AChE inhibitory alkaloids from Coptis chinensis.

Coptis chinensis is a widely used traditional Chinese herbal medicine. In this work, 6 new alkaloids (coptisine A-F, 1-6) and 26 known alkaloids (7-32) were isolated from the chloroform extract of the rhizomes of C. chinensis. Compounds 1-3 are α-carbonylated benzylisoquinolines, and 4-6 are berberidic acid type alkaloids. Their structures were elucidated on the basis of extensive NMR and MS analyses. Seven compounds (7, 20, 22, 23, 25, 26, 29) exhibited significant AChE inhibitory activities at 10 μM (inhibition rates >80%).

Functional analysis of norcoclaurine synthase in Coptis japonica.

(S)-Norcoclaurine is the entry compound in benzylisoquinoline alkaloid biosynthesis and is produced by the condensation of dopamine and 4-hydroxyphenylacetaldehyde (4-HPAA) by norcoclaurine synthase (NCS) (EC 4.2.1.78). Although cDNA of the pathogenesis-related (PR) 10 family, the translation product of which catalyzes NCS reaction, has been isolated from Thalictrum flavum, its detailed enzymological properties have not yet been characterized. We report here that a distinct cDNA isolated from Coptis japonica (CjNCS1) also catalyzed NCS reaction as well as a PR10 homologue of C. japonica (CjPR1

Coptis chinensis Polysaccharides Inhibit Advanced Glycation End Product Formation.

Coptis chinensis Franch (Huanglian) is commonly used to treat diabetes in China. In this study, the effects of the C. chinensis Franch polysaccharides (CCP) on advanced glycation end product (AGE) formation in vitro and in streptozotocin-induced diabetic mice were investigated. CCP significantly inhibited all the three periods of nonenzymatic protein glycation in vitro, including Amadori product, dicarbonyl compound, and AGE formation (P < .01). In diabetic mice, the administration of CCP not only improved both bodyweight and serum insulin and decreased fasting blood glucose and glycated serum

Coptis rhizome extract influence on Streptococcus pneumoniae through autolysin activation.

This study investigated the antibacterial properties of Coptis rhizome, a plant traditionally used for respiratory infections, against Streptoccus pneumonia (S. pneumoniae), for which there has been minimal empirical evidence of effectiveness. The study particularly examined autolysis, indirectly associated with antibacterial resistance, when using Coptis rhizome for bacterial infections. In our methodology, Coptis rhizome was processed with ethanol and distilled water to produce four different extracts: CRET30, CRET50, CRET70, and CRDW. The antibacterial activity of these extracts were tested

Alkaloids from Coptis chinensis root promote glucose uptake in C2C12 myotubes.

The root of Coptis chinensis Franch. (COCH) is regularly used for medicinal purposes, and has been prescribed alone or in combination with other traditional herbs for the treatment of diabetes. To investigate the effects of COCH on glucose utilization by skeletal muscles, we prepared an ethanol extract of COCH root (COCH-Et) partitioned with dichloromethane, n-butanol, and water and tested its effects on glucose uptake in differentiated C2C12 myotubes. We found that dichloromethane and n-butanol sub-fractions of COCH-Et promoted glucose uptake in differentiated C2C12 cells at 50 μg/mL. Further

Exploring cadmium uptake pathways in the roots of Coptis chinensis Franch.

To investigate the cadmium (Cd) uptake mechanism in the roots of Coptis chinensis Franch., we utilized non-invasive micro-test technology (NMT) to analyze Cd2+ and H+ fluxes, alongside ICP-MS to assess Cd content and fluxes under various inhibitors and ion channel blockers. Results revealed that Cd2+ uptake primarily occurs in the root meristematic zone. Cd2+ addition significantly inhibited H+ influx. The P-type ATPase inhibitor Na3VO4 and the metabolism inhibitor CCCP both reduced Cd2+ fluxes and Cd content in roots of C. chinensis Ca2+ channel blocker LaCl3 exhibited more pronounced inhibit

Cytokine storm-calming property of the isoquinoline alkaloids in Coptis chinensis Franch.

Coronavirus disease (COVID-19) has spread worldwide and its effects have been more devastating than any other infectious disease. Importantly, patients with severe COVID-19 show conspicuous increases in cytokines, including interleukin (IL)-6, monocyte chemoattractant protein (MCP)-1, IL-8, tumor necrosis factor (TNF)-α, IL-1, IL-18, and IL-17, with characteristics of the cytokine storm (CS). Although recently studied cytokine inhibitors are considered as potent and targeted approaches, once an immunological complication like CS happens, anti-viral or anti-inflammation based monotherapy alone

[Research advances on comprehensive utilization of non-medicinal parts of Coptis chinensis].

Non-medicinal parts(NMP) of Coptis chinensis are abundant but have long been discarded as agricultural waste, resulting in resource wastage and environmental pollution. Research indicates that NMP contains active components similar to those in rhizomes, including alkaloids, flavonoids, and phenylpropanoids. These components exhibit pharmacological activities such as hypoglycemic and lipid-regulating effects, antioxidant properties, and antibacterial activity, while demonstrating high safety profiles, offering broad development prospects. This review systematically summarized the chemical compo

Analysis of the Coptis chinensis genome reveals the diversification of protoberberine-type alkaloids.

Chinese goldthread (Coptis chinensis Franch.), a member of the Ranunculales, represents an important early-diverging eudicot lineage with diverse medicinal applications. Here, we present a high-quality chromosome-scale genome assembly and annotation of C. chinensis. Phylogenetic and comparative genomic analyses reveal the phylogenetic placement of this species and identify a single round of ancient whole-genome duplication (WGD) shared by the Ranunculaceae. We characterize genes involved in the biosynthesis of protoberberine-type alkaloids in C. chinensis. In particular, local genomic tandem d

Coptisine from Coptis chinensis blocks NLRP3 inflammasome activation by inhibiting caspase-1.

Inflammasome mediates the activation of caspase-1, which promotes the secretion of proinflammatory cytokines. In this work, we aimed to investigate whether natural compounds from a Traditional Chinese Medicine prescription called San-Huang-Xie-Xin-Tang exert its clinical efficacy by inhibiting inflammasome activation and the underlying mechanism. The inhibitory effects of compounds on caspase-1 were evaluated in recombinant expressed caspase-1 protein and macrophages. Molecular docking was conducted to examine the interaction between compounds and caspase-1. The effects of the compounds on pro

Regulatory Mechanisms of Benzylisoquinoline Alkaloid Biosynthesis in Coptis chinensis Under Light Quality Treatment.

Coptis chinensis Franch. is a perennial and valuable medicinal plant. The dried rhizome of C. chinensis has been extensively used in traditional Chinese medicine to treat heat-related disorders. Benzylisoquinoline alkaloids (BIAs), including berberine, epiberberine, palmatine, and coptisine, constitute the primary bioactive compounds in this species. Although light quality is recognized as a key regulator of plant growth and metabolism, the specific roles of distinct light spectra in modulating BIA biosynthesis remain poorly understood. In this study, an integrated transcriptomics and metabolo
