The language of the immune system: Part 1: Cytokine conversations and conversationalists.

The immune system is always acting to protect the body from the microbial world, especially at the openings of the body but also helps to maintain homeostasis. Normal functions include self-regulation and regeneration, facilitating the maintenance of organs, skin and mucosal membranes, protecting the openings of the body, providing surveillance for abnormal and cancer cells and eliminating molecular and cellular debris. In addition to status quo maintenance functions, the immune system responds to damage, disruption or danger when challenged by trauma, infection, cancer, autoimmunity and other

Mitochondrial transfer and transplantation in the immune cells: Mechanistic foundations, medical applications, and future prospects.

Mitochondria exhibit tissue-specific physiological functions and are central to the maintenance of cellular homeostasis. Emerging evidence indicates that intercellular mitochondrial transfer is regulated by multiple determinants and exerts a profound influence on the function of both innate and adaptive immune cells. The underlying mechanisms are highly heterogeneous, involving distinct cellular contexts, microenvironmental cues, and modes of intercellular communication. This review summarizes the major triggers and mechanistic pathways governing mitochondrial transfer in immune cells and immu

Emergence of curvature-dependent tissue growth from cell crowding and mechanical interactions.

Many biological tissues grow at rates that depend strongly on the local geometry of the tissue interface, yet the cellular mechanisms underlying this dependence remain poorly understood. In this work, we develop a two-dimensional mathematical model of tissue growth to investigate how curvature-dependent growth emerges from the complex interactions between cell-scale crowding and mechanical interactions. The tissue interface is represented as a confluent layer of mechanically interacting cells that simultaneously produce new tissue. We formally derive a continuum limit of this discrete model, o

A flashing success: A community-engaged approach to finding western U.S. fireflies.

Fireflies are well-documented in many regions of the USA. However, the presence of flashing fireflies in much of the western United States remains largely unknown. Leveraging citizen science, this study aimed to locate populations of bioluminescent fireflies across Utah. Through a multifaceted public outreach campaign and an interactive website for data collection, citizen scientists were invited to report firefly sightings. Over nearly a decade, the project successfully amassed reports from diverse locations, significantly expanding the known distribution of bioluminescent fireflies in Utah a

Nanosystem-Mediated Intercellular Crosstalk Modulation in the Tumor Microenvironment for Enhanced Cancer Immunotherapy.

Intercellular crosstalk in the tumor microenvironment (TME) is a central determinant of cancer progression and therapy response, in which malignant, immune, stromal and vascular compartments exchange information mainly through extracellular vesicles, signaling pathways, and metabolic cues. These communication circuits can enforce immune exclusion, sustain immunosuppressive cell states and enable adaptive resistance, thereby limiting the durability of otherwise effective therapies. Nanosystems have emerged as enabling tools to modulate this crosstalk across major tumor-centered interaction axes

Cell-Cell Interactome-Based Pathogenesis and Therapies for Osteosarcoma.

Osteosarcoma (OS), the most common primary malignant bone tumor in children and young adults, is characterized by aggressive behavior, frequent metastasis, and resistance to chemotherapy, resulting in poor clinical outcomes. Increasing evidence indicates that OS progression is not solely driven by tumor-intrinsic factors but is strongly influenced by dynamic interactions within the tumor microenvironment (TME). This literature review synthesizes current research on the roles of endothelial cells, fibroblasts, mesenchymal stromal cells, immune populations, and osteoclasts in OS pathogenesis, wi

Development of the neurohypophysis: A major neuroendocrine interface.

The neurohypophysis is a major central neuroendocrine interface regulating reproductive functions and water homeostasis. Distinct neurovascular cell types interact via evolutionarily conserved signaling molecules in the developing neurohypophysis, providing a model system for studying principles in neuroendocrine interface morphogenesis. This review provides an overview of neurohypophysis development with a focus on paracrine signaling and the intrinsic mechanisms that regulate the major cell types and neurovascular interface development.

Frozen in Place: Proximity Labeling Maps Glial Interactomes Across Cell States.

Glial cells, including radial glia, oligodendrocyte precursor cells (OPCs), oligodendrocytes, astrocytes, and microglia, are active and dynamic regulators of central nervous system (CNS) development, homeostasis, and disease. Through extensive interactions with neurons, other glial populations, and the vasculature, they form highly specialized communication networks that are essential for normal brain function. While transcriptomic approaches have revealed extensive glial heterogeneity and enabled the prediction of putative signaling networks, a critical challenge remains in validating and tra

Secreted phospholipase A2 regulates intercellular communications by coordinating extracellular phospholipid metabolism.

Lipids play fundamental roles in life. In essence, "phospholipase A2" (PLA2) indicates a group of enzymes that release fatty acids and lysophospholipids by hydrolyzing the sn-2 position of glycerophospholipids. To date, more than 50 enzymes that possess PLA2 or related lipid-metabolizing activities have been identified in mammals and are subdivided into several families in terms of their structures, catalytic mechanisms, tissue/cellular localizations, and evolutionary relationships. Among the PLA2 superfamily, the secreted PLA2 (sPLA2) family contains 11 isoforms in mammals, each of which has

Novel insights into osteocyte and inter-organ/tissue crosstalk.

Osteocyte, a cell type living within the mineralized bone matrix and connected to each other by means of numerous dendrites, appears to play a major role in body homeostasis. Benefiting from the maturation of osteocyte extraction and culture technique, many cross-sectional studies have been conducted as a subject of intense research in recent years, illustrating the osteocyte-organ/tissue communication not only mechanically but also biochemically. The present review comprehensively evaluates the new research work on the possible crosstalk between osteocyte and closely situated or remote vital

Beyond Macromolecules: Extracellular Vesicles as Regulators of Inflammatory Diseases.

Inflammation is the defense mechanism of the immune system against harmful stimuli such as pathogens, toxic compounds, damaged cells, radiation, etc., and is characterized by tissue redness, swelling, heat generation, pain, and loss of tissue functions. Inflammation is essential in the recruitment of immune cells at the site of infection, which not only aids in the elimination of the cause, but also initiates the healing process. However, prolonged inflammation often brings about several chronic inflammatory disorders; hence, a balance between the pro- and anti-inflammatory responses is essent

The Cellular Mechanism of Acupuncture for Ulcerative Colitis based on the Communication of Telocytes.

Acupuncture can ameliorate or treat diseases according to the meridian theory in traditional Chinese medicine (TCM); however, its mechanism has not been scientifically clarified. On the other hand, telocytes (TCs) are morphologically in accordance with the meridian system, which needs further cytological investigations and acupuncture confirmation. The present study showed that acupuncture could activate TCs in several ways, alleviating rabbit ulcerative colitis. TCs could cytologically communicate the acupoints, the acupuncture sites in skin with their corresponding large intestine by TC homo

Neuron-glia Interaction as a Possible Pathophysiological Mechanism of Bipolar Disorder.

Accumulating evidence has shown the importance of glial cells in the neurobiology of bipolar disorder. Activated microglia and inflammatory cytokines have been pointed out as potential biomarkers of bipolar disorder. Indeed, recent studies have shown that bipolar disorder involves microglial activation in the hippocampus and alterations in peripheral cytokines, suggesting a potential link between neuroinflammation and peripheral toxicity. These abnormalities may also be the biological underpinnings of outcomes related to neuroprogression, such as cognitive impairment and brain changes. Additio

Stellate Cell Networks in the Teleost Pituitary.

The folliculostellate cells of the mammalian pituitary are non-endocrine cells that are implicated in long-distance communication and paracrine signaling, but to date, these cells have yet to be characterized in teleosts. We found that the stellate cells of the teleost pituitary share many common attributes with mammalian folliculostellate cells. By labeling of stellate cells in live preparations of tilapia pituitaries we investigated their distribution, association with other endocrine cells and their anatomical and functional coupling. In the pars intermedia, stellate cells were arranged aro
