies on measuring prolactin levels in the blood, and elevated serum levels of prolactin are typically indicative of prolactinoma. The hook effect occurs in immunological tests such as the prolactin level test. When the amount of prolactin present in the sample is too high and exceeds the binding capacity of the antibodies being used, the test result may indicate …

Involvement of neurosteroids in the control of prolactin secretion in sheep under basal, stressful and pregnancy conditions.

Prolactin (PRL) secretion by the anterior pituitary (AP) is responsive to changes in physiological conditions and many external factors that also affect brain neurosteroid levels. This study tested the hypothesis that neurosteroids can affect PRL secretion in sheep under basal, stressful and advanced pregnancy conditions. In Experiment 1, luteal-phase sheep were subjected to a three-day series of intracerebroventricular (icv.) control (n = 12) or allopregnanolone (AL, 4 × 15 μg/60 μL/30 min at 30-min intervals, n = 12) infusions. Acute stressful stimuli, isolation and partial movement restrict…

Immunohistochemical localization of prolactin receptor (PRLR) to Hodgkin's and Reed-Sternberg cells of Hodgkin's lymphoma.

Prolactin receptor (PRLR), a type-1 cytokine receptor, is overexpressed in a number of cancer types. It has attracted much attention for putative pro-oncogenic roles, which however, remains controversial in some malignancies. In this study, we reported the localization of PRLR to the Hodgkin's and Reed-Sternberg (HRS) cells of Hodgkin's lymphoma (HL), a neoplasm of predominantly B cell origin. Immunohistochemistry performed on 5-μm thick FFPE sections revealed expression of PRLR in HRS cells. Cellular immunofluorescence experiments showed that the HL-derived cell lines, Hs445, KMH2 and L428 ov…
