Helminth Excretory/Secretory Proteins Ameliorate Colitis Through Preservation of Intestinal Homeostasis.

Inflammatory bowel disease (IBD) manifests as a sustained gastrointestinal condition driven by an ongoing dysregulated immunity, defective mucosal barrier, and microbiota dysbiosis. Parasitic helminths exert immunomodulation chiefly via excretory/secretory proteins (ESP). This study assessed the remedial effects alongside the mechanistic pathways of the ESP derived from Echinococcus multilocularis (Emu-ESP) within a dextran sulfate sodium (DSS)-provoked colitis paradigm in mice. Our results indicated that Emu-ESP treatment significantly mitigated DSS-induced colitis and notably alleviated gut

Helminth infection alters IgE responses to allergens structurally related to parasite proteins.

Immunological cross-reactivity between environmental allergens and helminth proteins has been demonstrated, although the clinically related implications of this cross-reactivity have not been addressed. To investigate the impact of molecular similarity among allergens and cross-reactive homologous helminth proteins in IgE-based serologic assessment of allergic disorders in a helminth-infected population, we performed ImmunoCAP tests in filarial-infected and noninfected individuals for IgE measurements to allergen extracts that contained proteins with high levels of homology with helminth prote

Effects of protein malnutrition on tolerance to helminth infection.

Infection tolerance is the ability of a host to limit the health effects of a given parasite load. A few recent studies have demonstrated genetic variation for tolerance, but little is known about how environmental factors affect tolerance. Here, we used the intestinal nematode Heligmosomoides polygyrus in laboratory mice to test for effects of protein malnutrition on tolerance. We performed an experiment where two different mouse strains (CBA and BALB/c) were fed either adequate-protein food or low-protein food, and trickle-infected with different doses of H. polygyrus larvae during four week

Healing helminths: A systematic review of the anti-inflammatory and disease-modifying potential of helminth-derived proteins.

Helminths secrete diverse immunoregulatory molecules that modulate host inflammatory responses, facilitating long-term persistence within the host. This capacity has prompted interest in helminth-derived molecules as potential therapeutics for diseases characterized by excessive or dysregulated inflammation. This systematic review with cross-study quantitative analysis synthesized evidence on helminth-derived immunomodulatory peptides, polypeptides, and proteins (HDIPs), focusing on their disease-modifying and anti-inflammatory effects in in vivo animal models of inflammatory disease. Intestinal helminths-an outlander in a case of protein losing enteropathy.

Malabsorption is the major disease burden in tropical countries. Both primary and secondary forms exist and a secondary form overshadows the primary category. Intestinal parasitic infections lead to secondary form of tropical malabsorption in both native and travelers and presentation varies from mild glossitis to severe protein losing enteropathy. The underlying condition is often masked unless an endoscopic biopsy is performed. This is followed by a histopathological examination which unravels the etiology.

Eosinophil granule proteins in serum and urine of patients with helminth infections and atopic dermatitis.

Eosinophil cationic protein (ECP) and eosinophil-derived neurotoxin (EPX) are cytotoxic molecules involved in helminth infections and allergic reactions. Hitherto most clinical chemical studies have been concerned with the analysis of serum ECP in allergic diseases. The aim of this study was to examine whether serum as well as urine levels of these proteins are useful clinical chemical parameters in helminthiases and allergic diseases such as atopic dermatitis. Comparing these diseases under the same methodological conditions, levels of ECP and EPX were generally higher in helminthiases than i

Helminth-derived proteins as immune system regulators: a systematic review of their promise in alleviating colitis.

Helminth-derived proteins have immunomodulatory properties, influencing the host's immune response as an adaptive strategy for helminth survival. Helminth-derived proteins modulate the immune response by inducing anti-inflammatory cytokines, promoting regulatory T-cell development, and ultimately favouring a Th2-biased immune response. This systematic review focused on helminth-derived proteins and explored their impact on reducing inflammatory responses in mouse models of colitis. A systematic search across Medline, EMBASE, Web of Science, and Cochrane Library identified fourteen relevant stu

Eukaryotic protein kinases (ePKs) of the helminth parasite Schistosoma mansoni.

BACKGROUND: Schistosomiasis remains an important parasitic disease and a major economic problem in many countries. The Schistosoma mansoni genome and predicted proteome sequences were recently published providing the opportunity to identify new drug candidates. Eukaryotic protein kinases (ePKs) play a central role in mediating signal transduction through complex networks and are considered druggable targets from the medical and chemical viewpoints. Our work aimed at analyzing the S. mansoni predicted proteome in order to identify and classify all ePKs of this parasite through combined computat

Small proline-rich protein 2A is a gut bactericidal protein deployed during helminth infection.

A diverse group of antimicrobial proteins (AMPs) helps protect the mammalian intestine from varied microbial challenges. We show that small proline-rich protein 2A (SPRR2A) is an intestinal antibacterial protein that is phylogenetically unrelated to previously discovered mammalian AMPs. In this study, SPRR2A was expressed in Paneth cells and goblet cells and selectively killed Gram-positive bacteria by disrupting their membranes. SPRR2A shaped intestinal microbiota composition, restricted bacterial association with the intestinal surface, and protected against Listeria monocytogenes infection.

Identification of paramyosin as a binding protein for calgranulin C in experimental helminthic keratitis.

PURPOSE: Calgranulin C (CaGC) is a protein released by activated neutrophils and involved in host defense against filarial infections. This study involved the identification of binding protein(s) of the helminth Brugia malayi to CaGC and the ability of binding complexes to induce keratitis. METHODS: Parasitic extracts prepared from B. malayi microfilariae and adult worms were incubated with recombinant CaGC protein. Parasite binding protein-CaGC complex was isolated by affinity chromatography. A B. malayi microfilariae cDNA library was immunoscreened with antisera from rats immunized with the

Increases in helminth-induced IL-21 protein levels disappear upon sample freezing.

IL-21 is a much studied cytokine that has been implicated in the regulation of TH1, TH2, TH17 and regulatory immune responses; its signalling is a promising therapeutic target for autoimmune, inflammatory and infectious diseases. Despite its biological importance, measuring IL-21 reliably has proved difficult. ELISAs are commonly used to measure cytokines in various biological samples. However, results obtained are only as good as the quality of the sample. Here, we show that when using fresh samples, a significant increase in IL-21 was measured in the intestinal homogenate of mice infected wi

Dendritic cells pulsed with unfractionated helminthic proteins to generate antiparasitic cytotoxic T lymphocyte.

Dendritic cells (DC) are sentinels of immunity. We determined their role in the induction of immunity against alveolar echinococcosis, caused by the larval stage of the cestode Echinococcus multilocularis. Furthermore, we evaluated if unfractionated protein from E. multilocularis (Em-Ag) can be used as loading agent for DC (comparable to unfractionated tumour proteins) in order to generate antiparasitic cytotoxic T lymphocyte (CTL). Interestingly, immature DC did not mature in the presence of 1 microg/ml Em-Ag as analysed by FACS and mixed leucocyte reactions. Yet, their capacity to take up de

Structural basis for IL-33 recognition and its antagonism by the helminth effector protein HpARI2.

IL-33 plays a significant role in inflammation, allergy, and host defence against parasitic helminths. The model gastrointestinal nematode Heligmosomoides polygyrus bakeri secretes the Alarmin Release Inhibitor HpARI2, an effector protein that suppresses protective immune responses and asthma in its host by inhibiting IL-33 signalling. Here we reveal the structure of HpARI2 bound to mouse IL-33. HpARI2 contains three CCP-like domains, and we show that it contacts IL-33 primarily through the second and third of these. A large loop which emerges from CCP3 directly contacts IL-33 and structural c

The immune-modulatory potential of helminth-derived proteins in cellular models of inflammation: a systematic review with cross-study quantitative data analysis.

Helminths are parasitic worms that secrete a plethora of immune regulatory molecules which allow them to moderate inflammatory responses by their host's immune system to ensure their survival within the host. This ability to drive a compatible existence with their host has led to research into the potential therapeutic effects of helminth-derived molecules the regulation of overactive immune and inflammatory responses in a wide variety of diseases. This systematic review with cross-study quantitative data analysis aims to synthesize the published data on helminth-derived immunomodulatory pepti

Does protein malnutrition modulate Schistosoma mansoni response to praziquantel? Assessment of adult helminth morphology.

Praziquantel (PZQ) is the only drug for schistosomiasis, a disease that frequently coexists with protein malnutrition in endemic populations. While this comorbidity is highly prevalent, its specific influence on PZQ's efficacy and the resulting parasite damage is poorly defined. This study addresses this knowledge by conducting a structural investigation into how the therapeutic efficacy of PZQ against Schistosoma mansoni in a hypoprotein diet murine model impacts on parasites. After four weeks of a normoprotein diet (22% protein) or hypoprotein diet (8% protein) feeding, mice were infected wi

An integrated transcriptomics and proteomics analysis of the secretome of the helminth pathogen Fasciola hepatica: proteins associated with invasion and infection of the mammalian host.

To infect their mammalian hosts, Fasciola hepatica larvae must penetrate and traverse the intestinal wall of the duodenum, move through the peritoneum, and penetrate the liver. After migrating through and feeding on the liver, causing extensive tissue damage, the parasites move to their final niche in the bile ducts where they mature and produce eggs. Here we integrated a transcriptomics and proteomics approach to profile Fasciola secretory proteins that are involved in host-pathogen interactions and to correlate changes in their expression with the migration of the parasite. Prediction of F.

A unique intestinal desensitization strategy: Blocking membrane protease enrichment on macrophages by helminth-derived cystatin-like protein.

Egg allergies are prevalent worldwide and pose serious risks in children; however, a permanent cure is lacking. The desensitization of Trichinella spiralis-secreted cystatin-like protein (Ts-CLP) is an alternative to helminthic therapy for food allergies. Based on the allergy model in which Escherichia coli (E. coli) delivers ovalbumin (OVA) to the digestive system of OT-II mice, various therapeutic interventions related to Ts-CLP partially (delayed bone marrow-derived macrophages (BMDMs) intervention) or completely relieved inflammation and the disease course. Safety of P28GST, a Protein Derived from a Schistosome Helminth Parasite, in Patients with Crohn's Disease: A Pilot Study (ACROHNEM).

Despite the development of novel therapies, inflammatory bowel diseases remain an innovative treatment challenge. Helminth therapy is a new promising approach, and a key issue is the identification of helminth-derived anti-inflammatory mediators. P28 glutathione-S-transferase (P28GST), a protein derived from schistosomes, a trematode parasitic helminth, was shown to reduce intestinal inflammation in experimental colitis by down-regulating the Th1/Th17 response. In this multicenter, open-label, pilot Phase 2a study, we evaluated the safety of P28GST administered to patients with mild Crohn's di

Extracellular vesicles from parasitic helminths contain specific excretory/secretory proteins and are internalized in intestinal host cells.

The study of host-parasite interactions has increased considerably in the last decades, with many studies focusing on the identification of parasite molecules (i.e. surface or excretory/secretory proteins (ESP)) as potential targets for new specific treatments and/or diagnostic tools. In parallel, in the last few years there have been significant advances in the field of extracellular vesicles research. Among these vesicles, exosomes of endocytic origin, with a characteristic size ranging from 30-100 nm, carry several atypical secreted proteins in different organisms, including parasitic proto
