Transcriptomic evidence for versatile metabolic activities of mercury cycling microorganisms in brackish microbial mats.

Methylmercury, biomagnifying through food chains, is highly toxic for aquatic life. Its production and degradation are largely driven by microbial transformations; however, diversity and metabolic activity of mercury transformers, resulting in methylmercury concentrations in environments, remain poorly understood. Microbial mats are thick biofilms where oxic and anoxic metabolisms cooccur, providing opportunities to investigate the complexity of the microbial mercury transformations over contrasted redox conditions. Here, we conducted a genome-resolved metagenomic and metatranscriptomic analys

The Effect of Crank Resistance on Arm Configuration and Muscle Activation Variances in Arm Cycling Movements.

Arm cycling on an ergometer is common in sports training and rehabilitation protocols. The hand movement is constrained along a circular path, and the user is working against a resistance, maintaining a cadence. Even if the desired hand trajectory is given, there is the flexibility to choose patterns of joint coordination and muscle activation, given the kinematic redundancy of the upper limb. With changing external load, motor noise and changing joint stiffness may affect the pose of the arm even though the endpoint trajectory is unchanged. The objective of this study was to examine how the c

Running after cycling induces inter-limb differences in muscle activation but not in kinetics or kinematics.

Overuse injuries are a common problem to triathletes' population. Overuse injuries may arise from inter-limb biomechanical differences during running, but the literature lacks information regarding inter-limb differences in triathletes. In this study inter-limb differences were investigated in injury-free triathletes during the running portion of a simulated cycle-run transition. Thirteen triathletes performed a 5 km run preceded by a 20 min cycling trial at 70% of maximal power output. During the Start, Mid and End stages of running, kinetic, kinematic and muscle activation variables were com

Beclin-1-mediated activation of autophagy improves proximal and distal urea cycle disorders.

Urea cycle disorders (UCD) are inherited defects in clearance of waste nitrogen with high morbidity and mortality. Novel and more effective therapies for UCD are needed. Studies in mice with constitutive activation of autophagy unravelled Beclin-1 as druggable candidate for therapy of hyperammonemia. Next, we investigated efficacy of cell-penetrating autophagy-inducing Tat-Beclin-1 (TB-1) peptide for therapy of the two most common UCD, namely ornithine transcarbamylase (OTC) and argininosuccinate lyase (ASL) deficiencies. TB-1 reduced urinary orotic acid and improved survival under protein-ric

Differential activity of the corticosteroidogenic enzymes in normal cycling women and women with polycystic ovary syndrome.

The phenotypic complex of patients with definitive diagnosis of polycystic ovary syndrome may include patients with normal and high serum androgen levels. Patients with hyperandrogenemia seem to present higher risk of changes to the glucose and lipid metabolism and, eventually, of earlier development of cardiovascular diseases than normoandrogenemic patients or healthy women. From a laboratory and clinical point of view, it is important to check androgen levels in patients with polycystic ovary syndrome. The identification of partial insufficiency of a given corticosteroidogenic enzyme is also

Upregulation of Krebs cycle and anaerobic glycolysis activity early after onset of liver ischemia.

The liver is a highly vascularized organ receiving a dual input of oxygenated blood from the hepatic artery and portal vein. The impact of decreased blood flow on glucose metabolism and how hepatocytes could adapt to this restrictive environment are still unclear. Using the left portal vein ligation (LPVL) rat model, we found that cellular injury was delayed after the onset of liver ischemia. We hypothesized that a metabolic adaptation by hepatocytes to maintain energy homeostasis could account for this lag phase. Liver glucose metabolism was characterized by 13C- and 1H-NMR spectroscopy and a

Regulation of TBK1 activity by Optineurin contributes to cell cycle-dependent expression of the interferon pathway.

The innate immune system has evolved to detect and neutralize viral invasions. Triggering of this defense mechanism relies on the production and secretion of soluble factors that stimulate intracellular antiviral defense mechanisms. The Tank Binding Kinase 1 (TBK1) is a serine/threonine kinase in the innate immune signaling pathways including the antiviral response and the host defense against cytosolic infection by bacteries. Given the critical roles of TBK1, important regulatory mechanisms are required to regulate its activity. Among these, Optineurin (Optn) was shown to negatively regulate

Prion-induced neurotoxicity: Possible role for cell cycle activity and DNA damage response.

Protein misfolding neurodegenerative diseases arise through neurotoxicity induced by aggregation of host proteins. These conditions include Alzheimer's disease, Huntington's disease, Parkinson's disease, motor neuron disease, tauopathies and prion diseases. Collectively, these conditions are a challenge to society because of the increasing aged population and through the real threat to human food security by animal prion diseases. It is therefore important to understand the cellular and molecular mechanisms that underlie protein misfolding-induced neurotoxicity as this will form the basis for

Differential modulation of activity related to the anticipation of monetary gains and losses across the menstrual cycle.

Estradiol and progesterone interact with the dopaminergic and other neurotransmitter systems that are involved in the processing of rewards. On the systems level, these hormones modulate responses to stimulants as well as neuronal activity related to the anticipation of monetary gains. As different mechanisms might underlie the processing of gains and losses, the current study aims to investigate whether neural correlates of gain and loss anticipation are differentially modulated by menstrual cycle phases. Therefore, young, naturally cycling women were examined by means of functional neuroimag

Probing the GTPase cycle with real-time NMR: GAP and GEF activities in cell extracts.

The Ras superfamily of small GTPases is a large family of switch-like proteins that control diverse cellular functions, and their deregulation is associated with multiple disease processes. When bound to GTP they adopt a conformation that interacts with effector proteins, whereas the GDP-bound state is generally biologically inactive. GTPase activating proteins (GAPs) promote hydrolysis of GTP, thus impeding the biological activity of GTPases, whereas guanine nucleotide exchange factors (GEFs) promote exchange of GDP for GTP and activate GTPase proteins. A number of methods have been developed

Activation of the RpoN-RpoS regulatory pathway during the enzootic life cycle of Borrelia burgdorferi.

BACKGROUND: The maintenance of Borrelia burgdorferi in its complex tick-mammalian enzootic life cycle is dependent on the organism's adaptation to its diverse niches. To this end, the RpoN-RpoS regulatory pathway in B. burgdorferi plays a central role in microbial survival and Lyme disease pathogenesis by up- or down-regulating the expression of a number of virulence-associated outer membrane lipoproteins in response to key environmental stimuli. Whereas a number of studies have reported on the expression of RpoS and its target genes, a more comprehensive understanding of when activation of th

Influence of experimental intrauterine infusion of Arcanobacterium pyogenes solution on ovarian activity in cycling cows.

To examine the ovarian response to Arcanobacterium pyogenes (A. pyogenes) in uterus, bacterial solution was infused into the uteri of cows, and the follicle and corpus luteum (CL) development were monitored with a real-time ultrasound instrument. In addition, the plasma concentrations of progesterone (P(4)) and 13, 14-dihydro-15-keto-PGF(2alpha) (PGFM) were determined. A 10 ml bacterial solution that contained A. pyogenes (8 to 15 x 10(8) CFU/ml) was infused into the uterus of eight cows transcervically three days after natural ovulation. As a control, sterile physiological saline was infused

The effects of weight cycling on serum leptin levels and lipogenic enzyme activities in adipose tissue.

Weight cycling is one of the widely used weight reduction strategies; however, the adverse effects of this method include regaining significant amounts of weight. The molecular mechanisms underlying weight gain following cycles of dietary deprivation and refeeding are still poorly understood. One of the possibilities is that repeated loss and gain of weight may promote fat deposition in adipose tissue. To test this hypothesis we investigated serum leptin levels and lipogenic enzyme activities in white adipose tissue (WAT) of male Wistar rats during 12 days of ad libitum feeding following multi

Cell proliferation activity and the expression of cell cycle regulatory proteins in oral lichen planus.

BACKGROUND: In oral lichen planus (OLP), destruction of the basal cell layer, which is one of the characteristic histological features, is seen and many changes in cell proliferation, cell repair and cell death occur in the injured mucosal epithelium. METHODS: We studied mucosal tissues from 19 patients of OLP and 10 controls, with immunohistochemistry for Ki-67, p53, cyclin dependent kinase inhibitors (CDKI) and cyclins. Mitotic count was calculated. TUNEL assay was also performed for evaluation of apoptotic cell death. RESULTS: Mitotic count, Ki-67 and cyclin D1 labeling indices in the bas

Effect of reproductive hormones on ovarian epithelial tumors: I. Effect on cell cycle activity.

We examined the effects of the 4 major female reproductive hormones, estradiol (E2), progesterone (P4), follicle stimulating hormone (FSH), and luteinizing hormone (LH) on thymidine incorporation in benign and malignant ovarian epithelial tumors cultured in vitro. Treatment of these tumors with E2, FSH and LH resulted in increased thymidine incorporation while treatment with P4 inhibited growth as well as thymidine incorporation. The inhibitory effect of progesterone could not be reproduced by treating the cells with ligands for other steroid hormone receptors known to interact with P4 such as

Angiotensin-converting enzyme activity in the bovine uteroplacental unit changes in relation to the cycle and pregnancy.

The expression of the angiotensin-forming enzymes, renin and angiotensin-converting enzyme (ACE), were examined in the bovine uteroplacental unit. The ACE activity was determined in cell membrane fractions, and ACE and renin were localized by autoradiography and immunohistochemistry, respectively. In the myometrium, the ACE activity was significantly higher in dioestrous than in oestrous. ACE activity correlated negatively with the day of gestation in the endometrium and myometrium but positively in the placentome and allantoamniotic membrane. Autoradiography showed, that ACE was localized in

Targeted disruption of CDK4 delays cell cycle entry with enhanced p27(Kip1) activity.

The mechanism by which cyclin-dependent kinase 4 (CDK4) regulates cell cycle progression is not entirely clear. Cyclin D/CDK4 appears to initiate phosphorylation of retinoblastoma protein (Rb) leading to inactivation of the S-phase-inhibitory action of Rb. However, cyclin D/CDK4 has been postulated to act in a noncatalytic manner to regulate the cyclin E/CDK2-inhibitory activity of p27(Kip1) by sequestration. In this study we investigated the roles of CDK4 in cell cycle regulation by targeted disruption of the mouse CDK4 gene. CDK4(-/-) mice survived embryogenesis and showed growth retardation

Changes in the gonadotropin-releasing hormone pulse generator activity during the estrous cycle in the goat.

The hypothalamic GnRH pulse generator activity, which was assessed by the characteristic increases in the multiple unit activity (MUA) specifically associated with the pulsatile LH secretion (MUA volleys), was monitored throughout the estrous cycle of the female goat. Blood samples were collected daily for analysis of progesterone, estradiol and LH levels to monitor ovarian cyclicity. The frequency of the MUA volleys showed a reciprocal relationship with the plasma progesterone profile; being low during the luteal phase, whereas increased during the follicular phase until the onset of the subs

Activation of the p34 CDC2 protein kinase at the start of S phase in the human cell cycle.

Using a protocol for selecting cells on the basis of both size and age (with respect to the preceding mitosis), we isolated highly synchronous human G1 cells. With this procedure, we demonstrated that the p34 CDC2 kinase was activated at the start of S phase. Cyclin A synthesis began at the same time, and activation of the p34 CDC2 kinase at the start of S phase was, at least in part, due to its association with cyclin A. Furthermore, cells synchronized in late G1 by exposure to the drug mimosine contain active cyclin A/p34 CDC2 kinase, indicating that p34 CDC2 activation can occur before DNA

[Immunoradiometric assay for active renin and plasma prorenin during cycles stimulated by IVFET or GIFT].

Prorenin (PR) an inactive high molecular weight form of renin normally circulates in human plasma at a concentration of about 10 times that of active renin (AR) and this proenzyme seems to be linked to the reproductive function. It has been demonstrated that AR and PR are present at high concentrations in follicular fluid when the ovaries are stimulated with gonadotropins and that the PR plasma levels increase steadily after hCG injection with a correlation between blood PR and the number of developing follicles and corpora lutea. From September 1986 we studied the profile of immunoreactive ac
