Spontaneous intravenous catheter fracture and embolization from an implanted venous access port and analysis by scanning electron microscopy.

A spontaneous fracture occurred in the midportion of a catheter that was indwelling in the left subclavian vein and embolized to the pulmonary artery. Scanning electron microscopy studies were performed upon the fractured catheter. Possible reasons for the fracture are discussed. Sudden chest pain in a patient with an implanted catheter may be the first clinical manifestation of an embolized catheter.

Mouse DNA-cytosine-5-methyltransferase: sequence specificity of the methylation reaction and electron microscopy of enzyme-DNA complexes.

Monoclonal antibodies prepared against DNA methyltransferase from human placenta undergo immune complex formation also with DNA methyltransferase from P815 mouse mastocytoma cells. One of these monoclonal antibodies, M2B10, was used for the immunoaffinity purification of this enzyme. Complexes of the immunoaffinity-purified mouse DNA methyltransferase with DNA were visualized by electron microscopy. DNA methyltransferase was found to be distributed along linearized plasmid DNA with a higher incidence of enzyme molecules at the terminal segments. This binding to strand ends was significantly in

Pityriasis rubra pilaris: a clinico-pathological and therapeutic study with special reference to histochemistry, autoradiography, and electron microscopy.

Five patients with pityriasis rubra pilaris (PRP) were analyzed by means of light and electron microscopy as well as by histochemistry and autoradiography. The results were compared with findings in psoriasis vulgaris. In PRP we found a moderate increase of the labeling index of epidermal cells, a highly increased labeling index of dermal infiltrating cells, and a mild spongiosis, and in the stratum granulosum, a decreased number of tonofilaments and an increased number of keratinosomes. The horny layer in PRP showed a pronounced histochemical and electron microscopical parakeratosis, even whe

South American pemphigus foliaceus: electron microscopy and immunoelectron localization of bound immunoglobulin in the skin and oral mucosa.

Five skin and two oral biopsies from patients with South American pemphigus foliaceus (SAPF) were studied by electron and immunoelectron microscopy for the ultrastructural localization of bound immunoglobulin in epidermal and oral lesions. Electron microscopy showed the tonofilament-desmosome complex to be preserved in the various layers of the epidermis. Immunoglobulin was bound over the plasma membrane and permeated the desmosomal junctions both in the skin and oral mucosa, thus suggesting that pemphigus antibodies are attached to the glycocalyx. It appears that the initial injury in SAPF ac

Fetal hypothalamic transplants in the third ventricle of the adult rat brain. Correlative scanning and transmission electron microscopy.

Blocks of anterior hypothalamus were transplanted from 19 day-old fetuses of Wistar/Lewis rats into the third ventricle of adult male Brattleboro rats. Physiological changes in graft recipients and in sham-operated animals were monitored daily. Twenty days after surgery, the graft recipients and sham-operated animals were killed and their brains examined by correlative scanning and transmission electron microscopy. Host animals that exhibited both decreased polydipsia and increased urine concentration were found to have viable grafts within the third ventricle. The observed physiological chang

Immunopathology of NZB/BL mice. V. Viruslike (filtrable) agent separable from lymphoma cells and identifiable by electron microscopy.

A filtrable agent separable from transplantable malignant lymphomas of NZB/Bl mice was capable, upon inoculation into preweanling NZB/Bl mice, of inducing lymphoid cell hyperplasia, hypergammaglobulinemia, proteinuria, hypoalbuminemia, and pathological changes (focal membranous glomerulonephritis) of kidneys-renal functional and structural changes qualitatively similar to those of spontaneous occurrence in older NZB/Bl mice. Viruslike particles with close resemblance to the typical "C" type murine oncogenic virus particles were identified by means of electron microscopy in NZB/Bl mouse tissues

Visualising redox-driven metal-oxide interface dynamics in Ni/GDC electrodes via in situ Auger-Meitner electron microscopy.

Metal/oxide interfaces play a decisive role in both high-temperature electrochemical energy conversion using solid oxide cells (SOCs) and heterogeneous catalysis. However, their morphological stability under dynamic redox conditions remains poorly understood. In the present work, we investigate model electrodes of Ni/GDC (GDC = Gd-doped ceria = Gd0.1Ce0.9O2-δ ) composites in thin-film and porous form. Electrochemical polarization of these electrodes enables electrochemical control of the oxygen chemical potential in ceria and thus provides direct experimental access to redox-driven interface d

Ultrastructural characterization of secretory canals in Peucedanum praeruptorum roots using microscopic sectioning, transmission electron microscopy, and computed tomography.

Plant secondary metabolites are mainly synthesized and stored in secretory tissues. Secretory canal development has been mainly characterized in Apiaceae. The secretory canals of Peucedanum praeruptorum contain pharmacologically active coumarins, but their organ-specific distribution and developmental dynamics remain poorly understood. This study integrated light microscopy (LM), transmission electron microscopy (TEM), X-ray microcomputed tomography (µ-CT), and high-performance liquid chromatography (HPLC) to investigate canal development, distribution, ultrastructure, 3D architecture, and cou

Protocol for fixation and staining of postmortem human brain tissue optimized for focused ion beam-scanning electron microscopy.

Here, we present a protocol for generating postmortem human brain tissue samples suitable for volume electron microscopy (vEM) imaging via focused ion beam-scanning electron microscopy (FIB-SEM). We describe steps for immersion fixation, sectioning, long-term cryopreservation, and enhanced-contrast staining that produce samples suitable for high-resolution FIB-SEM. This approach generates ultrastructure of sufficiently high preservation to enable both dense and targeted three-dimensional (3D) reconstructions of cellular, subcellular, synaptic, and organellar features in postmortem human brain

PdGa Alloy Dynamics under CO2 Hydrogenation from Surface Organometallic Chemistry on a Chip and Operando Transmission Electron Microscopy.

Industrial processes rely on the use of promoter elements to improve the performance of catalysts. Despite considerable efforts, the fundamental understanding of the origin of promotion remains elusive, partly due to the inherent complexity of catalytic systems. In this work, we develop a surface organometallic chemistry approach on an in situ MEMS chip for the preparation of tailored PdGa nanoparticles enabling to monitor the dynamic response of their structure and reactivity under a range of conditions ─ including CO2 hydrogenation ─ via operando (scanning) transmission electron microscopy.

Ultrastructural and elemental characterization of gallstones in patients ≤40 years: a scanning electron microscopy study with histopathology correlation.

Gallstone disease affects individuals of different ages, yet age-related variations in stone morphology, histology, and chemical composition remain insufficiently defined. Understanding these patterns may clarify pathogenic mechanisms and their potential links to clinical or metabolic profiles. This study investigated age-related differences in gallstone characteristics. Thirty-seven gallstones were analyzed and grouped into three age categories (<20, 21-30, and 31-40 years), with additional stratification by sex. Morphological classification was performed macroscopically and by scanning elect

Comparative electron microscopy analysis of internal limiting membrane and epiretinal membrane ultrastructure from vitrectomy surgery: A study protocol.

The epiretinal membrane (ERM) is a fibrocellular layer that forms on the inner surface of the retina, often leading to visual impairment and significantly impacting visual function. Understanding the pathophysiology of ERM formation is crucial for advancing ophthalmic care and improving patient outcomes. This research aims to investigate the pathophysiology of ERM by comparing the internal limiting membrane (ILM) pathology in patients with and without ERM. The study involves a comprehensive protocol of ILM and ERM specimens collection during pars plana vitrectomy and membrane peeling procedure

Development and validation of an interface for automated image acquisition during high-temperature environmental scanning electron microscopy experiments.

An interface that enables automatic image acquisition during high-temperature experiments in an environmental SEM is developed. It is optimized to work on multiple regions of interest at multiple magnifications, performing image focusing (focus and astigmatism) and automatic re-centering of regions of interest. Its operation has been validated by monitoring two regions of interest of a nickel-based superalloy undergoing oxidation at 950 °C at different magnifications. Recording series of images at different magnifications on different regions of interest makes it possible to qualify the behavi

Epigenetic Modifier Drug Valproic Acid Enhances Cancer Metaphase Chromosome Elasticity and Electron Transport: An Atomic Force Microscopy Approach.

The structural integrity of the chromosomes is essential to every functional process within eukaryotic nuclei. Chromosomes are DNA-histone complexes that are essential for the inheritance of genetic information to the offspring, and any defect in them is linked to mitotic errors, cancer growth, and cellular aging. Changes in the mechanical properties of a chromosome could lead to its compromised function and stability, leading to chromosome breaks. Here, we studied the changes in chromosome physical properties using metaphase chromosomes isolated from moderately malignant (MCF7) and highly mal

Pathological characterization of hepatic and mesenteric neotropical Echinococcosis in Brazilian Amazonian patients using light and scanning electron microscopy.

Neotropical Echinococcosis (NE) is an emerging parasitic zoonosis of significant public health concern. The disease is caused by the larval stage (metacestodes) of Echinococcus vogeli and affects humans in tropical forests of Central and South America. While clinical presentation, radiological imaging, and surgical procedures have been investigated, the pathological features of NE remain partially understood. We performed a comparative study of hepatic and mesenteric metacestodes obtained from Brazilian patients in Acre and Amazonas States during surgical procedures, using both light and scann

Large-field-of-view scanning electron microscopy of the paraventricular nucleus of the hypothalamus during diet-induced obesity.

Dysregulation in the paraventricular nucleus of the hypothalamus (PVN) is associated with a variety of diseases including those related to obesity. Although most investigations have focused on molecular changes, structural alterations in PVN neurons can reveal underlying functional disruptions. Although electron microscopy (EM) can provide nanometer resolution of brain structures, an inherent limitation of traditional transmission EM is the single field of view nature of data collection. To overcome this, we used large-field-of-view high-resolution backscatter scanning electron microscopy (bSE
