Neurotoxic alkaloids: saxitoxin and its analogs.

Saxitoxin (STX) and its 57 analogs are a broad group of natural neurotoxic alkaloids, commonly known as the paralytic shellfish toxins (PSTs). PSTs are the causative agents of paralytic shellfish poisoning (PSP) and are mostly associated with marine dinoflagellates (eukaryotes) and freshwater cyanobacteria (prokaryotes), which form extensive blooms around the world. PST producing dinoflagellates belong to the genera Alexandrium, Gymnodinium and Pyrodinium whilst production has been identified in several cyanobacterial genera including Anabaena, Cylindrospermopsis, Aphanizomenon Planktothrix an

[Saxitoxins and tetrodotokxins as a new biological weapon].

Saxitoxins (STX) and tetrodotoxins (TTX) are a group of chemical compounds produced by certain species of marine algae and fish. Lethal dose for a human is about 0.5-2.0 mg when the toxin enters the body via food, and 0.05 mg of poisoning at the time of injection. In the case of aerosol the lethal dose for human being is 5 mg/min/m(3). STX and TTX poisoning cause mostly symptoms from the nervous system in the form of: paresthesia around the lips, tongue, gums, distal segments of the limbs, headache, dysphonia, astigmatism, floating feeling, muscle weakness, paralysis of cranial and peripheral

Binding of tetrodotoxin and saxitoxin to sodium channels.

A useful first step in any chemical characterization of the sodium channels in nerve membrane would clearly be the identification of some measurable property of the channel that does not depend on the intactness of the tissue. To this end, tetrodotoxin and saxitoxin, which bind specifically to sodium channels, have been triated and their binding to rabbit, lobster and garfish non-lyelinated nerve fibres examined. In each case, a component of the binding curve was found that saturated at concentrations of a few nanomolar. In addition, non-specific binding, indicated by a linear dependence of th

Binding to saxitoxin to electrically excitable neuroblastoma cells.

Saxitoxin inhibits the action potential Na+ ionophore of electrically excitable neuroblastoma cells with a KI of 3.7 nM. Binding experiments detect a single class of saturable binding sites with KD = 3.9 nM and a binding capacity of 156 fmol/mg of cell protein (78 sites per micrometer2 of cell surface). Saturable binding is completely inhibited by tetrodotoxin but is unaffected by scorpion toxin or batrachotoxin. No saturable binding is observed in cultures of clone N103, a variant neuroblastoma clone lacking the action potential Na+ response. Thus, saxitoxin binds specifically to the action p

Respiratory and circulatory effects of saxitoxin in the cerebrospinal fluid.

1 In cats anaesthetized with pentobarbitone, saxitoxin and, on a few occasions, tetrodotoxin were injected into a lateral cerebral ventricle or into the subarachnoid space of the lower brain stem. Observations were made on frequency and tidal volume of breathing, on CO(2) responsiveness and on electrical responsiveness of the respiratory centre. Effects on the blood pressure were observed simultaneously.2 A single large dose of toxin, e.g., 250 ng, produced within minutes apneustic breathing and a rise in blood pressure which were converted rapidly to respiratory failure and hypotension. In co

Extraneuronal saxitoxin binding sites in rabbit myelinated nerve.

The changes in binding of 3H-labeled saxitoxin (STX) to rabbit sciatic nerve during axonal regeneration (after nerve crush) and during axonal degeneration (after nerve section) were measured and compared with the corresponding changes in the sciatic nerves of other mammals (rat, guinea pig, and cat). In the rabbit and rat, regeneration after nerve crush is associated with a 2- to 4-fold increase in STX binding capacity, consistent with the known corresponding increase in the number of nodes of Ranvier in regenerating nerve. Furthermore, consistent with the disappearance of nodes that occurs wi

Scalable total synthesis of saxitoxin and related natural products.

Saxitoxin (STX, 1), a potent neurotoxin from shellfish, first isolated in 1957 (ref. 1), offers immense pharmaceutical potential owing to its interaction with voltage-gated sodium channels2, which are ubiquitously present in all excitable cells of the central and peripheral nervous system3. Hundreds of studies towards its synthesis have been disclosed so far, yet a fully modular and scalable approach to the family remains elusive4-12. Here we show how a tactical combination of radical retrosynthesis, biocatalysis and C-H functionalization logic can be used to solve this problem, resulting in a

Comparative cytotoxicity of aflatoxin B1 and saxitoxin in cell cultures.

Human and animal cell cultures were evaluated for their susceptibility to two environmental toxins found as contaminants in human food supplies: aflatoxin B1, a hepatotoxin produced by the mold Aspergillus flavus, and saxitoxin, a paralytic neurotoxin produced by the marine dinoflagellate Gonyaulax catenella. Both toxins cause food poisoning in humans and other animals. The acute cytotoxicity of both toxins was measured and compared by inhibition of cell growth and by progressive cytopathogenicity resulting in cell destruction. Aflatoxin B1 was cytotoxic to all of the 11 primary kidney culture

The binding of labelled saxitoxin to the sodium channels in nerve membranes.

1. Tritium labelled saxitoxin has been prepared and purified, and its binding both to intact rabbit vagus nerves and to a solubilized preparation of garfish olfactory nerve membranes has been examined.2. In intact and solubilized nerves there is a saturable binding component of magnitude equal to that previously obtained for labelled tetrodotoxin.3. This component of bound saxitoxin is displaced competitively by tetrodotoxin, and it is concluded that the two toxins bind to the same site.4. The saturable saxitoxin (STX) interaction with the nerve membrane is reversible and can be described by t

In this study, we proposed label-free saxitoxin (STX) sensor using STX specific aptamer in combination with spectroscopic ellipsometry (SE) and attenuated internal reflection (AIR) spectroscopic ellipsometry method which is operated under surface plasmon resonance (SPR) conditions. Besides the other surface plasmon resonance-based applications, AIR-SE applications have unique advantages in terms of sensitivity and it was used herein for real-time detection of STX in real samples. Another method, SE, was also used and compared with AIR-SE. Analytical performances were satisfactory with low dete

Structural basis for saxitoxin congener binding and neutralization by anuran saxiphilins.

Dinoflagellates and cyanobacteria produce saxitoxin (STX) and ~50 congeners that disrupt bioelectrical signals by blocking voltage-gated sodium channels (NaVs). Consuming seafood carrying these toxins causes paralytic shellfish poisoning (PSP). Although NaVs and anuran STX binding proteins (saxiphilins, Sxphs) use convergent STX binding modes, the structural basis for STX congener recognition is unknown. Here, we show that American bullfrog (Rana catesbeiana) RcSxph and High Himalaya frog (Nanorana parkeri) NpSxph sequester STX congeners using a 'lock and key' mode shared with STX. Importantly

The saxitoxin (STX) receptor has been purified 740-fold from rat brain by a combination of ion exchange chromatography, wheat germ agglutinin chromatography, and sedimentation on sucrose gradients to a specific activity of 1488 pmol/mg of protein. The best fractions were estimated to be 47% pure from their specific activity or 66% pure on the basis of NaDodSO4 gel electrophoresis. Two polypeptides, alpha (Mr approximately equal to 270,000 +/- 10,000) and beta (Mr approximately equal to 38,300 +/- 2000) (mean +/- SD) copurify with STX binding activity. Enhanced stability of CH3NH3PbBr3 quantum dots for electrochemiluminescence detection of saxitoxin.

Saxitoxin (STX) is a highly potent phycotoxin that poses a severe threat to both human health and aquatic ecosystems. Given its extremely low permissible level in food products, there is an urgent need for a sensitive, specific, and cost-effective screening method. In this study, an electrochemiluminescence (ECL)-based method was developed for STX detection, capitalizing on the inherent advantages of ECL, including high sensitivity and minimal background interference. To achieve selective recognition of STX, molecularly imprinted polymers (MIPs) featuring specific memory cavities were modified

Discovery of Selective Inhibitors of NaV1.7 Templated on Saxitoxin as Therapeutics for Pain.

The voltage-gated sodium channel isoform NaV1.7 has drawn widespread interest as a target for non-opioid, investigational new drugs to treat pain. Selectivity over homologous, off-target sodium channel isoforms, which are expressed in peripheral motor neurons, the central nervous system, skeletal muscle and the heart, poses a significant challenge to the development of small molecule inhibitors of NaV1.7. Most inhibitors of NaV1.7 disclosed to date belong to a class of aryl and acyl sulfonamides that preferentially bind to an inactivated conformation of the channel. By taking advantage of a se

Binding and Pharmacokinetics of the Sodium Channel Blocking Toxins (Saxitoxin and the Tetrodotoxins).

Tetrodotoxin (TTX) found in diverse variety of animals including puffer fishes, some newts, frogs and limited number of non-vertebrate species (6 different phyla). The saxitoxin (STX) and the TTX are small molecules composed of 7,8,9 guanidinium and 1,2,3 guanidinium groups, respectively in their structures. These groups provide positive charge to the molecules and are believed to interact with negatively charged Glu755 and Asp400 residues in domain II and I of the sodium channel strongly. The pharmacokinetic studies (absorption, distribution and accumulation) reported on Takifugu rubripes, Ta

Transcriptomic comparison unveils saxitoxin biosynthesis genes in the marine dinoflagellate Gymnodinium catenatum.

The marine dinoflagellate Gymnodinium catenatum is known to produce saxitoxins (STXs) that are responsible for paralytic shellfish poisoning (PSP); however, the genes involved in STX synthesis are insufficiently understood. In the present study, we determined transcriptome sequences of toxic G. catenatum isolated from Korea (Gc-KR) and compared them with G. catenatum strains reported in other regions such as Spain (Gc-SP) and the United States (Gc-US). Toxin analysis showed that the Korean strain produced the toxins at 6.0 ± 1.9 STXs eq fmol/cell. Comparative transcriptomics of the three strai

Electrophysiological Evaluation of Pacific Oyster (Crassostrea gigas) Sensitivity to Saxitoxin and Tetrodotoxin.

Pacific oysters (Crassostrea gigas) may bio-accumulate high levels of paralytic shellfish toxins (PST) during harmful algal blooms of the genus Alexandrium. These blooms regularly occur in coastal waters, affecting oyster health and marketability. The aim of our study was to analyse the PST-sensitivity of nerves of Pacific oysters in relation with toxin bio-accumulation. The results show that C. gigas nerves have micromolar range of saxitoxin (STX) sensitivity, thus providing intermediate STX sensitivity compared to other bivalve species. However, theses nerves were much less sensitive to tetr

Saxiphilin functions as a toxin sponge protein that counteracts the effects of saxitoxin poisoning.

Saxitoxin (STX) is among the most potent toxins known, is classified as a chemical weapon, and is the archetype of the paralytic shellfish toxin (PST) family produced by marine and freshwater harmful algal blooms (HAB). STX causes paralysis and death through inhibition of voltage-gated sodium channels (NaVs), has no antidote, and poses a public health and commercial fishing threat due to its accumulation in seafood and increasing HAB occurrences. Although STX is lethal to varied vertebrates, including humans, certain frogs resist STX poisoning. This phenotype is thought to depend on soluble to

Transcriptome response of a marine copepod in response to environmentally-relevant concentrations of saxitoxin.

Paralytic shellfish toxins (PSTs) can pose a serious threat to human health. Among them, saxitoxin (STX) is one of the most potent natural neurotoxins. Here, the copepod Tigriopus japonicus, was exposed to environmentally relevant concentrations (2.5 and 25 μg/L) STX for 48 h. Although no lethal effects were observed at both concentrations, the transcriptome was significantly altered, and displayed a concentration-dependent response. STX exposure decreased the copepod's metabolism and compromised immune defense and detoxification. Additionally, STX disturbed signal transduction, which might af
